Genome-wide identification of target genes for the transcription factor Klf13 in differentiated primary mouse oligodendrocytes.

ABSTRACT: To identify target genes of the Klf13 transcription factor in oligodendroglia in a genome-wide and unbiased approach, we cultured primary mouse oligodendroglia of wild type and Klf13-knockout mice, kept them under differentiating conditions, and isolated total RNA which was followed by RNA-Seq studies. Among others we detected substantial changes in the expression of genes associated with differentiation and myelination of oligodendrocytes.

Among others we detected substantial changes in the expression of genes associated with the onset of terminal differentiation of oligodendrocytes.

Transcription factors Sox8 and Sox10 contribute differentially to maintenance of the differentiated state in oligodendrocytes

Project description:To identify target genes of the transcription factors Sox8 and Sox10 in oligodendrocytes in a genome-wide and unbiased approach, we induced oligodendocyte-specific deletion of Sox8, Sox10 or a combination thereof (Sox8 knockout, Sox10 knockout, double knockout) in 2 months-old mice, dissected the corpus callosum of control and mutant mice after 21 days and isolated total RNA for ensuing RNA-Seq studies. Among others we detected substantial changes in the expression of genes associated with differentiation of oligodendrocytes and myelination in Sox10 knockout mice as well as double knockout mice but not in Sox8 knockout mice.

Genome-wide mapping of Olig2 targets in primary oligodendrocytes

Project description:Myelination by oligodendrocytes in the central nervous system (CNS) is essential for proper brain function, yet the molecular determinants that control this process remain poorly understood. The basic helix-loop-helix transcription factors Olig1 and Olig2 promote myelination, whereas bone morphogenetic protein (BMP) and Wnt/?-catenin signaling inhibit myelination. Here we show that these opposing regulators of myelination are functionally linked by the Olig1/2 common target Smad-interacting protein-1 (Sip1). We demonstrate that Sip1 is an essential modulator of CNS myelination. Sip1 represses differentiation inhibitory signals by antagonizing BMP receptor-activated Smad activity while activating crucial oligodendrocyte-promoting factors. Importantly, a key Sip1-activated target, Smad7, is required for oligodendrocyte differentiation and partially rescues differentiation defects caused by Sip1 loss. Smad7 promotes myelination by blocking the BMP- and ?-catenin-negative regulatory pathways. Thus, our findings reveal that Sip1-mediated antagonism of inhibitory signaling is critical for promoting CNS myelination and point to new mediators for myelin repair. ChIP-seq was performed to identify Olig2 direct target genes in oligodendrocytes during oligodendrocyte differentiation.

Expression data from mouse CD3+ T-cells from WT and KLF13 KO mice

Project description:Kruppel-like factors are a subclass of zinc finger transcription factors that play important roles in different aspects of cell growth, development, and differentiation. Our group have identified KLF13 as an essential transcription factor for the late expression of chemokine RANTES in T lymphocytes. However, very little is known about the role of KLF13 in T cells and other potential transcriptional targets. To address this, we sought to identify genes that are regulated by KLF13 in mouse T cells. Using microarray analysis, we compared gene expression in activated CD3+ T lymphocytes from wild type and Klf13-/- animals. We isolated CD3+ T-cells from WT and KO mouse spleen. Isolated cells were activated with CD3 and CD28 for 4 days and RNA was extracted. All samples were hybrdized and gene expression were analyzed with Affymetrix microarrays.

RNA-sequencing of Zdhhc5 knockout oligodendrocytes

Project description:Myelin sheath is an important structure to maintain normal functions of the nerves in central nervous system. Protein palmitoylation has been established as a sorting determinant for the transport of myelin-forming proteins to the myelin membrane. However, its function in the regulation of oligodendrocyte development remains unknown. Here, we show that an Asp-His-His-Cys (DHHC) motif-containing palmitoyl acyltransferases, DHHC5, is involved in the control of oligodendrocyte development. Loss of Zdhhc5 in oligodendrocytes inhibits myelination and remyelination by a reduction on total myelinating oligodendrocyte population. STAT3 is the primary substrate for DHHC5 palmitoylation in oligodendrocytes. Zdhhc5 ablation reduces STAT3 palmitoylation, and then suppresses STAT3 phosphorylation and activation. As a result of the decreased STAT3 transactivity, the transcription of the myelin-related and anti-apoptosis genes is therefore inhibited, leading to suppressed oligodendrocyte development and myelination. Our findings demonstrate the key role DHHC5 in the control of myelinogenesis.
