The effect of fructose vs glucose supplementation on stable AKR1B1 knockdown in A549 lung cancer cells

ABSTRACT: To investigate the cytotoxic effect of AKR1B1 depletion, we induced stable AKR1B1 knockdown via shRNA under different growth conditions (glucose vs fructose) We then performed gene expression profiling analysis using data obtained from RNA-seq of control (non-targeting shRNA) or shAKR1B1 A549 cells 5 days after shRNA delivery.

SMARCA5 deficiency reprograms AKR1B1-mediated endogenous fructose metabolism to eradicate leukemia stem cells in acute myeloid leukemia

Project description:A significant variation in chromatin accessibility is an epigenetic feature in leukemia cells. However, the regulators and consequences of the chromatin accessibility variations in leukemia remain elusive. Here we identified SMARCA5, a core ATPase of the ISWI chromatin remodeling complex, as the primary regulator of chromatin accessibility in acute myeloid leukemia (AML) by CRISPR-Cas9 library screening. We found that SMARCA5 is required for self-renewal of leukemia stem cells and myeloid leukemogenesis. Mechanistically, SMARCA5 promotes the transcriptional activation of AKR1B1, an aldo/keto reductase, by recruiting transcription co-activator DDX5 and transcription factor SP1 to its promoter. Clinical sample analysis has shown that high expression of AKR1B1 predicts a poor prognosis in AML patients. Interestingly, AKR1B1-mediated endogenous fructose metabolism is reprogrammed by Smarca5 in AML. Overexpression of AKR1B1 rescued the leukemogenesis impaired by Smarca5 deficiency. Moreover, pharmacological inhibition of AKR1B1 displayed significant therapeutic effects in the AML patient-derived xenograft (PDX) mouse model. Thus, our findings link chromatin state dynamics regulated by SMARCA5 to AKR1B1-mediated endogenous fructose metabolism reprogramming, and uncover an essential role of AKR1B1 in AML, which may provide a therapeutic target for AML patients.

Metformin Impairs Intestinal Fructose Metabolism

Project description:Objective: To investigate the effects of metformin on intestinal carbohydrate metabolism in vivo. Method: Male mice preconditioned with a high-fat, high-sucrose diet were treated orally with metformin or a control solution for two weeks. Fructose metabolism, glucose production from fructose, and production of other fructose-derived metabolites were assessed using stably labeled fructose as a tracer. Results: Metformin treatment decreased intestinal glucose levels and reduced incorporation of fructose-derived metabolites into glucose. This was associated with decreased intestinal fructose metabolism as indicated by decreased enterocyte F1P levels and diminished labeling of fructose-derived metabolites. Metformin also reduced fructose delivery to the liver. Proteomic analysis revealed that metformin coordinately down-regulated proteins involved carbohydrate metabolism including those involved in fructolysis and glucose production within intestinal tissue. Conclusion: Metformin reduces intestinal fructose metabolism, and this is associated with broad-based changes in intestinal enzyme and protein levels involved in sugar metabolism indicating that metformin's effects on sugar metabolism are pleiotropic.

Transcriptome analysis of the pfkB1 deletion mutant cultivated with glucose or fructose

Project description:In the analysis of a carbohydrate metabolite pathway, we found that a mutant strain of Corynebacterium glutamicum deficient in pfkB1, which encodes fructose-1-phosphate kinase, showed interesting characteristics. After aerobically cultivated with fructose as a carbon source, this mutant consumed glucose and produced lactate more than 2-fold as compared with the wild-type under conditions of oxygen deprivation. This considerably higher fermentation capacity was unique for the combination of the pfkB1 deletion and fructose cultivation. On the basis of the metabolome and transcriptome analyses, we identified marked intracellular accumulation of fructose-1-phosphate and significant upregulation of several genes related to the phosphoenolpyruvate:carbohydrate phosphotransferase system, glycolysis, and organic acid synthesis in this strain. Therefore, the considerably enhanced glucose consumption and organic acid production presumably resulted from a relief of transcriptional repression driven by global regulator SugR owing to the accumulated fructose-1-phosphate. Furthermore, we demonstrated that engineered strains overexpressing the above upregulated genes showed enhanced glucose consumption and organic acid production. The ppc deletion mutant of the engineered strain consumed 1,250 mM glucose and produced 2,390 mM lactate in 48 h under oxygen deprivation, which are 2.6- and 2.7-fold higher than the corresponding parameters of the ppc-deleted wild-type.

Gene expression altered by high-fructose diet feeding and Chrebp deficiency

Project description:Epidemiologic and animal studies implicate overconsumption of fructose in the development of non-alcoholic fatty liver disease, but the molecular mechanisms underlying fructose-induced chronic liver diseases remains largely unknown. We present evidence supporting the essential function of the lipogenic transcription factor ChREBP in mediating adaptation response to fructose and protecting against fructose-induced hepatotoxicity. High-fructose diet (HFrD) activates hepatic lipogenesis via a ChREBP-dependent manner in wildtype mice, while inducing steatohepatitis in Chrebp-KO mice. In Chrebp-KO mouse livers, HFrD reduces levels of molecular chaperones and activates the CHOP-dependent unfolded protein response, whereas administration of chemical chaperone or Chop shRNA rescues liver injury. Gene expression profiling revealed elevated expression of cholesterol biosynthesis genes in Chrebp-KO livers after HFrD, in parallel with increased abundance of nuclear SREBP2. genes expression were compared between livers of wildtype mice fed 70%-fructose-diet v.s. regular chow, and between livers of Chrebp-/- mice v.s. wildtype mice fed 70%-fructose-diet.

Polyphenols protection against fructose overload

Project description:To decipher the beneficial effects of Polyphenols on health, healthy volunteers were randomized into two groups and were submitted to either a red grape polyphenol rich extract supplemented diet (PP) or a placebo diet (PCB) during 8 weeks. Then they were submitted to a fructose load (3g/kg Fat Free Mass/day) during 7 days. Muscle biopsies were taken before the protocol, 8 weeks after PP or PCB diet and after the 7 days fructose load. We have employed whole genome microarray expression profiling as a discovery platform to identify genes regulated by fructose and to identify the mechanism of action of Polyphenols. A Pilot Study to Determine Fructose Uptake by Primary Human Colorectal Tumors

Project description:This proposed study is designed to investigate the specific uptake of fructose by human colorectal tumors. In this study, subjects with colorectal cancer undergoing surgery will receive an oral sugar solution containing fructose or xylose prior to surgery. The tumor will then be resected, and a portion of the tissue will be used to measure the abundance of fructose and xylose. The study hypothesis is that the tumors will take up fructose sugar but not xylose sugar. A comparison of the sugar uptake between the tumor and normal tissues from the adjacent intestinal epithelium and smooth muscle and the liver will be conducted. This proposal will confirm that human colorectal cancer tumors can directly absorb dietary sugars, which has never been demonstrated.
