Project description:Circular DNAs have been reported for decades and have been linked to tumorigenesis in humans. Previous sequencing-based genomic approaches capture circular DNA and average their readout over millions of cells, thereby losing insight into intercellular variation. Here, we introduce single-cell Circle-seq and show that the occurrence of most circular DNA species is stochastic, with some oncogene-containing large circular DNAs observed relatively frequently among different single cells. Despite this stochasticity, circular DNA is cell type-specific, which is concordant with H3K9me3 and H3K4me3 chromatin modification specificity of the corresponding cell lines. Moreover, we identify a cell type-specific circular DNA signature in cancer cells, which, we believe holds potential for early cancer diagnosis. In sum, our novel method single-cell Circle-seq dissects the randomness and cell type-specificity of circular DNAs at the single-cell level and uncovers the potential clinical application of circular DNA.

Project description:Polytetrafluoroethylene (PTFE) fine powder with 93% crystallinity was irradiated by an electron beam (EB) at various temperatures under a nitrogen atmosphere. Trapped free radicals in PTFE were studied using electron spin resonance (ESR) spectroscopy. The observed spectra of the samples exposed to air after irradiation at various temperatures showed asymmetrical signals, which are middle-chain type peroxide macroradicals derived from fluoroalkyl radicals. The radical yields at each irradiation temperature increased with increasing absorbed dose, and eventually saturated. The higher irradiation temperature resulted in higher radical yields when compared at the same exposed dose. Furthermore, the G-value of the radicals (G(R·)) increases with increasing irradiation temperatures corresponding to each relaxation and transition temperature. It is concluded that the chain reaction by the fluorine extraction from the main chain due to the end-chain radical generated via β-scission after dissociative electron attachment (DEA) is enhanced by the synergistic effect of heat and radiation.

Project description:The influence of electron-beam irradiation on polypropylene (PP) and high-density polyethylene (HDPE) was investigated with a focus on crystallization. A high-temperature (200°C) creep test revealed that the HDPE gradually increased cross-linking density in the range 30-120 kGy, while the PP underwent a chain scission which was quantitatively evaluated by gel permeation chromatography. The mechanical properties were measured in the range -150 to 200°C by a dynamic mechanical analysis. A small presence of C=C and C=O bonds was found in the irradiated PP by a Fourier transform infrared spectroscopy. Crystallization kinetics measured by differential scanning calorimetry and hot-stage optical microscopy results were influenced tremendously by irradiation for HDPE and to a lesser extent for PP. Irradiation caused a decrease in both the number of nucleation centres and the growth rate of individual spherulites. Crystallization was analysed in detail with the help of Hoffman-Lauritzen, Avrami and Arrhenius equations. Interestingly an increasing β-crystal formation with an increasing irradiation level was discovered for PP by X-ray diffraction. A generation of defects in the crystalline structure owing to irradiation was discussed.

Project description:Transparent heaters have attracted increasing attention for their usefulness in vehicle windows, outdoor displays, and periscopes. We present high performance transparent heaters based on Ag nanowires with electron beam irradiation. We obtained an Ag-nanowire thin film with 48 ohm/sq of sheet resistance and 88.8% (substrate included) transmittance at 550 nm after electron beam irradiation for 120 sec. We demonstrate that the electron beam creates nano-soldering at the junctions of the Ag nanowires, which produces lower sheet resistance and improved adhesion of the Ag nanowires. We fabricated a transparent heater with Ag nanowires after electron beam irradiation, and obtained a temperature of 51 °C within 1 min at an applied voltage of 7 V. The presented technique will be useful in a wide range of applications for transparent heaters.

Project description:An electron beam was irradiated on polyacrylonitrile (PAN) fibers prior to thermal stabilization. The electron-beam irradiation effectively shortened the thermal stabilization process by one fourth compared with the conventional thermal stabilization process. A comprehensive mechanistic study was conducted regarding this shortening of the thermal stabilization by electron-beam irradiation. Various species of chain radicals were produced in PAN fibers by electron-beam irradiation and existed for a relatively long duration, as observed by electron spin resonance spectroscopy. Subsequently, these radicals were gradually oxidized to peroxy radicals in the presence of oxygen under storage or heating. We found that these peroxy radicals (CO) enabled such an effective shortcut of thermal stabilization by acting as intermolecular cross-linking and partial aromatization points in the low temperature range (100-130?°C) and as earlier initiation seeds of successive cyclization reactions in the next temperature range (>130-140?°C) of thermal stabilization. Finally, even at a low irradiation dose (200?kGy), followed by a short heat treatment (230?°C for 30?min), the PAN fibers were sufficiently stabilized to produce carbon fibers with tensile strength and modulus of 2.3 and 216?GPa, respectively, after carbonization.

Project description:This research investigated the effect of irradiation with an electron beam energy of 10 MeV in doses of 26-156 kGy on polytetrafluoroethylene (PTFE) with a 15% and 20% graphite additive. The research has shown that mechanical (compression strength, hardness, and Young's modulus) and sclerometric (coefficient of wear micromechanism and coefficient of resistance to wear) properties improve and tribological wear decreases as graphite content increases. Electron beam irradiation increases the degree of crystallinity of both materials to a similar extent. However significant differences in the improvement of all examined properties have been demonstrated for PTFE with higher (20%) graphite content subjected to the electron beam irradiation. This polymer is characterized by higher hardness and Young's modulus, reduced susceptibility to permanent deformation, higher elasticity, compression strength, and above all, a nearly 30% reduction in tribological wear compared to PTFE with a 15% graphite additive. The most advantageous properties can be obtained for both of the examined composites after absorbing a dose of 104 kGy. The obtained results hold promise for the improvement of the operational life of friction couples which do not require lubrication, used for example in air compressors and engines, and for the possibility of application of these modified polymers. Project description:Electron beam irradiation is an efficient cold sterilization technique that has been widely utilized in food preservation. To understand the biological and morphological changes of microorganisms by electron beam irradiation, the D10 value, growth curve, and morphological characterization by scanning electron microscope (SEM) of Staphylococcus aureus were studied. Furthermore, Tandem mass tag (TMT)-based quantitative proteomic was performed to investigate the responses of Staphylococcus aureus to electron beam. The results showed that electron beam sensitized the irradiated strain and prolonged the lag phase of bacterial growth. There were 53, 96, 1055, and 431 differentially expressed proteins (DEPs) detected in 1-kGy irradiated strain (0 h-1-kGy), 2-kGy irradiated strain (0 h-2-kGy), 1-kGy irradiated strain after 24 h cultivation (24 h-1-kGy), and 2-kGy irradiated strain after 24 h cultivation (24 h-2-kGy) groups compared to unirradiated strain (control) group, respectively. Most of DEPs involved in DNA repair were found to be up-regulated. However, DEPs involved in biofilm formation and staphylococcal enterotoxin production were down-regulated. There were no noticeable morphological changes observed in irradiated S. aureus. These findings provided valuable information in the responses of S. aureus to electron beam irradiation, leading to a better understanding of the effects of electron beam irradiation on food pathogens.

Project description:Electron-beam irradiation (EBI) is a cold sterilization technology used in the irradiation processing of food, including rice. Herein, the effects of EBI on the swelling power, color, pasting, and sensory properties of white rice after short-term storage were analyzed. Samples were electron-beam irradiated at 0, 2, 4, 6, or 8 kGy and stored at 25 °C or 37 °C for up to 75 days. Results showed that swelling power and major pasting viscosities (including peak, breakdown, and setback viscosities) at both storage temperatures decreased with increased irradiation dose. Negative correlations were also observed between the major viscosities of pasting properties and irradiation dose at both storage temperatures. During sensory evaluation, extremely low scores for rice hardness, appearance, taste, and overall acceptability were obtained for rice subjected to high EBI dose (>4 kGy). However, rice stored at 37 °C showed lower performance than rice at 25 °C in terms of the abovementioned parameters. By contrast, the sensory properties at irradiation doses between 2 and 4 kGy were better than those of the control group at both storage temperatures. All these findings indicated the potential of low-dose (<4 kGy) EBI as pretreatment for improving the quality of white rice during storage.
