Project description:In order to evaluate the identification of genes and pathways, the global gene expression profiles were assessed in response to GO and rGO on Human hepatoma (HepG2) cells. We performed whole genome DNA microarray experiments using HepG2 cells exposed to GO and rGO for 24h. We used whole genome microarrays to screen for global changed in HepG2 transcription profiles and with subsequent quantitative analysis conducted on selected genes. 24h GO and rGO exposed HepG2 cells were used for total RNA extraction and hybridization on Affymetrix microarrays.

Project description:Antibacterial surfaces have an enormous economic and social impact on the worldwide technological fight against diseases. However, bacteria develop resistance and coatings are often not uniform and not stable in time. The challenge is finding an antibacterial coating that is biocompatible, cost-effective, not toxic, and spreadable over large and irregular surfaces. Here we demonstrate an antibacterial cloak by laser printing of graphene oxide hydrogels mimicking the Cancer Pagurus carapace. We observe up to 90% reduction of bacteria cells. This cloak exploits natural surface patterns evolved to resist to microorganisms infection, and the antimicrobial efficacy of graphene oxide. Cell integrity analysis by scanning electron microscopy and nucleic acids release show bacteriostatic and bactericidal effect. Nucleic acids release demonstrates microorganism cutting, and microscopy reveals cells wrapped by the laser treated gel. A theoretical active matter model confirms our findings. The employment of biomimetic graphene oxide gels opens unique possibilities to decrease infections in biomedical applications and chirurgical equipment; our antibiotic-free approach, based on the geometric reduction of microbial adhesion and the mechanical action of Graphene Oxide sheets, is potentially not affected by bacterial resistance.

Project description:The application of membrane technology to remove pollutant dyes in industrial wastewater is a significant development today. The modification of membranes to improve their properties has been shown to improve the permeation flux and removal efficiency of the membrane. Therefore, in this work, graphene oxide nanoparticles (GO-NPs) were used to modify the polyethersulfone (PES) membrane and prepare mixed matrix membranes (MMMs). This research is dedicated to using two types of very toxic dyes (Acid Black and Rose Bengal) to study the effect of GO on PES performance. The performance and antifouling properties of the new modified membrane were studied using the following: FTIR, SEM, AFM, water permeation flux, dye removal and fouling, and by investigating the influence of GO-NPs on the structure. After adding 0.5 wt% of GO, the contact angle was the lowest (39.21°) and the permeable flux of the membrane was the highest. The performance of the ultrafiltration (UF) membrane displayed a rejection rate higher than 99% for both dyes. The membranes showed the highest antifouling property at a GO concentration of 0.5 wt%. The long-term operation of the membrane fabricated from 0.5 wt% GO using two dyes improved greatly over 26 d from 14 d for the control membrane, therefore higher flux can be preserved.

Project description:Regulatory T cell (Treg)-based therapeutics are receiving increased attention for their potential to treat autoimmune disease and prevent transplant rejection. Adoptively transferred Tregs have shown promise in early clinical trials, but cell-based therapies are expensive and complex to implement, and "off-the-shelf" alternatives are needed. Here, we investigate the potential of artificial antigen presenting cells (aAPCs) fabricated from a blend of negatively charged biodegradable polymer (poly(lactic-co-glycolic acid), PLGA) and cationic biodegradable polymer (poly(beta-amino ester), PBAE) with incorporation of extracellular protein signals 1 and 2 and a soluble released signal 3 to convert naïve T cells to induced Foxp3+ Treg-like suppressor cells (iTregs) both in vitro and in vivo in a biomimetic manner. The addition of PBAE to the aAPC core increased the conjugation efficiency of signal proteins to the particle surface and resulted in enhanced ability to bind to naïve T cells and induce iTregs with potent suppressive function. Furthermore, PLGA/PBAE tolerogenic aAPCs (TolAPCs) supported the loading and sustained release of signal 3 cytokine TGF-β. A single dose of TolAPCs administered intravenously to C57BL/6 J mice resulted in an increased percentage of Foxp3+ cells in the lymph nodes. Thus, PLGA/PBAE TolAPCs show potential as an "off-the-shelf" biomimetic material for tolerance induction. STATEMENT OF SIGNIFICANCE: Regulatory T cells (Tregs) are promising for basic research and translational medicine as they can induce tolerance and have the potential to treat autoimmune diseases such as type 1 diabetes and multiple sclerosis. As cell-based therapies are expensive and difficult to manufacture and implement, non-cellular methods of engineering endogenous Tregs are needed. The research reported here describes a new type of biomimetic particle, tolerogenic artificial antigen presenting cells (TolAPCs) fabricated from a blend of negatively charged biodegradable polymer, poly(lactic-co-glycolic acid), and positively charged biodegradable polymer, poly(beta-amino ester), along with key biomolecular signals: extracellularly presented protein signals 1 and 2 and a soluble released signal 3. These TolAPCs bind to naïve T cells and induce Foxp3+ Treg-like suppressor cells with potent suppressive function. In both in vitro and in vivo studies, it is shown that this non-cellular approach is useful to induce tolerance.

Project description:The mercury ion (Hg2+) is one of the heavy metal ions, and its presence in trace amounts can cause physiological damage to an organism. Traditional methods of Hg2+ detection have been useful but have also had numerous limitations and challenges, and as a result, it is important to design new and sophisticated methods that can aid in the detection of Hg2+. In this paper, two fluorescent dyes, carboxyfluorescein (FAM) and SYBR Green I, were used to label and intercalate DNA probes immobilized on the surface of graphene oxide (GO) for sensors to detect Hg2+. FAM and SYBR Green I dye share close excitation and emission wavelength spectra, which can promote and amplify the detection of signals, and also increase the limit of detection (LOD). The results showed that the limit of detection in this method was 0.53 nM. Moreover, when the sensors with double amino groups on the surface of GO were carried out to detect Hg2+, a limit of detection was improved to 0.43 nM. The sensors were then applied in the real sample. The results show that this method has a promising potential in Hg2+ detection.

Project description:Graphene oxide (GO)-phosphor hybrid nanoscrolls were synthesized using a simple chemical method. The GO-phosphor ratio was varied to find the optimum ratio for enhanced optical characteristics of the hybrid. A scanning electron microscope analysis revealed that synthesized GO scrolls achieved a length of over 20 μm with interior cavities. The GO-phosphor hybrid is extensively analyzed using Raman spectroscopy, suggesting that various Raman combination modes are activated with the appearance of a low-frequency radial breathing-like mode (RBLM) of the type observed in carbon nanotubes. All of the synthesized GO-phosphor hybrids exhibit an intense luminescent emission around 540 nm along with a broad emission at approximately 400 nm, with the intensity ratio varying with the GO-phosphor ratio. The photoluminescence emissions were gauged using Commission Internationale d'Eclairage (CIE) coordinates and at an optimum ratio. The coordinates shift to the white region of the color spectra. Our study suggests that the GO-phosphor hybrid nanoscrolls are suitable candidates for light-emitting applications.

Project description:Nanocomposites from native cellulose with low 2D nanoplatelet content are of interest as sustainable materials combining functional and structural performance. Cellulose nanofibril-graphene oxide (CNF-GO) nanocomposite films are prepared by a physical mixing-drying method, with focus on low GO content, the use of very large GO platelets (2-45μm) and nanostructural characterization using synchrotron x-ray source for WAXS and SAXS. These nanocomposites can be used as transparent coatings, strong films or membranes, as gas barriers or in laminated form. CNF nanofibrils with random in-plane orientation, form a continuous non-porous matrix with GO platelets oriented in-plane. GO reinforcement mechanisms in CNF are investigated, and relationships between nanostructure and suspension rheology, mechanical properties, optical transmittance and oxygen barrier properties are investigated as a function of GO content. A much higher modulus reinforcement efficency is observed than in previous polymer-GO studies. The absolute values for modulus and ultimate strength are as high as 17 GPa and 250 MPa at a GO content as small as 0.07 vol%. The remarkable reinforcement efficiency is due to improved organization of the CNF matrix; and this GO-induced mechanism is of general interest for nanostructural tailoring of CNF-2D nanoplatelet composites.
