Project description:Adventitious roots (AR) develop from tissues other than the primary root, in a developmental process physiologically regulated by phytohormones. Adventitious roots provide structural support and contribute to water and nutrient absorption, and are critical for commercial vegetative propagation of several crops. Here we quantified the number of AR, root architectural traits and root biomass in cuttings from a pseudo-backcross population of Populus deltoides and Populus trichocarpa. Project description:Strigolactone (SL) signalling plays many roles in plants, however, the downstream responsive genes remain unclear. SUPPRESSOR OF MORE AXILLARY GROWTH2-LIKE 7 (SMXL7) is degraded upon SL signalling. Here, we generated Arabidopsis carrying a transgene SMXL7pro:SMXL7d53-GR, where a SL-insensitive version of SMXL7 is tagged with the ligand binding domain of the glucocorticoid receptor (GR). SMXL7d53-GR will be translocated to the nucleus upon dexamethasone (dex), and we profiled the gene expression in mock or dex treated plants and revealed the downstream responsive genes of SL signalling.

Project description:To reveal transcriptome dynamics during adventitious root formation in a coniferous tree, C. japonica, we conducted custom microarry experiments. Three parts from cuttings of easy-to-root clone of C. japonica were collected at eight time points during adventitious root formation. The results revealed major turning points on transcriptome toward adventitious root formation and the expression behavior of genes related to carbohydrate, plant hormone and others suggested the important biological changes for adventitious root formation.

Project description:Strigolactones are plant metabolites that act as phytohormones and rhizosphere signals. Whereas most research on unraveling the action mechanisms of strigolactones is focused on plant shoots, we investigated proteome adaptation during strigolactone signaling in the roots of Arabidopsis thaliana. Through large-scale, time-resolved, and quantitative proteomics, the impact of the strigolactone analog rac-GR24 was elucidated on the root proteome of the wild type and the signaling mutant more axillary growth 2 (max2). Our study revealed a clear MAX2-dependent rac-GR24 response: an increase in abundance of enzymes involved in flavonol biosynthesis, which was reduced in the max2-1 mutant. Mass spectrometry-driven metabolite profiling and thin-layer chromatography experiments demonstrated that these changes in protein expression lead to the accumulation of specific flavonols. Moreover, quantitative RT-PCR revealed that the flavonol-related protein expression profile was caused by rac-GR24--induced changes in transcript levels of the corresponding genes. This induction of flavonol production was shown to be activated by the two pure enantiomers that together make up rac-GR24. Finally, our data provide much needed clues concerning the multiple roles played by MAX2 in the roots and a comprehensive view of the rac-GR24--induced response in the root proteome.

Project description:Arctic alpine species experience extended periods of cold and unpredictable conditions during flowering. Thus, often, alpine plants use both sexual and asexual means of reproduction to maximise fitness and ensure reproductive success. We used the arctic alpine perennial Arabis alpina to explore the role of prolonged cold exposure on adventitious rooting. We exposed plants to 4 °C for different durations and scored the presence of adventitious roots on the main stem and axillary branches. Our physiological studies demonstrated the presence of adventitious roots after 21 weeks at 4 °C saturating the effect of cold on this process. Notably, adventitious roots on the main stem developingin specific internodes allowed us to identify the gene regulatory network involved in the formation of adventitious roots in cold using transcriptomics. These data and histological studies indicated that adventitious roots in A. alpina stems initiate during cold exposure and emerge after plants experience growth promoting conditions. While the initiation of adventitious root was not associated with changes of DR5 auxin response and free endogenous auxin level in the stems, the emergence of the adventitious root primordia was. Using the transcriptomic data, we discerned the sequential hormone responses occurring in various stages of adventitious root formation and identified supplementary pathways putatively involved in adventitious root emergence, such as glucosinolate metabolism. Together, our results highlight the role of low temperature during clonal growth in alpine plants and provide insights on the molecular mechanisms involved at distinct stages of adventitious rooting.

Project description:Strigolactones (SLs), a class of phytohormones, play a pivotal role in governing critical aspects of plant development, including shoot branching and root development. DWARF53 (D53) is a key repressor in the SL signaling. Nevertheless, its involvement and regulatory mechanisms in cotton fiber development—a representative single-cell model, has remained elusive. In this study, we unveil GhD53 represses the transcription of SL-inducible genes GhFAD3-2 and GhFAD3-4, which are involved in linolenic acid biosynthesis—a stimulant of cell elongation, thus inhibiting cotton fiber elongation. We demonstrate that GhD53's inhibitory influence on cotton fiber elongation is relieved in the presence of SLs, as GhD53 undergoes degradation. This alleviation of GhD53 repression triggers the activation of GhFAD3 expression, promoting fiber cell elongation. Remarkably, GhD53 serves as a dual-function transcription repressor, impeding SLs’ role in fiber elongation. Firstly, as a transcription factor, GhD53 directly binds to GhFAD3 promoters, suppressing their transcription. Secondly, GhD53 physically interacts with GhSLRF, a bHLH-type transcription factor, preventing it from binding to GhFAD3 promoters and repressing its transcription activation activity. GhD53 emerges as a pivotal component in the regulatory network governing SL signaling transduction. Overall, our findings provide novel insights into D53’s role in SL signaling and delineate a comprehensive framework for transmitting SL signals to modulate plant cell elongation behavior.

Project description:affy_strigolactone_sunflower - affy_strigolactone_sunflower - Abiotic stress and more specifically drought is the major limiting factor for sunflower production. Sunflower response to drought includes root plasticity to adapt to water availability and reach soil water. We identified genotype-specific responses of root architecture to strigolactone application. This experiment aims at identifying strigolactone responsive pathways in 6 genotypes in order to better understand molecular control of root development in sunflower and therefore its response to drought. The ultimate goal will be to improve sunflower breeding through selection of key drought response genes.-The experiment consisted of 2 repeats of four 12-day-old-plantlets of 6 sunflower genotype SF193 (INRA code: XRQ), SF326 (INRA code: PSC8), SF056 (INRA code: FU), SF306 (INRA code: PAZ2), SF302 (INRA code: PAC2), SF268 (INRA code: RHA266), grown in growth chamber conditions and submitted to a 24-hour-treatment of 100 nM strigolactone analogue rac-GR24 (Chiralix, Nijmegen, Netherland) or not. Growth conditions were 14h light at 23°C and 10h night at 20°C under fluorescent bulbs. Plants were grown in hydroponic boxes containing 20 litres of aerated liquid culture medium (as described in Massonneau et al., 2001 Planta).
