Targeting aberrant sialylation and fucosylation in prostate cancer cells using potent metabolic inhibitors

ABSTRACT: To identify signalling pathways altered by treating prostate cancer cells with the metabolic inhibitors P-SiaFNEtoc, Fucotrim I and Fucotrim II, we treated CWR22Rv1 cells with P-SiaFNEtoc, Fucotrim I and Fucotrim II for 72 hours and their respective vehicle control with DMSO. We then performed differential gene expression, Gene Ontology and GSEA analyses using data obtained from RNA-seq of the control and treated cell lines.

Characterization of co-transcriptional splicing in acute lymphoblastic leukemia

Project description:Alternative splicing is a promising therapeutic target in acute lymphoblastic leukemia. Here, we report the alternative splicing changes in B-ALL cell lines including NALM6 cells and MUTZ-5 cells treated with DYRK1 inhibitor, EHT 1610 and GNF 2133. Dysregulation of co-transcriptional splicing is a therapeutic target in acute lymphoblastic leukemia (ALL). Here, we detected the alternative splicing in NALM6 cells or MUTZ-5 cells treated with EHT 1610 or GNF2133. We detected the alternative splicing in NALM6 cells cells treated with THAL-SNS-032. We also determined the alternative splicing in MEF2D-BCL9 ALL and MEF2D-HNRNPUL1 ALL patient xenografted samples treated with E7820, a anticancer sulfamide, or DMSO. Furthermore, ChIP-seq for RNA polymerase II (Pol II), p-Ser2 Pol II, and p-Ser5 Pol II were performed after DMSO or EHT 1610 treatment in NALM6 cells. ChIP-seq for p-Ser2 Pol II, and p-Ser5 Pol II were performed after DMSO or THAL-SNS-032 treatment in NALM6 cells.PRO-seq in NALM6 cells treated with DMSO, EHT 1610, and THAL-SNS-032 were performed.

Tissue selective effects of topoisomerase II inhibitors in vivo

Project description:One major class of anti-cancer drugs targets topoisomerase II to induce DNA double-strand breaks and cell death of fast growing cells. In vitro experiments showed that doxorubicin can induce histone eviction as well as DNA damage, while etoposide can only induce DNA damage. Here, we compare the transcription responses of different tissues to doxorubicin or etoposide treatment in vivo. Total RNA from respective tissues in FVB mice 1 day or 6 days post indicated drug treatment were extracted and compared to un-treated mice. Two mice were used for each treatment.

Project description:We identified the first Spt5-Pol II inhibitors (SPIs). SPIs faithfully reproduced Spt5 knockdown effects on proximal-promoter-pausing, NF-κB activation and the expanded-repeat huntingtin gene in neuronal cells. Using SPIs we identified Spt5 target genes that responded with profoundly diverse kinetics and a novel regulatory element of proximal-promoter-pausing. To obtain a genome-wide view of the impact of SPIs on transcription, we performed Global run-on sequencing (GRO-seq) using nuclei of HeLa cells treated with either DMSO (control) or SPI-21.

Inflammatory Signaling is Regulated by Fucosylation in Metastatic Breast Cancer Cells

Project description:N-linked glycosylation results in a large branched tree-like sugar structure composed of several types of individual carbohydrates being attached to a protein. Fucosylation is one specific type of glycosylation that is defined by the addition of α-L-fucose to a carrier protein. Although fucosylation has been relatively well defined as a biomarker for progression in some human cancers, for example, pancreatic and hepatocellular carcinoma, its role in breast cancer is much less well defined. A growing body of evidence indicates that levels of fucosylation correlate with breast cancer progression and contributes to metastatic disease. However, very little is known about the signaling and functional outcomes that are driven by fucosylation. We performed tandem- mass-tag (TMT) proteomics on 4T1 metastatic mammary tumor cells that were treated with a fucosylation inhibitor, 2-fluorofucose (2FF) or vehicle (DMSO) control. This analysis identified >400 proteins that were significantly increased or decreased in 2FF treated samples compared to the DMSO treated samples. We found that two proteins, Tollip and Bcl-10, were downregulated in 4T1 cells in response to 2FF treatment. Functionally, Tollip and Bcl10 signaling induce pro-inflammatory responses through activation of NF-κB, a transcription factor that is pro-tumorigenic and a prime target in human cancer. Our results show that treatment of 4T1 cells with 2FF leads to a reduction in NF-κB expression and activity through increased IκBα. This observation is consistent with a reduction in inflammatory response due to the loss of Tollip and Bcl-10 expression. Collectively, our results indicate that fucosylation is a key mediator of inflammatory signaling in metastatic breast cancer cells.
