DSF effects on Arabidopsis

ABSTRACT: To investigate the effects of DSF on Arabidopsis in Xcc infecction, we used 2 μM DSF pretreated the roos of Arabidopsis for 48h and then inoculated with Xcc, We then performed gene expression profiling analysis using data obtained from RNA-seq of pretreated/unpretreated at two time points (inoculated with Xcc or not)

Gene expression profiling of early events of DSF elicitor based defense signalling in Arabidopsis thaliana

Project description:To study the early events of priming response leading to changes in gene expression in Arabidopsis thaliana Col0 wild type plant on infiltration with the potent elicitor molecule Diffusible Signal Factor (DSF). In the present experimentation we have employed total RNA microarray expression profiling as a discovery platform to identify differentially expressing genes that are upregulated and down regulated in the various pathways involved in priming the defense responses by the elicitor molecule DSF on infiltration in the host plant Arabidopsis thaliana. This would help understand the possible defense pathways elicited by the elicitor molecule during interaction. The rosette leaves of Arabidopsis thaliana Col0 plants of 4 weeks old were infiltrated with 100uM DSF and the total RNA samples at 4hpi, 8hpi and 16hpi were extracted along with methanol treatment serving as a control in two biological replicates 1A and 2A. Further, these RNA samples were checked for the quality and subjected to microarray analysis. The differentially expressing candidate genes were identified specific to DSF treatment and were quantified by real-time PCR post DSF treatments.

The transcriptome pattern of inoculated and non-inoculated plants of Arabidopsis was compared at three time points after inoculation (24 hours, 48 hours and one-week ai). Sixty plants were studied by comparison (inoculated-non inoculated) and by kinetic point. RNAs of the 30 inoculated plants and the RNAs of the 30 non-inoculated plants were pooled to eliminate variability between individual plants and thus to target for genes for which differences in expression were linked to the inoculation by the pathogen. Five comparisons have been performed: inoculated-non inoculated at 24 hours ai; inoculated-non inoculated 48 hours ai; inoculated-non inoculated one-week ai; inoculated 24h ai-inoculated 48h ai; inoculated 48h ai- inoculated one-week ai. The entire experiment was repeated (biological repetition) and each comparison was performed in two independent hybridisations (technical repetition). Quantitative proteomics analysis reveals novel insights into mechanisms of action of Disulfiram (DSF)

Project description:The new uses of the old drugs may reduce cost and shortens the production cycle of research and development, especially that approved by FDA for important clinical conditions. Alcohol abuse Disulfiram (DSF) has been proven safe and shows the promising anti-tumor effect in may preclinical studies. However, the potential side effects of DSF on tumor remain unknown. In this study, we explored the role of DSF in cancer cells and searched for the differential expressed protein after DSF treatment in the HeLa cells. To fully understand the mechanism of action of DSF with a systems perspective, we employed a quantitative proteomics strategy to systematically identify potential targets of DSF. In total, 201 proteins were dys-regulated significantly after DSF exposure, implying that they may be potential targets of DSF. Analysis of this data on a system level revealed major changes of proteins involved in diverse biological processes, including metabolic process and response to stimulus.

The pathogen infects and multiplies in plant vascular tissues and, as the disease progresses, the veins of infected tissues turn black and characteristic V-shaped lesions appear along the margins of leaves.The aim of this work is to identify differentially expressed genes from Brassica oleracea during early infection by Xcc, in an attempt to identify proteins related to resistance. Cabbge seedlings were inoculated with Xanthomonas campestris pv campestris (Xcc) suspension and cabbage gene expression at 6h., 24h. And 48h. After inoculation was assessed with help of Brassica 95k EST microarray chip.

Application of Functional Genomics to the Study of Solanacea's Defense Mechanisms against Xanthomonas sp. Pathogenicity

Project description:Xanthomonas campestris pv. campestris (Xcc) is a major bacterial pathogen of cruciferous plants worldwide. The pathogen produces polysaccharides including extracellular cyclic glucan, xanthan, and extracellular enzymes that are key virulence factors. Different Xcc mutants (8397-defective in xanthan and 8523- defective in extracellular glucans) have been obtained and characterized in previously, wich shown to be less infective than the wild type strain when inoculated in N.benthamiana, wich has shown to be an excellent model for the study of Xcc-plant interaction. The objective of this work is evaluate this compounds functions in the plant-pathogen interaction, in particular in the plant transcriptoma modulation to confer susceptibility or resistance to the infection. Plant gene expression profiles would be obtained from independently inoculated leaves of N.benthamiana with the following strains of xanthomonas: Xcc. 8004 (wild type), Xcc. 8397 (xanthan minus), Xcc. 8523 (glucan minus), ant water (control). Leaves discs will be collected at 24 hs post infection and immediately submerged in liquid N2. Total RNA will be extracted with plant RNA specific kits (RNA easy QIAGen), treated with DNase, purified and quantified. Keywords: Reference design
