Transcriptomics and proteomics approach for the identification of altered blood microRNAs and plasma proteins in Parkinson’s disease by Custom Brain Specific miRNA OpenArray Real-time PCR panel

ABSTRACT: The blood samples of PD patients and Healthy controls were collected from the general OPD of Department of Neurology, KGMU, Lucknow. RNA (including small RNAs) was extracted by Ribopure Blood RNA Isolation Kit by Thermo Fisher as suggested protocol by manufacturer. A custom Taqman OpenArray panel was used to profile the miRNAs expression.

Project description:12 week male wistar rats were exposed with rotenone (2.5mg/kg b.wt.) and control rats exposed with vehicle (corn oil) intraperetonially for two months. Gene expression profiling of blood in ruptured intracranial aneurysms

Project description:In the present study we aimed to investigate the systemic response to a rupture of intracranial aneurysms by an analysis of global gene expression profiles in peripheral blood cells. In addition, we sought to determine whether this approach could provide biomarkers related to clinical status of subarachnoid hemorrhage patients. Patients with subarachnoid hemorrhage from ruptured aneurysm were prospectively recruited from patients consecutively admitted to the Departments of Neurology or Neurosurgery and Neurotraumatology, University Hospital, Krakow, Poland in 2010 and 2011. Control subjects were recruited from patients of the Department of Neurology suffered from headaches.

Project description:Total RNA was extracted from 400 ul of serum with the miRNeasy Serum/Plasma advanced Kit (Qiagen) and quantified using the Qubit microRNA assay kit (Thermo Fisher Scientific). cDNA templates were prepared using the TaqMan Advanced miRNA cDNA Synthesis Kit (Thermo Fisher Scientific), starting from 10 ng of RNA. RT-qPCR carried out on a QuantStudio 12K Flex (Applied Biosystems) using the TaqMan OpenArray miRNA panel.

A retrospective clinical study to explore diagnostically significant markers in circulating exosomes of patients with cerebral small vessels disease (CSVD)

Project description:Often characterized by insidious onset, CSVD is the leading cause of vascular dementia.Nonetheless, early screening and diagnosis of CSVD remain challenging. Exosomal miRNAs in circulation have been found to be diagnostic markers for many diseases and to play an essential role in neurological disorders. The research will use peripheral blood samples from CSVD patients hospitalized and relatively healthy outpatients in our neurology department from 2018 to 2020. Specifically, microarray results from patients with CSVD will be obtained and then validated in peripheral blood samples, aiming to investigate abnormal expression indicators in circulating exosomes of patients with CSVD, for further mechanism exploration and diagnosis of CSVD.

Effect of milk intake on circulating plasma miRNAs of adult humans

Project description:This study examines circulating plasma miRNAs of adult human subjects before and after bovine milk intake. We sought to confirm and expand findings previously reported by Baier, et al., J Nutr, 2014, PMID 25122645, using the same samples. Plasma samples were provided by the Zempleni lab (see Baier, et al., J Nutr, 2014, PMID 25122645) and had been taken from five healthy donors prior to milk intake (T0) and at three hours post-intake (T3). miRNA profiles were generated for each sample by OpenArray (Thermo Fisher).
