Human Skeletal Stem Cell Diversity

ABSTRACT: We have used SmartSeq2 to sequence single phenotypic human skeletal stem cell (SSCs) and downstream lineage populations purified via FACS from fetal skeletal tissues and patient specimens. SSC populations were isolated based on their previously reported surface marker profiles. A human SSC (CD45-CD235a-CD31-TIE2-CD146-PDPN+CD73+CD164+), a human bone-cartilage-stroma-progenitor (BCSP; CD45-CD235a-CD31-TIE2-CD146+PDPN+) and an osteoprogenitor (OP; CD45-CD235a-CD31-TIE2-CD146+PDPN-) were investigated.

Comparison of periosteal, adipose, and dermal CD146+ human pericytes

Project description:Human pericytes demonstrate multilineage differentiation potential, and their descendants participate in tissue homeostasis and repair. Increasing evidence from developmental biology and tissue engineering suggest that regional specification by tissue of origin exists among human pericytes. Here, we sought to define the differentiation of CD146+ human pericytes from skeletal and soft tissue sources. Uncultured CD146+CD31-CD45- pericytes were derived by fluorescent activated cell sorting from human periosteum, adipose, or dermal tissue. Periosteal CD146+CD31-CD45- cells retained canonical features of pericytes, including cell surface marker expression, multilineage differentiation potential, and paracrine induced tubulogenesis. Periosteal pericytes demonstrated a striking tendency to undergo osteoblastogenesis, while soft tissue pericytes did not in vitro or in vivo. Microarray analysis demonstrated substantive differences between periosteal pericytes in comparison to their soft tissue pericyte counterparts. In sum, skeletal and soft tissue pericytes differ in their relative lineage differentiation potential and ability to form bone. Transcriptome analysis of synovial osteoprogenitor populations in mice with antigen-induced arthritis and non-immunized mice

Project description:The aim of this experiment was to analyze differences in gene expression between potential osteoprogenitor populations in synovial compartment of non-immunized mice and mice with antigen-induced arthritis (AIA). We analyzed transcriptome of TER119–CD31–CD45–CD51+CD200+CD105– cells from nonimmunized mice, TER119–CD31–CD45–CD51+CD200+CD105– cells from mice with arthritis, and TER119–CD31–CD45–CD51+CD200–CD105+ from mice with arthritis. Comparison between nonimmunized and arthritic mice served to determine changes in TER119–CD31–CD45–CD51+CD200+CD105–population induced by arthritis. Comparison of TER119–CD31–CD45–CD51+CD200+CD105– and TER119–CD31–CD45–CD51+CD200–CD105+ from mice with arthritis aimed to define transcriptomic differences between those two populations.

Signaling pathway manipulation of CXCR4 among soft tissue pericytes led to an increase in osteoblastogenesis and bone formation. In sum, skeletal and soft tissue pericytes differ in their relative lineage differentiation potential and ability to form bone. Plasticity exists, however, and manipulation of CXCR4 signaling pathway may ‘coax’ a soft tissue pericyte toward an osteoblastogenic cell fate.

Vessel Forming Mesenchymal Stromal Cell Populations

Population 1 (P1) is CD45-Ter119-Tie2+PDGFRa-CD31+CD105highSca1low and gives rise to stunted vessels (incomplete tubular structures) in a transplant setting, and Population 2 (P2) which is CD45-Ter119-Tie2+PDGFRa+CD31-CD105lowSca1high and forms stunted vessels and fat. To test the homogeneity and signal interactions of these populations we performed 10X single cell RNA-sequencing on them.
