Project description:Purpose: Microgravity is one of crucial factors which affects astronauts’ health in space. The goals of this study are to compare SMG-pretreated macrophages infected with EPEC by RNA-sequencing to investigate the immunity-related differential expression genes and signaling pathway and to make for prevention and therapy of infectious diseases in space. Methods: 3-day SMG-pretreatment macrophages were infected with EPEC (a kind of enteropathogen) and macrophages infected with EPEC under normal gravity as control and then we used RNA sequencing on Illumina platform. The sequence reads that passed quality filters were analyzed at the transcript isoform level by Volcano Plot, KEEG database and InnateDB database. qRT–PCR validation was performed using SYBR Green assays. Results: After RNA sequencing, we got 1272 differential expression genes (DEGs) of which 542 were up-regulated, 720 were down-regulated DEGs. KEGG pathway enrichment scatter plot showed that cytokine-cytokine receptor interaction and MAPK signaling pathway were optimal-related to infection immunity under simulated microgravity. Because of our study involved in infection immunity, we chose a database on immunity called InnateDB to screen the DEGs related to immunity. We found 14 DEGs in cytokine-cytokine receptor interaction, of which 7 were up-regulated, and 7 were down-regulated and 36 DEGs in MAPK signaling pathway, of which 11 were up-regulated, and 25 were down-regulated. In cytokine-cytokine receptor interaction signaling pathway, except for Il 33 (P>0. 05), the results of mRNA expression of DEGs in macrophages infected with EPEC were consistent with the results of RNA sequencing. In MAPK signaling pathway, the mRNA expression of Il 1b, Cd14, Hspa1a, Pdgfrb, Il 1a, Fgf7, Igf1r and Pak1 was not consistent with the RNA sequencing. Compared with normal gravity, simulated microgravity didn’t affect the mRNA expression of Il 1b, Cd14, Hspa1a and Pdgfrb (P>0. 05). The Il 1a and Fgf7 down regulated and Igf1r and Pak1 up regulated under simulated microgravity at the level of mRNA with a comparison to normal gravity, and the results of the rest of DEGs were consistent with the results of RNA sequencing. Conclusions: This study is the first high-throughput analysis on simulated microgravity of infection immunity. The comparative analysis demonstrates that cytokine-cytokine receptor interaction and MAPK signaling pathway participants in the innate immune response.

Project description:To reveal the potential mechanisms involved in the dysfunction of antiviral immune responses under simulated microgravity conditions, we investigated the transcriptional changes related to the status of innate immune responses by RNA-seq with poly I:C or mock PBS treatment under Normal gravity or simulated microgravity conditions. Our results indicate that the retinoic acid inducible gene (RIG)-I-like receptor (RLR) and Toll-like receptor (TLR) signal pathways, which are both involved in the type-I interferon induction, are significantly inhibited by simulated microgravity effects.

Project description:Regenerative life support systems for space crews recycle organic and inorganic waste into water, food and oxygen using different organisms. For instance, the European Space Agency's MELiSSA uses Limnospira indica PCC8005 for air revitalisation and food production. Before use in space, the components' compatibility with reduced gravity must be tested. This innovative study introduces a novel ground analog designed specifically for microgravity experiments involving cyanobacteria, employing a cutting-edge random positioning machine (RPM). Limnospira indica PCC8005 was shown to grow slower under simulated microgravity and whole proteome analysis revealed a downregulation of e.g. ribosomal proteins, glutamine synthase and nitrate uptake transporters while an upregulation was found for gas vesicle proteins, carboxysome proteins and phycobiliproteins. All together our results suggested that L. indica experienced carbon limitation when cultivated in simulated microgravity conditions.

Project description:Astronauts have been previously shown to exhibit decreased salivary lysozyme and increased dental calculus and gingival inflammation in response to space flight, host factors that could contribute to oral diseases such as caries and periodontitis. However, the specific physiological response of caries-causing bacteria such as Streptococcus mutans to space flight and/or ground-based simulated microgravity has not been extensively investigated. In this study, High Aspect Ratio Vessel (HARV) S. mutans simulated microgravity and normal gravity cultures were assessed for changes in metabolite and transcriptome profiles, H2O2 resistance, and competence in sucrose-containing biofilm media. Stationary phase S. mutans simulated microgravity cultures displayed increased killing by H2O2 compared to normal gravity control cultures, but competence was not affected. RNA-seq analysis revealed that expression of 153 genes was up-regulated ≥ 2-fold and 94 genes down-regulated ≥ 2-fold during simulated microgravity HARV growth. These included a number of genes located on extrachromosomal elements, as well as genes involved in carbohydrate metabolism, translation, and stress responses. Collectively, these results suggest that growth under microgravity analog conditions promotes changes in S. mutans gene expression and physiology that may translate to an altered cariogenic potential of this organism during space flight missions.

Project description:Background/Objectives: The waterflea Daphnia is an interesting candidate for biore- generative life support systems (BLSS). These animals are particularly promising be- cause of their central role in the limnic food web and its mode of reproduction. How- ever, the response of Daphnia to altered gravity conditions has to be investigated, especially on the molecular level, to evaluate the suitability of Daphnia for BLSS in space. Methods: In this study, we applied a proteomic approach to identify key proteins and pathways involved in the response of Daphnia to simulated microgravity gener- ated by a 2D-clinostat. We analysed 5 biological replicates using 2D-DIGE proteomic analysis. Results: We identified 109 protein spots differing in intensity (p < 0.05). Substan- tial fractions of these proteins are involved in actin microfilament organisation, in- dicating the disruption of cytoskeletal structures during clinorotation. Furthermore, proteins involved in protein folding were identified, suggesting altered gravity in- duced break-down of protein structures in general. In addition, simulated micro- gravity increased the abundance of energy metabolism related proteins, indicating an enhanced energy demand of Daphnia. Conclusion: The affected biological processes were also described in other studies using different organisms and systems either aiming to simulate microgravity con- ditions or providing real microgravity conditions. Moreover, most of the Daphnia protein sequences are well conserved throughout taxa, indicating that the response to altered gravity conditions in Daphnia follows a general concept.

Project description:Cellular and molecular dynamics of human cells are constantly affected by gravity. Alteration of the gravitational force disturbs the cellular equilibrium, which might modify physiological and molecular characteristics. Nevertheless, biological responses of cancer cells to reduced gravitational force remains obscure. Here, we aimed to comprehend not only transcriptomic patterns but drug responses of colorectal cancer (CRC) under simulated microgravity. We established four organoids directly from CRC patients, and organoids cultured in 3D clinostat were subjected to genome wide expression profiling and drug library screening. Our observations revealed changes in cell morphology and an increase in cell viability under simulated microgravity compared to their static controls. Transcriptomic analysis highlighted a significant dysregulation in the TBC1D3 family of genes. The upregulation of cell proliferation observed under simulated microgravity conditions was further supported by enriched cell cycle processes, as evidenced by the functional clustering of mRNA expressions using cancer hallmark and gene ontology terms. Our drug screening results indicated an enhanced response rate to 5-FU under conditions of simulated microgravity, suggesting potential implications for cancer treatment strategies in simulated microgravity.

But, regulation and modulation of smallRNAs in pathology conditions and normal conditions are poorly recognized, supremely in wound care management. Current study focused to identify the smallRNA regulatory network in simulated microgravity sensitized Human Umbilical Cord Vein Endothelial cells (HUVECs) and gravity (1g) as a background. HUVECs were purchased from Lonza (Cat.No) and cultured in EGM2 medium (Cat.No CC3162) supplemented with 10% Fetal Bovine, 1% penicillin-streptomycin (W/V). Cells were cultured in RPM (Randomized positioned machine) for 2 hours at 37°C.

Project description:Microgravity is known to affect the organization of the cytoskeleton, cell and nuclear morphology and to elicit differential expression of genes associated with the cytoskeleton, focal adhesions and the extracellular matrix. Although the nucleus is mechanically connected to the cytoskeleton through the LInker of Nucleoskeleton and Cytoskeleton (LINC) complex, the role of this group of proteins in these responses to microgravity has yet to be defined. Therefore, we used simulated microgravity achieved by growing cells on a 3d clinostat to investigate whether the LINC complex acts to mediate responses to the microgravity environment. We show that nuclear shape and differential gene expression are both responsive to simulated microgravity in a LINC-dependent manner and that this response changes with the duration of exposure to simulated microgravity. These LINC-dependent genes likely represent elements normally regulated by the mechanical forces imposed by gravity on Earth.
