Project description:We systematically evaluated the expression pattern and function of ENO1 in AML. Knockdown ENO1 significantly inhibited ERK pathway phosphorylation, ERK pathway inhibitors significantly inhibited proliferation, clonal formation, migration and invasion of AML cells in vitro, blocked cell cycle and promoted apoptosis of AML cells, and activators of ERK pathway could reverse the effects of knockdown ENO1 on AML cells. The results showed that knockdown ENO1 inhibited the proliferation and invasion of AML cells in mice by inhibiting the activation of ERK pathway, and induced a significant increase in the overall survival rate of mice. Overall, this study aims to explore the role of ENO1 in the development and progression of AML and its potential mechanism, in order to provide a potential target for the treatment of AML.

Project description:cMyc plays a major role in lymphoma development. In addition, missense mutations including T58A and T58I the substitution mutations augment cMyc activity during lymphoma development. In this study, we characterized lymphoma-associated gene expression changes as increasing levels of wild type or mutant cMyc proteins progressively drive conversion of B-cells to lymphoma-like cells. To this end we established a cell system in which primary mouse B-cells were transduced with constructs that over-express the anti-apoptotic proteins, BMI1 and BCLXL and allow variable expression of cMyc (wild type, T58A or T58I) from a doxycycline-regulated promoter. Increased cMyc expression is associated with cell cycle entry, increased cell size and increased cell proliferation. Generally, gene programs required for cell growth and proliferation are induced while specific functions associated with B-cells are repressed. Mutant cMyc proteins cause differential effects, relative to each other and to wild type cMyc, that are focused on particular aspects of the cMyc-induced program.

Project description:Enolase 1 (ENO1) is a glycolytic enzyme that plays essential roles in various pathological activities including cancer development. However, the mechanisms underlying ENO1-contributed tumorigenesis are not well explained. Here, we uncover that ENO1, as an RNA-binding protein, binds to the cytosine-uracil-guanine-rich elements of YAP1 messenger RNA to promote its translation. ENO1 and YAP1 positively regulate alternative arachidonic acid (AA) metabolism by inverse regulation of PLCB1 and HPGD (15-hydroxyprostaglandin dehydrogenase). The YAP1/PLCB1/HPGD axis-mediated activation of AA metabolism and subsequent accumulation of prostaglandin E2 (PGE2) are responsible for ENO1-mediated cancer progression, which can be retarded by aspirin. Finally, aberrant activation of ENO1/YAP1/PLCB1 and decreased HPGD expression in clinical hepatocellular carcinoma samples indicate a potential correlation between ENO1-regulated AA metabolism and cancer development. These findings underline a new function of ENO1 in regulating AA metabolism and tumorigenesis, suggesting a therapeutic potential for aspirin in patients with liver cancer with aberrant expression of ENO1 or YAP1.

Project description:We performed a biological and molecular characterization of the novel human multiple myeloma cell line CMA03/06, an IL-6-independent variant of CMA03 cell line previously established in our Institution. We showed that the CMA03/06 cells grows in the absence of IL-6 with a doubling time comparable to CMA03; the addition of IL-6 to the culture medium or co-culture with multipotent mesenchymal stromal cells does not induce an increase in proliferation rate. Interestingly, we provided evidence that IL-6 independence of CMA03/06 cells is not a consequence of the development of an autocrine IL-6 loop, even though the cells maintain the IL-6 signaling pathway responsiveness as demonstrated by STAT1 and STAT3 induction by IL-6. A slight constitutive activation of STAT3 has been observed in CMA03/06 cell line; however STAT3 silencing in CMA03/06 cells did not affect their viability and proliferation suggesting that this pathway is not the only responsible for the IL-6 independency of CMA03/06 cell line. The new cell line showed a lower susceptibility to camptothecin-induced apoptosis compared to CMA03 cells, whereas an increased induction of apoptosis was observed in CMA03/06 cell line after Bortezomib treatment which may suggest the involvement NF-kB pathway in the IL-6 independent growth and survival of CMA03/06 cells. Finally, global gene expression profiling analysis allowed the identification of a list of 308 modulated genes in CMA03/06 versus CMA03 cells, many of which particularly relevant for MM biology. Overall, the novel CMA03/06 cell line may represent a suitable model for studies investigating molecular mechanisms involved in clonal evolution towards IL-6 and/or stroma-independent growth and survival of myeloma cells.

Project description:Purpose:Sipunculus nudus is a marine organism with high application value, and the fibrinolytic enzyme (SNFE) extracted from the visceral homogenate has been proved to have an antithrombotic effect by experiments. MicroRNAs(MiRNAs) play an important role in the molecular mechanisms related to plasmin. However, the research on plasmin-related miRNA of Sipunculus nudus is relatively rare. Methods:In this paper, high-throughput sequencing technology was used to sequence and analyze the small RNA and transcriptome of Sipunculus nudus. Conclusion:After data analysis, 198 known miRNAs were identified and 46 new miRNAs were predicted, followed by statistical analysis of miRNA base preference. At the same time, 76 miRNAs differentially expressed in different tissues were found, and a total of 76 known and new miRNAs were predicted for target genes, from which 7,161 candidate target genes were obtained. In addition, we found that six plasmin-related target genes were significantly annotated with 28 GO terms, and three plasmin-related target genes were rich in eight KEGG biological pathways. Finally, four joint regulatory networks including four candidate target genes, 32 differentially expressed miRNA and related proteins were constructed according to the four related species, and it was found that there existed interactions between the candidate target genes Cluster-15745.8440 and Cluster-3821.0 with unclear mechanisms.This study will provide a reference for in-depth exploration of the regulatory relationship between the plasmin-related genes and miRNA in Sipunculus nudus and understanding the molecular mechanisms related to plasmin in Sipunculus nudus.
