Gene expression profile at single cell level of hESC H9 derived hemognic niches at Day 18 of in vitro differentiation

ABSTRACT: Small drug (SB/CHIR) treatment and orbital shear treatment promotes the generation of hemogenic endothelium and multipotent hematopoieitic progenitor cells. We used single cell RNA sequencing (scRNA-seq) to analyze the diversity of hemogenic niches derived from hESC H9 line at day 18 of in vitro differentiation.

Hemogenic Endothelium transcriptome along the timeline of hESC differentiation

Project description:The differentiation of human embryonic stem cells to hematopoietic lineages initiates with the specification of hemogenic endothelium, a transient specialized endothelial precursor of all blood cells.Unfortunately, absence of hemogenic endothelium-specific markers as well as lack of consensus in the timing of hemogenic potential analysis and methodologies used to study the hematopoietic potential of this precursor prevents reaching clear and definite conclusions. Here, we demonstrate that the hemogenic potential of the endothelium precursor population sharply decline over the course of the differentiation process. Poly(A) RNA-sequencing on CD31+CD144+ population at day 6, day 8 and day 10 of EB diffferentiation with or without the addition of cytokines. Comparasion with hematopoietic committed population CD31+CD144- from day 10 of EB differentiation.

β-catenin regulates primitive streak induction through collaborative interactions with SMAD2/3 and OCT4

Project description:Canonical Wnt and Nodal signaling are both required for induction of the primitive streak (PS), which guides organization of the early embryo. The Wnt effector β-catenin is thought to function in these early lineage specification decisions via transcriptional activation of Nodal signaling. Here, we demonstrate a broader role for β-catenin in PS formation by analyzing its genome-wide binding in a human embryonic stem cell model of PS induction. β-catenin occupies regulatory regions in numerous PS and neural crest genes, and direct interactions between β-catenin and the Nodal effectors SMAD2/3 are required at these regions for PS gene activation. Furthermore, OCT4 binding in proximity to these sites is likewise required for PS induction, suggesting a collaborative interaction between β-catenin and OCT4. Induction of neural crest genes by β-catenin is repressed by SMAD2/3, ensuring proper lineage specification. This study provides mechanistic insight into how Wnt signaling controls early cell lineage decisions. Project description:Purpose: Characterize the gene expression profile of knocked-down ZEB1 and ZEB2 H9 cells during myogenic differentiation through RNA sequencing Methods: RNA sequencing of RNA from cell cultures treated for 2 days with CHIR from the three genotypes used in this study: H9shCTL, H9shZEB1 and H9shZEB2 Results: Knock-down of ZEB1 and ZEB2 in H9 cells results in differential gene expression upon differentiation. Conclusions: Compairson of knocked-down ZEB1 and ZEB2 H9 cells display differences in gene expression at day 2 of the differentiation protocol.
