ABSTRACT: To investigate whether the gene expression profiles of synovial fluid mesenchymal stem cells (SF-MSCs) from rat OA knees were favorable for endogenous cartilage repair, we performed RNA sequencing for SF-MSCs from intact knees and knees 4 weeks after the pMx and sham surgery.

Synovial fluid proteome changes in ACL injury-induced posttraumatic osteoarthritis: proteomics analysis of porcine knee synovial fluid

Project description:In the current work, we used an isotopically labeled high resolution LC MS/MS-based proteomics approach to analyze the protein profile of synovial fluid at 6 and 12 months after ACL transection in untreated and repaired porcine knees. Our primary aim was to determine how the synovial fluid proteome differs between the two groups in an effort to identify candidate proteins that may be associated with the development of posttraumatic OA. We hypothesized that the development of macroscopic cartilage damage following surgical ACL transection would be accompanied by differential changes in synovial fluid proteome in untreated knees compared to repaired joints.

Multiomics reveals diverse effects of silk fibroin and sericin on mesenchymal stem cells: implications for tissue regeneration

Project description:In the present study, we employed multiomics analysis to investigate the biological interactions of SF and SS with mesenchymal stem cells (MSCs) to elucidate the triggered cellular responses and pathways. The integrated transcriptomics and proteomics revealed multiple biological processes of MSCs with widespread gene and protein changes initiated by SF and SS. Specifically, both SF and SS strongly potentiated the paracrine activities of MSCs via activating multiple but distinct signaling pathways instead of directly regulating cell behaviors. Additionally, we investigated whether the paracrine functions of MSCs in response to SF and SS could mediate the local signaling processes in vivo and have implications for tissue regeneration. We found that SF and SS triggered specific paracrine signals of MSCs that promote cell proliferation, migration, angiogenesis, immunomodulation, and tissue healing processes. Moreover, the high consistency of transcriptomic and proteomic analyses in vitro and in vivo indicated the strong cellular responses of MSCs to SF and SS and the stable and powerful biological effects of these paracrine signals. This work thus provides a comprehensive and reliable insight into the cellular interactions with SF and SS via multiomics profiling, which enables the further development of silk-based therapeutics for tissue engineering and regenerative medicine

MicroRNA-29a Counteracts Synovitis in Knee Osteoarthritis Pathogenesis by Targeting VEGF.

Project description:Synovitis contributes to the development of osteoarthritis (OA) of the knee. MicroRNAs regulate joint microenvironment homeostasis and deterioration. This study was undertaken to characterize the actions of microRNA-29a (miR-29a) to synovial remodeling in OA joints. Synovial specimens isolated from patients with end-stage OA knees showed abundant fibrotic matrix and vessel histopathology concomitant with weak miR-29a expression. In vitro, miR-29a knockdown caused synovial fibroblasts to exhibit high expressions of collagen III, TGF-?1, MMP9, MMP13, and ADAMTS5, whereas miR-29a overexpression diminished these joint-deleterious factors. In collagenase-mediated OA pathogenesis, miR-29a-overexpressing transgenic mice showed minor responses to hyperplasia, macrophage infiltration, fibrosis, hyperangiogenesis, and VEGF expression in synovial lesions. These effects mitigated articular cartilage loss and gait aberrance of injured joints. Intra-articular administration of miR-29a precursor lessened the collagenase aggravation of excessive synovial remodeling reactions and thereby sustained joint tissue integrity. miR-29a lowered VEGF production and angiogenic activities in synovial fibroblasts through targeting the 3'-UTR of VEGF. Taken together, miR-29a deficiency exacerbated synovitis pathogenesis in the end-stage OA knees. miR-29a signaling fends off excessive synovial angiogenesis and fibrosis, which delays joint destruction. This study sheds new light on the protective effects against synovial deterioration and the therapeutic advantage of miR-29a in OA knees.

Can optical spectral transmission assess ultrasound-detected synovitis in hand osteoarthritis?

Project description:ObjectiveTo determine whether optical spectral transmission (OST) can be used to assess synovitis in hand and wrist joints of patients with hand osteoarthritis (OA).DesignHand and wrist joints of 47 primary hand OA patients with at least one clinically inflamed hand or wrist joint were assessed for synovitis by OST and ultrasound (US). Associations between standardized OST and US synovitis were studied in linear mixed effects models, across all joint types together and individually for wrist, proximal interphalangeal (PIP), and distal interphalangeal (DIP) joints, and were adjusted for OA features that showed associations with US synovitis. Diagnostic performance was determined using receiver operator characteristic (ROC) curves analysis, with US as reference standard.ResultsAltogether, 6.7% of joints showed US synovitis. Statistically significant associations between OST scores and US synovitis were found for all joints combined (Δ0.37SD, p<0.001) and PIP joints (Δ0.81SD, p<0.001), but not for DIP (Δ0.14SD, p = 0.484) or wrist joints (Δ0.37SD, p = 0.178). All associations were independent of other OA features, i.e. osteophytes and dorsal vascularity. Analysis of diagnostic performance of OST, revealed an area under the ROC curve (AUC-ROC) of 0.74 for all joints together (p<0.001), 0.69 for PIP joints (p<0.001), 0.54 for DIP joints (p = 0.486), and 0.61 for wrist joints (p = 0.234).ConclusionsOST scores and US synovitis are statistically significantly associated, independent of osteophytes and dorsal vascularity. At this stage, OST performs fair in the assessment of synovitis in PIP joints of hand OA patients.
