Single-cell transcriptome profiling of primary tumors and paired organoids of pancreatic cancer

ABSTRACT: Pancreatic cancer (PC) is the most lethal common gastrointestinal malignancy with an overall 5-year survival rate of 11%. Recently, a variety of patient-derived models, including patient-derived xenograft (PDX) and cancer organoid models, have paved a way for identifying common responders to chemotherapy. However, the utility of PC organoid for recapitulating hallmarks of corresponding primary tumor at transcriptome level remains to be investigated. We conducted single-cell RNA sequencing (scRNA-seq) for three paired primary tumors and organoids using the 10x Genomics platform to identify common and different phenotypes between them.
