Next-generation sequencing of wild type and CD4 conditional Bob1 deficient follicular helper T cells

ABSTRACT: Accumulating evidence has shown that a transcriptional coactivator of B-cell Oct-binding protein-1 (Bob1), in which Oct represents a DNA binding molecule specific to the octamer motif with an 8-bp element, ATTTGCAT, is preferentially expressed in human and mouse foollicular helper T (Tfh) cells. Meanwhile, the mechanism of how Bob1 contributes to the control of Tfh cells remains to be addressed. Studies using T-dependent antigens further showed that Bob1 would control the maintenance and development of memory Tfh cells from the central memory T-cell pool to sustain specific humoral responses. This was probably attributable to the regulatory program by Bob1 in energy metabolism as well as the expression of ICOS and IL-4. Collectively, Bob1 has the fundamental capacity to regulate long-term responses of Tfh cells to maintain specific humoral immunity.
