Gene expression alterations with Nkapl knockout or NKAPL frameshift mutation (M349fs)

ABSTRACT: This study aims to explore the transcriptomic changes in Nkapl-KO, Sox30-KO and Nkapl TGdel/TGdel mouse testes comparing with their wild-type testes. Fresh testes were collected from 21d wild-type, Nkapl-KO, Sox30-KO and Nkapl TGdel/TGdel mice, respectively.

Transcription profiling of mouse WT and Nxf2 KO post-natal day 21 testes

Project description:In euakryotes, mRNAs must be exported from the nucleus to the cytsoplasm. NXF2 is highly expressed in the mouse male germ cells. We are interested in its function in spermatogenesis, espically in the nuclear RNA export in the testis. To this end, we made Nxf2 mutant mice by gene targeting. In an attempt to identify the mRNA substrates of NXF2, we perform the microarray experiments on testes. We used microarrays to check the expression profiles of the Nxf2 WT and KO 21d testes on C57BL/6 background. Experiment Overall Design: To examine the expression difference between WT and Nxf2 KO testes, we collected testes from juvenile mice of three ages ( 21d, 26d, 28d). Testis weight was similar between WT and KO mice at post-natal day 21. Three pairs of WT and KO 21d testes were chosen for microarray analysis.

Project description:We challenge bristol strain (wild-type), metl-9 KO strain (short as KO, has a 101bp insertion, leads to a truncated protein of 258aa) and metl-9 catalytic-activity mutated strain (short as mut, has N172K, D274G mutations in full-length protein) with P.aeruginosa (P.A14), and observe a discrepant transcriptome pattern between wild-type and KO/mut strains. Plenty of innate immune response genes show different expression patterns upon P.A14 infection between the wild-type strain and KO/mut strain. It indicates the important role of metl-9 and 6mA in worm innate immune response modulation.
