Transcription profiling of mouse prostate after castration and hormone replacement

Project description:In this study, we used microarray analysis to determine gene expression profile changes in the mouse prostate following castration and hormone replacement. We first identified genes with significant expression changes in each of these two processes and then generated a list of androgen responsive genes and a list of genes whose expression were inversely correlated with the presence of androgen. The analysis of this data set is described in Wang et al., Differentiation, 2006 Experiment Overall Design: C57/B6 mice of 10-12 weeks old (Charles River, Hollister, CA) were used. Mice were castrated, and at 3 or 14 days after surgery (C3 or C14), mice from these groups were sacrificed and whole prostates were collected. Prostates were also collected from sham-treated mice for control (N3). To obtain re-growing or regenerating prostate, mice that had been castrated 14 days before were implanted with testosterone (T) pellet (15 mg/pellet/mouse, Innovative Research, Sarasota, FL). Three days after pellet implantation, mice were sacrificed and prostates were collected (C14+T3). Five mice were used for each treatment/time point. Total RNA was purified with RNeasy kit.

Hemodialysis exacerbates proteolytic imbalance and pro-fibrotic platelet dysfunction.

Project description:Multi-organ fibrosis among end stage renal disease (ESRD) patients cannot be explained by uremia alone. Despite mitigation of thrombosis during hemodialysis (HD), subsequent platelet dysfunction and tissue dysregulation are less understood. We comprehensively profiled plasma and platelets from ESRD patients before and after HD to examine HD-modulation of platelets beyond thrombotic activation. Basal plasma levels of proteolytic regulators and fibrotic factors were elevated in ESRD patients compared to healthy controls, with isoform-specific changes during HD. Platelet lysate (PL) RNA transcripts for growth and coagulative factors were elevated post-HD, with upregulation correlated to HD vintage. Platelet secretome correlations to plasma factors reveal acutely induced pro-fibrotic platelet phenotypes in ESRD patients during HD characterized by preferentially enhanced proteolytic enzyme translation and secretion, platelet contribution to inflammatory response, and increasing platelet dysfunction with blood flow rate (BFR) and Vintage. Compensatory mechanisms of increased platelet growth factor synthesis with acute plasma matrix metalloproteinase (MMP) and tissue inhibitor of MMPs (TIMP) increases show short-term mode-switching between dialysis sessions leading to long-term pro-fibrotic bias. Chronic pro-fibrotic adaptation of platelet synthesis were observed through changes in differential secretory kinetics of heterogenous granule subtypes. We conclude that chronic and acute platelet responses to HD contribute to a pro-fibrotic milieu in ESRD.

Liver tissue from hormone treated C57BL/6J mice versus pool C57BL/6J mice of chow diet Liver

Project description:The purpose of this experiment was to determine the expression traits in animals of inbred strain C57BL/6J, treated with gonadal hormones. (N=40, 20 males and 20 females). Liver tissue from the hormone treated group consists of 40 female and male C57BL/6J mice fed a chow diet containing 4% fat (Ralston-Purina Co., St. Louis, MO) until 8 weeks of age and then were gonadectomized at 8 weeks of life, implanted with hormone pellets at 12 weeks, and sacrificed at 14 weeks. Male and female mice of the hormone treated groups received subcutaneous implants of either 0.5 mg estradiol (E2) pellet (plasma yield of 300 picogram/ml) or a 5 mg dihydrotestosterone (DHT) pellet (plasma yield of 1-2 nanogram/ml), designed to release over 21 days, (Innovative Research of America, 17B-estradiol: Cat. No. E-121 0.5mg/pellet, 5a-dihydrotestosterone: Cat. No. A-16). Control mice were treated with the placebo pellet (Innovative Research of America, placebo: Cat. No. C-111). At 14 weeks mice were sacrificed, after a 12-hour fast, Liver tissue were dissected and flash frozen in LN2 and stored at -80Â°C. YAP mediates TNF effect on pro-fibrotic genes expression

Project description:YAP/TAZ are transcription co-factors recently described responsive to pro-inflammatory cytokines and involved in inflammatory-related disorders. However, the role of tumor necrosis factor (TNF), a major pro-inflammatory cytokine, on YAP signaling is not well understood and controversial. Here, we observe in vitro, using wild type and YAP knockout HEK293 cells, that TNF triggers YAP nuclear translocation and transcriptional activity, thus being dependent on Rho family of GTPases. In response to TNF, YAP transcriptional activity orientates cell fate toward survival. Transcriptional analysis with Nanostring technology reveals that YAP modulates TNF-induced increase in fibro-inflammatory pathways such as NF-κB, inflammasomes, cytokines or chemokines signaling as well as pro-fibrotic pathways involving TGF-β and extracellular matrix remodeling. Therefore, in response to TNF, YAP acts as a sustainer of the inflammatory response and as a molecular link between inflammation and fibrotic processes. This work identifies that YAP is critical to drive several biological effects of TNF which are involved in cancer and inflammatory disorders.

Estrogen and high-fat diet induced alterations of endometrial transcriptome profile in ovariectomized C57BL/6 mice

Project description:Unopposed estrogen stimulation and insulin resistance are known to play important roles in endometrial cancer (EC), but the relationship between these two factors and how they contribute to endometrial lesions is not completely understood. To investigate the endometrial transcriptome profiles and the associated molecular pathway alterations, we established an ovariectomized C57BL/6 mouse model treated with subcutaneous implantation of the 17-β estradiol (E2) pellet and/or high-fat diet (HFD) to mimic sustained estrogen stimulation and insulin resistance. We used microarrays to detail the global program of gene expression underlying the mice endometrium and indentified distinct classes of E2 and/or high-fat diet related ncRNA during this process.
