Project description:Myometrial stem/progenitor cells (MyoSPCs) have been proposed as the cells of origin for uterine fibroids, which are benign tumors that develop in the myometrium of most reproductive age women, but the identity of the MyoSPC has not been well established. We previously identified SUSD2 as a possible MyoSPC marker, but the relatively poor enrichment in stem cell characteristics of SUSD2+ over SUSD2- cells compelled us to find better discerning markers for more rigorous downstream analyses. We combined bulk RNA-seq of SUSD2+/- cells with single cell RNA-seq to identify markers capable of further enriching for MyoSPCs. We observed seven distinct cell clusters within the myometrium, with the vascular myocyte cluster most highly enriched for MyoSPC characteristics and markers, including SUSD2. CRIP1 expression was found highly upregulated in both techniques and was used as a marker to sort CRIP1+/PECAM1- cells that were both enriched for colony forming potential and able to differentiate into mesenchymal lineages, suggesting that CRIP1+/PECAM1- cells could be used to better study the etiology of uterine fibroids.

Project description:We previously identified Dclk1, a tuft cell marker, marks tumor stem cells (TSCs) in mouse intestinal tumors. In this study, we have identified IL17RB as a cell surface marker distinctively expressed by Dclk1+ tuft-like tumor cells in mouse intestinal tumors. Using this tuft cell marker, we compared and analyzed the transcriptome of Lgr5-tuft marker-, Lgr5+tuft marker-, Lgr5-tuft marker+, and Lgr5+tuft marker+ tumor cells. These analyses revealed that tuft-like tumor cells in the intestinal tumors comprise two distinct subsets: highly differentiated tuft-like tumor cells (Lgr5-tuft marker+ cells) and tuft-like tumor cells with TCS potential (Lgr5+tuft marker+ cells).

Project description:Background: Identification of a lineage-specific marker plays a pivotal role in understanding developmental process and is necessary to isolate a certain cell type with high purity for therapeutic purposes. Here, we report a new cardiac-specific marker and demonstrate its functional significance in cardiac development. Methods: When mouse pluripotent stem cells (PSCs) were stimulated with BMP4, Activin A, and bFGF, they differentiated into cardiac cells. To screen molecules expressed on cardiac progenitor cell (CPC) surfaces compared to those on undifferentiated PSCs, we isolated Flk1+/PdgfRa+ cells at differentiation day 4 and performed microarray analysis. Results: Among candidates, we identified a new G protein-coupled receptor, latrophilin-2 (Lphn2). Here, we report this new cardiac-specific surface marker, Lphn2, expressed specifically on CPCs and cardiomyocytes (CMCs) during mouse and human PSC differentiation in vitro and exclusively in the heart during mouse embryonic development. Lphn2 knockout in mice was embryonically lethal because of severe heart, but not vascular, defects. PSC-derived Lphn2+ cells, but not Lphn2- cells, differentiated into CMCs and regenerated the myocardium when transplanted into infarcted hearts. Transplanted Lphn2+ cells improved left ventricular systolic function and reduced infarct sizes. Analysis of the signaling pathway indicated that cyclin-dependent kinase 5 is downstream of Lphn2 and collaborates with Src kinase to induce P38MAP kinase phosphorylation, subsequently activating cardiac-related gene transcription. Conclusion: Lphn2 is a functionally significant cell-surface marker for cardiac progenitor and cardiomyocytes. These findings provide a valuable tool for isolating cardiomyogenic progenitors and CMCs from PSCs and shed light on heart development and regeneration.

Project description:The generation and purification of cardiomyocyte sub-population are critical for precise modelling of cardiac diseases, drug screening, and regenerative medicine due to their distinct phenotypic and functional characteristics. Therefore, identifying surface markers for cardiomyocyte subtypes that facilitate live cell sorting of homogenous population of cardiomyocytes is of utmost importance for successful translation of cardiovascular research and therapeutic applications. Using MARIS method coupled with RNA sequencing analysis on MLC2A- and MLC2V-expressing cardiomyocytes, we identified JAK2 as a potential cell surface marker for purifying ventricular cardiomyocytes. We demonstrated that that SIRPA+/JAK2+ population sorted expressed high levels of ventricular-specific marker and had electrophysiological properties that corresponded to ventricular-like cardiomyocytes. Functionally, SIRPA+/JAK2+ cardiomyocytes do not response to vernakalant, an atrial-selective anti-arrhythmic agent, confirming their identity as ventricular-like cells.

Project description:The human uterus undergoes profound physiological tissue remodelling during pregnancy. In the myometrium, altered gene expression must underlie these extensive molecular and structural changes. The purpose of this study was to compare expression profiles of pregnant and non-pregnant myometrium, in order to identify genes that participate in this process. mRNA from 14 non-pregnant and four pregnant human myometrial samples were analysed using a human UniGEM V microarray with 7075 cDNA elements. A total of 602 transcripts from the microarray were up-regulated >=2.0-fold in pregnant myometrium, with 37 transcripts up-regulated >=4.0-fold. In contrast, eight transcripts were down-regulated >=2.0-fold in pregnancy. To ensure accurate representation of differential gene expression, Northern blot analyses using total RNA from 16 samples of non-pregnant and pregnant myometrium were used to examine mRNA levels for four of the genes that were differentially expressed by microarray analysis, namely plasminogen activator inhibitor type 1 (PAI-1), milk fat globule-EGF factor 8 protein (MFGE8), secreted frizzled-related protein 4 (sFRP4) and estrogen receptor {alpha} (ER{alpha}). On the microarray these transcripts were up-regulated 7.5-fold for PAI-1 and 4.9-fold for MFGE8 in pregnant myometrium, and down-regulated 3.7-fold for sFRP4 and 2.9-fold for ER{alpha} in pregnancy. Northern blot analyses confirmed these changes. Our findings suggest that microarray technology is a useful tool for examining global changes in gene expression that occur as the myometrium differentiates from non-pregnant to pregnant status. Defining these changes provides new insight into the structural and functional adaptations of human myometrium during pregnancy. Keywords: other

Project description:The identification of cell surface proteins on stem cells or stem cell derivatives is a key strategy for the functional characterization, isolation, and understanding of stem cell population dynamics. Here, using an integrated mass spectrometry and microarray based approach, we analyzed the surface proteome and transcriptome of cardiac progenitor cells (CPCs) generated from the stage-specific differentiation of mouse and human pluripotent stem cells. Through bioinformatic analysis, we have identified and characterized FZD4 as a new marker for lateral plate mesoderm. Additionally, we utilized FZD4, in conjunction with FLK1 and PDGFRA, to further purify CPCs and increase cardiomyocyte (CM) enrichment in both mouse and human systems. Moreover, we have shown that NORRIN presented to FZD4 further increases CM output via proliferation through the canonical WNT pathway. Taken together, these findings demonstrate a role for FZD4 in mammalian cardiac development.
