Levo-tetrahydropalmatine Ameliorates Neuropathic Pain via Suppressing the Activation of the Clec7a-MAPK/NF-κB-NLRP3 Inflammasome Axis

ABSTRACT: Due to the complex pathogenesis of neuropathic pain (NP), the therapeutic effect of existing drugs on NP is not satisfactory, which seriously affects the life quality of patients. Accumulating studies have indicated that neuroinflammation in the spinal cord may play a vital role in NP development and progression. Levo-tetrahydropalmatine (l-THP) has been extensively used for the treatment of a variety of chronic pain for decades. However, the analgesic effects and the underlying mechanisms of I-THP against NP have not been fully elucidated. To address this problem, we herein carried out RNA-seq and bioinformatics analyses to identify the effective target profiling of I-THP including 1113 upregulated and 132 downregulated genes in spinal cord obtained from Chronic constrictive injury (CCI) rats with or without I-THP administration.
