Characterizing transcriptomic changes at different stage of hepatobiliary reprogramming in the adult mouse liver

ABSTRACT: Recent studies have demonstrated that hepatocytes can be reprogrammed into biliary epithelial cell-like cells. Here, by RNA-seq, we dissect the transcriptomic changes involving the reprogramming process at different stages.

However, the epigenetic basis underlying this phenomenon is largely unknown. Here, we profiled cells after knockout of Nsd1 or Kdm2a at two reprogramming stages to assess the transcriptional differences imposed by epigenetic dysregulation.

Project description:Following liver injury, a subset of hepatocytes adopt features of biliary epithelial cells (BECs) in a process known as biliary reprogramming. The aim of this study is to elucidate the molecular events accompanying this dramatic shift in cellular identity. Wild type hepatocytes, wild type biliary cells, DDC-treated hepatocytes, DDC-treated biliary cells, and DDC-treated reprogrammed cells were isolated by FACS, RNA isolated, and sequenced.

Transcriptomic characterization of hepatobiliary plasticity of hepCLiPs and bilCLiPs

Project description:Recent studies have demonstrated extensive proliferative capacity of adult hepatocytes and biliary epithelial cells (BECs) in vitro, thereby allowing derivation of clonal cell lines named hepCLiPs and bilCLiPs, respectively. Both cell lines undergo hepatobiliary plastiticity in that they exhibit a BEC-like phenotype under a 2D culture condition, while they exhibit a hepatocyte-like phenotype under a 3D culture condition. This data set was obtained to comprehensively characterize the gene expression profiles of hepCLiPs and bilCLiPs which were cultured under 2D and 3D culture conditions.
