Gene expression profiles of EpiSC-derived neural stem cell lines in comparison with EpiSC and embryonic CNS

ABSTRACT: Epiblast stem cells (EpiSCs) maintained in the medium with activin were freed from activin to elicit neural development, exposed to various strengths of Wnt signals, which may provide the initial stage of anteroposterior regional specificities, and placed in the culture condition to arrest neural development but expand cells as neural stem cells (NSCs) with epidermal growth factor (EGF) and fibroblast growth factor 2 (FGF2).

Project description:The requirements for self-renewal differ between EpiSCs and ES cells and the underlying mechanism is largely unknown. Here we show that mouse EpiSCs can be efficiently derived and robustly propagated even from single cells, using two small-molecule inhibitors: CHIR99021 and XAV939. The whole-genome microarray analyses is performed to confirm the identity of EpiSC maintained in CHIR/XAV by comparing the expression profile in EpiSC-CHIR/XAV to those in ESC maintained in 2i and EpiSC maintained in FGF2/activin. Sigle-cell transcriptome analysis of EpiSC-derived neural stem cell lines derived from EpiSCs under different Wnt signal inputs

Project description:The four neural stem cell lines, XN4, XN1, KN1, and CN4, were derived from EpiSCs under different Wnt signal inputs (Nakamura et al., to be submitted). Microarray analysis of the transcripts indicated that these cells have different anteroposterior regional characteristics of the embryonic CNS from the forebrain to the thoracic spinal cord. However, each cell line has a relatively broad anteroposterior regional coverage. For instance, the XN1 line expressed midbrain-characteristic genes and hindbrain-characteristic genes. Single-cell transcriptome analysis of the four cell lines was performed to confirm that the broad anteroposterior specificity is intrinsic to individual cells of the cell line rather than the cell lines are composite of separable subpopulations with narrower anteroposterior specificities.

Time-series of gene expression in female murine ES cells during EpiSC differentiation

Project description:Embryonic stem cells (ESC) are derived from the inner cell mass of the blastocyst in the presence of leukemia inhibitory factor (LIF). In vivo these cells then differentiate into epi stem cells (EpiSC) that can be derived from the Epiblast in presence of Fgf2 and ActivinA. In this study, female ESCs cultured in 2i medium have been differentiated into EpiSC for 3.5 days in vitro by addition of Fgf2 and Activin A. The gene expression profile was analyzed every 4-12 h using mouse exon arrays.

Project description:All data are derived from the same batch of human ES cells (line H9, WA-09). With the exception of the â??UDâ?? file all files represent neural cell types derived from the undifferentiated hESCs. Following mechanical isolation, rosettes were maintained for various passages in the presence of defined growth factors and morphogen factors as indicated below at the R-NSC stage. Cells at the NSCFGF/EGF stage were derived from mechanically isolated neural rosettes that were purified and long-term expanded as attached monolayer cultures in serum free medium in the presence of FGF2/EGF. Expression profiles in HMGN1-overexpressed neural precursor cells

Project description:Neural precursor cells (NPCs) are multipotent cells that can generate neurons, astrocytes, and oligodendrocytes in the mammalian central nervous system. Although high mobility group nucleosomal binding domain 1 (HMGN1) was highly expressed in NPCs, its functions in neural development are not fully understood. We performed microarray analysis to examine changes in gene expression between control and HMGN1-overexpressed NPCs. NPCs derived from E11.5 mouse forebrains were infected with control or HMGN1 retrovirus in the presence of fibroblast growth factor 2 (FGF2) and epidermal growth factor (EGF). Three days after infection, the virus-infected NPCs were dissociated and seeded on poly-D-lysine -coated dishes, and subsequently the cells were cultured for 1 day without FGF2 and EGF.
