Project description:Anoxia induces several heat shock proteins and a heat pre-treatment can acclimatize Arabidopsis seedlings to a subsequent anoxic treatment. In this work we analyzed the response of Arabidopsis seedlings to anoxia, heat and a combined heat+anoxia stress. A significant overlapping between the anoxic and heat shock responses has been observed by whole-genome microarray analysis. Experiment Overall Design: We treated Arabidopsis seedling, 4-days old, dark germinated with: Experiment Overall Design: -Control (23°C, dark, liquid Murashige-Skoog medium containing 30mM sucrose). Experiment Overall Design: -Heat-treated (38°C for 90 minutes, dark, liquid Murashige-Skoog medium containing 30mM sucrose). Experiment Overall Design: -Anoxia-treated (23°C, under anoxia for 6h, dark, liquid Murashige-Skoog medium containing 30mM sucrose). Experiment Overall Design: -combined heat+Anoxia-treatment (23°C, treated at 38°C for 90 min and thereafter under anoxia for 6h, dark, liquid Murashige-Skoog medium containing 30mM sucrose). Project description:Dark grown Arabidopsis seedlings (Columbia gl1) were grown in the dark at 23°C for 4 days before adding 90 mM sucrose for 6h. Experiment Overall Design: Arabidopsis thaliana ecotype Columbia glabra (gl1-1) was used in this study. Seeds were sterilized with diluted bleach (10 min incubation in 1.7% sodium hypochlorite, rinsing and washing thoroughly in sterile water), and incubated in 2.5 ml liquid growing medium [Murashige-Skoog (MS) half strength solution] in 6-well plates (fully aerobic, with shaking). Plates were incubated in the darkness at 4°C for 2 days and finally transferred to 23°C for 4 days before the sugar treatments (with 90 mM sucrose, lasting 6h). Two independent, replicated experiments were performed for sucrose treatments. Each independent experiment consisted of four replicated seedlings cultures, pooled after RNA extraction. Project description:The ppi2 mutant introgressed into the Columbia-0 ecotype as previously reported. Doublemutant ppi2xrpn8a was crossed with the above referenced genotypes using heterozygous ppi2 plants. After 2 days of stratification at 4 °C the plants were grown on half-strength Murashige and Skoog (M&S) medium supplemented with 0.8% (w/v) plant agar (Duchefa) and 0.8% (w/v) sucrose under short day conditions.

Project description:rs05-05_atrazine - effect of atrazine and sucrose on arabidopsis transcriptome - Growth in the presence of sucrose was shown to confer to Arabidopsis thaliana seedlings, under conditions of in vitro culture, a very high level of tolerance to the herbicide atrazine and to other photosynthesis inhibitors (Sulmon et al., 2004). The CATMA investigation will be useful to reveal the gene networks implicated in the mechanisms of xenobiotics tolerance. - Seeds of Arabidopsis thaliana (ecotype Columbia) were surface-sterilized inbayrochlore/ethanol (1:1, v/v), rinsed in absolute ethanol and dried overnight. Germination and growth were carried out under axenic conditions in square Petri dishes. After seeds were sowed, Petri dishes were placed at 4degreeC for 48 h in order to break dormancy and homogenize germination, and then transferred to a control growth chamber at 22degreeC under a 16-h light period regime at 4500 lux for 4 weeks. Growth medium consisted of 0.8% (w/v) agar in 1x Murashige and Skoog (MS) basal salt mix (Sigma, St. Louis, MO, USA) adjusted to pH 5.7. After 4 weeks of cultivation on vertical plates plantlets were transferred on fresh medium complemented or not with atrazine 10 uM and with sucrose 80mM or mannitol 80 mM which were directly added during preparation of agar-MS media prior to sterilisation. Atrazine was sterilized by microfiltration through 0.2 um cellulose acetate filters (Polylabo, Strasbourg, France) and then axenically added to melted agar-MS medium prior to pouring into Petri dishes. Project description:To understand how plants respond to anoxia-reoxygenation, we have employed a global microarray expression profiling as a basic platform to characterize genes with the potential to mediate the recovery responses in Arabidopsis. 7-day-old seedlings were trated with 4hr and 8hr anoxia, and then reoxygenated in air for 0, 0.5, 1, 3, 6 hrs. Genes changed in both condition were designated as reoxygenation-regulated genes. They included the genes in ROS detoxification, dehydration, metabolic process, and many other responses. Interestingly, ethylene was also involved in this recovery process. We further adopted ein2-5 and ein3eil1 microarray to investigate ethylene signaling. Our results showed ethylene partially regulate reoxygenation regulated genes and is reruired for plant survival in reoxygenation. Genes expression during anoxia-reoxygenation in Arabidopsis seedlings was measured at 0, 0.5, 1, 3, 6 hours after anoxia treatment, and normal condition before anoxia. Three independent experiments were performed at each time (Nor, 0, 0.5, 1, 3, 6) under 4hr anoxia-reoxygenation condition by using Col-0, ein2-5, and ein3eil1. Four independent experiments were performed at each time (Nor, 0, 0.5, 1, 3, 6) under 8hr anoxia-reoxygenation condition by using Col-0.

Project description:The rainbow trout, Oncorhynchus mykiss, is relatively sensitive to hypoxia and does not survive deep hypoxia or anoxia. Given this lack of hypoxia-tolerance in the whole animal, the aim of this experiment was to investigate the in vitro responses of cultured rainbow trout cells following anoxia exposure. Rainbow trout hypodermal fibroblast (RTHDF) cells were exposed to anoxia for 12 and 24 h, whilst control cells were held under normoxic conditions for 24 h. Differential gene expression as a consequence of the treatments was analysed using an oligoarray composed of approximately 21,500 BLAST-identified sequences fabricated on the Agilent microarray platform. 57 genes were found to display significant responses to oxygen deprivation and these genes were assigned Gene Ontology terms and IDs. Of the 57 differentially expressed genes, 6 genes associated with carbohydrate metabolism were up-regulated following anoxia, these included phosphoglucomutase (PGM) and glycogen phosphorylase, consistent with glycogen serving as an important energy source during anoxia. Other genes up-regulated in response to anoxia included a number of putative targets of hypoxia inducible factor 1 alpha (HIF1A), namely phosphoglycerate kinase, triosephosphate isomerase and the glucose transporter, GLUT2. Egl9 homolog 3, the proline hydroxylase responsible for HIF1A regulation, was also induced. This analysis indicates that cultured trout cells have the capacity for adaptive gene responses when subjected to oxygen levels that are lethal to the whole organism.
