Tire-wear particle leachate toxicity to Americamysis bahia: analysis of sublethal and molecular effects

ABSTRACT: Tire-wear particles (TWPs) are considered among the largest contributors of microplastics to the environment. They are subject to break-down due to environmental weathering, which allows for potentially toxic chemicals to be leached from and sorbed onto the particles. In this study, leachate generated from “weathered” and “un-weathered” TWPs were used for sublethal toxicity tests with Americamysis bahia.

Selenomethionine against titanium particle-induced osteolysis by regulating the ROS-dependent NLRP3 inflammasome activation via the β-catenin signaling pathway

Project description:Wear debris-induced osteolysis, especially titanium (Ti) particles-induced osteolysis, is the most common cause of arthroplasty failure with no effective therapy. Previous studies have suggested that inflammation and impaired osteogenesis are associated with Ti particles -induced osteolysis. Selenium (Se) is an essential trace element in the human body, which forms selenomethionine (Se-Met) in nature, and selenoproteins has strong anti-inflammatory and antioxidant stress effects. In this study, the effects of Se-Met on Ti particles-induced osteolysis were observed and the potential mechanism was explored. We found that exogenous Se-Met relieved osteolysis induced by Ti particles in two animal models and MC3T3-E1 cells. We found that the addition of Se-Met effectively inhibited Ti particle-induced inflammation by regulating ROS-dependent NLRP3 inflammasome activation. These therapeutic effects were abrogated in MC3T3-E1 cells that had received a β-catenin antagonist, suggesting that Se-Met alleviates inflammatory osteolysis via the β-catenin signaling pathway. Collectively, these findings indicated that Se-Met may serve as a potential therapeutic agent for treating Ti particle-induced osteolysis.

Gene expression profiling in wear-particle induced and infectious endoprosthesis loosening

Project description:The aim of the study was to identify markers for the early diagnosis of endoprosthesis loosening, for the differentiation between wear-particle induced and septic loosening, as well as to gather new insights into the pathogenesis. 824 genes were differentially expressed with a fold change greater than 2. Among these were Chitinase 1, CD52, Calpain 3, Apolipoprotein, CD18, Lysyl oxidase, Cathepsin D, E-Cadherin, VE-Cadherin, Nidogen, Angiopoietin 1 and Thrombospondin 2, and the differential expression levels were validated by RT-PCR. The chitinase activity was significantly higher in the blood from patients with wear-particle induced prosthesis loosening (p=0.001). However, using chitinase activity as a marker for early diagnosis, it has a specificity of 83% and a sensitivity of only 52%, due to a high variability both in the disease and in the control group. Project description:Purpose: In order to clarify the molecular response mechanism of Tetranychus truncatus to sublethal concentrations of abamectin. Methods: We sequenced the transcriptome of Tetranychus truncatus treated with sublethal concentration of abamectin and control. Results: we provide a comprehensive transcriptome resource for Tetranychus truncatus derived from treating with sublethal concentration abamectin and control. Conclusions: The new genomic resources and data that we present in this study for Tetranychus truncatus will substantially facilitate molecular studies of underlying mechanisms involved in abamectin resistance.

Spinetoram sublethal exposure stimulates Dengue virus replication in Aedes aegypti by dysregulating oxidative phosphorylation

Project description:Vector-borne diseases are closely linked to the environment by the ecology of the vectors and their hosts. Elucidating these causal relationships is one of the most pressing challenges faced by researchers and public health scientists as increasing anthropogenic alternations in the environment can drive shifts in vector-borne disease dynamics. However, as one of the major environmental stressors, the impact of sublethal exposure of insecticides on vector behavior remains poorly investigated. Here, we analyzed how sublethal exposure of the promising vector-control bioinsecticide spinetoram on Aedes aegypti larvae, the latest generation of the spinosyns that have been suggested as a promising new bioinsecticide for vector control, at the larval stage of Ae. aegypti alters adult performance and susceptibility to dengue virus (DENV) infection. While spinetoram sublethal exposure leads to extended immature development time, these survivors were significantly smaller and exhibited weaker blood-feeding capacity than normal females. More importantly, surviving females presented higher DENV susceptibility than the control group after spinetoram sublethal exposure on larvae. Mechanistically, the transcriptomic analysis showed that inhibition of oxidative phosphorylation (OXPHOS) may function in stimulating DENV production in adult Ae. aegypti. In vitro study revealed that spinetoram induces apoptosis via mitochondrial reactive oxygen species (mtROS)-regulated OXPHOS dysregulation in Aag2 cells, and thereby may stimulate DENV production through the metabolic switch between OXPHOS and glycolysis. Altogether, our data demonstrate that sublethal spinetoram exposure could act as a crucial factor on life traits and vector competence in Ae. aegypti. Given that other insecticides are known to induce mitochondrial dysfunction in mosquito vectors, this research may have wider implications.

Assessment of transcriptomic and apical responses of Daphnia magna exposed to a 3 polyethylene microplastic in a 21-day chronic study

Project description:There is global concern regarding the fate and effects of microplastics in the environment, particularly in aquatic systems. In this study, ethylene acrylic acid copolymer particles were evaluated in a chronic toxicity study with the aquatic invertebrate, Daphnia magna. The study design included a natural particle control treatment (i.e., silica) in order to discern any potential physical effects of a particlefrom intrinsic toxicity of the test material. In addition to the standard endpoints of survival, growth, and reproduction, the transcriptomic profile of control and ethylene acrylic acid copolymer-exposed D. magna were evaluated at the termination of the 21-day toxicity study. No significant effects on D. magna growth, survival, or reproduction were observed in the study in comparison to both particle and untreated control groups. Significant transcriptomic alterations were induced in the highest treatment level of 2.3 x 1012 particles of the ethylene acrylic acid copolymer/ L in key pathways linked to central metabolism and energy reserves, oxidative stress, as well as ovulation and molting indicating a global transcriptomic response pattern. To put the results in perspective is challenging at this time, since, to date, microplastic environmental monitoring approaches have not been equipped to detect particles in the nano size range. However, the results of this study indicate that ethylene acrylic acid copolymer microplastics in the upper nano-size range are not expected to adversely affect D. magna growth, survival, or reproductive outcomes at concentrations up to 1012 particles/L.
