Project description:Infection is a devastating post-surgical complication, often necessitating additional procedures and prolonged antibiotic therapy. This is especially relevant for craniotomy and prosthetic joint infections, both of which are characterized by biofilm formation on the bone or implant surface, respectively, with S. aureus representing a primary cause. The effectiveness of immune responses to these infections is predicated on both host- and pathogen-derived signals in the infection microenvironment. However, the extent to which these signals differ across distinct tissue niches and influence immune function remains relatively unknown. Using mouse models of S. aureus craniotomy and prosthetic joint infection complemented with patient samples from both infectious modalities, we show profound metabolomic, transcriptomic, and functional differences that are dependent on tissue niche. These signatures were both spatially and temporally distinct, differing not only between surgical site infections but evolving over time within a single model. These findings highlight the unique immune attributes of biofilms that are heavily influenced by the local tissue microenvironment, which will likely have important implications when designing therapeutic approaches to target specific infections.

Project description:The ability of Entamoeba histolytica to phagocytose host cells correlates to observed virulence in vivo. To better understand the mechanism of phagocytosis we used paramagnetic beads coated with host ligand and sorted trophozoites based on phagocytic ability. Gene expression was then measured in both the sorted phagocytic and non-phagocytic populations using a custom Affymetrix chip for E. histolytica. Feed forward regulation of phagocytosis by Entamoeba histolytica. Infection and Immunity. PMID 23045476 M280 Streptavidin Dynabeads (Invitrogen) were labeled with 20ug/mL biotinylated Human C1q (Quidel). Entamoeba histolytica (strain HM1) were washed twice with PBS then resuspended with the labeled beads at a 10:1 ratio of beads to trophozoites. Samples were incubated at 37°C for 45 minutes, washed twice with agitation to remove adherent beads, then resuspended in MACS buffer. Project description:We report the application of RNA-Seq to assess transcriptional profiles of S. aureus CC30 strains with allelic replacements (knockouts) of two genes in the LFR genomic islet: 1) fatty acid desaturase (fad) and 2) a MocR regulator. The overall results of our study suggest that the LFR islet enhances metabolic plasticity of the CC30 lineage which contributes to increased colonization, survival and persistence in the host. The background strain used for these experiments was UAMS-1. Allelic replacements of fad and MocR were made in UAMS-1. The RNA-Seq experiments compared the transcriptional profiles of UAMS-1 vs. delta fad, and UAMS-1 vs. delta MocR. Two biological replicates were used for each RNA-Seq analysis.

Few differences were observed between the expression properties of S. aureus harvested from diabetic vs. nondiabetic cardiac vegatations. Significant differences were observed between the expression properties of S. aureus harvested from cardiac vegetations in comparison to exponential and/or stationary phase planktonically grown cells.

Project description:WalKR is an essential two component regulatory system in S. aureus, thought to control cell wall metabolism. Using genome sequencing of 5 paired clinical isolates of vancomycin-susceptible and vancomycin-intermediate S. aureus we found frequent, but unique, mutations in this locus. To investigate the contribution of these mutations to vancomycin resistance allelic replacement WalK (G223D) and WalR (K208R) mutants were generated and compared to the parent strains. Mutations in walk and walR led to increased vancomycin resistance, reduced biofilms formation and attenuation of virulence, demonstrating that minor genetic changes in this locus can lead to significant changes in bacterial resistance and virulence. Microarray transcriptional comparisons were performed to investigate the regulatory effects of the WalK (G223D) and WalR (K208R) mutations, and demonstrated that while changes in genes affecting cell wall metabolism were detected, more dramatic changes were found in regulation of cellular metabolism. Transcriptional profiling of laboratory derived S. aureus walKR mutants compared to the parent isolates. TPS3130 has a single point mutation in walK, and TPS3190 has a single point mutation in walR.
