MicroRNA and mRNA expression profiles of hepatobiliary tumor patients' tissue together with those of non-tumor part tissues and serum exosomes. (mRNA)

ABSTRACT: 10 cholangiocarcinoma (including 4 younger cases) and 14 control (10 hepatocellular carcinomas and 4 benigns) patients' tumor and non-tumor parts were obtained and then devided into 8 groups. Total RNA were extracted and purified from tissues and microRNA signal intensities are measured using Toray 3D gene microarrays designed for expression profiles. mRNA signal intensities were measured using agilent one color microarrays designed for expression profiles.

Project description:TORAY microarray assays were performed on HPMCs derived from four independent donors. miRNA expression was measured in control samples and after 1 ng/ml of TGF-β1 treatment for 48 h. TGF-β1-driven mesothelial-to-mesenchymal transition (MMT) was confirmed in all samples by analysis of the eight molecular markers previously described (data not shown). A total of 1 μg in a volume of ≤2 μl of total RNA from each sample was subsequently used for Toray microarray analysis, performed by Central Biotechnology Services at Cardiff University.

Indel array-based genotyping of pupariation site preference among Drosophila sechellia backcross progeny

Project description:We identified 6,975 insertion/deletion events of between 10 and 100 bp in length from the Drosophila simulans and Drosophila sechellia Mercator/MAVID genomic sequence alignment. Replicate pure samples of Drosophila simulans and Drosophila sechellia gDNA were competitively hybridized to measure the expected relative hybridization intensity of alleles from each species. We used these measured intensities to assess the likelihood that the hybridization signal at each probe in an experimental animal reflected homozygosity or heterozygosity at that locus.
