Bulk RNA-seq of mouse ileum of Lactobacillus rhamnosus GG or Ruminococcus gnavus monoassociated mice.

ABSTRACT: Lacticaseibacillus rhamnosus GG (LGG) is a widely consumed probiotic whose potential beneficial effects in humans have been examined in over 250 clinical trials. However, the mechanisms by which LGG modulates host gut physiology remains unknown. R. gnavus is a pathobiont that is strongly associated with inflammatory bowel diseases. Germ free mice were mono-associated with L. rhamonosus (LGG) or R. gnavus (RG) for 21 days, ileum bulk RNA-seq were performed to compare the transcriptomic profiles of LGG or RG associated mice with the transcriptome of germ-free mouse ileum.

Milk protein based cryogel monoliths as novel encapsulant of probiotic bacteria. Part II: L. rhamnosus GG storage stability and bioactivity under in vitro digestion

Project description:The aim of the study was to assess the ability of milk protein-based cryogel monoliths to preserve the biological role of Lacticaseibacillus rhamnosus GG (LGG) under cryogenic processing, temperature and water activity controlled static storage and in vitro gastrointestinal transit conditions. Moreover, the matrix disintegration, digestibility, and release of bioactive peptides in the simulated gastrointestinal fluids was determined. The cryogelation process did not confer any significant cellular injuries (losses < 0.5 logCFU g-1) due to the achievement of a vitreous state under the implemented cryogenic conditions. Among the tested storage conditions i.e., temperature (5, 20 and 37 °C) was the most influential parameter of the LGG inactivation kinetics (kLGG = 0.002-0.008 to 0.25-0.56 day-1) followed by the protein composition (i.e., the presence of sodium caseinate alleviated the lethality of LGG). Cryogels stored at aw ≤ 0.33 (found in the vicinity or below Tg) allowed good LGG survivability (kLGG 0.033-0.045 day-1) compared to their rubbery analogues (i.e. aw > 0.33). All cryogels exerted a very limited disintegration during the oral and gastric processing steps helping the LGG cells to overcome the harsh gastric stressors. A complete disintegration of the cryogels was achieved during the intestinal step favouring the release of LGG cells and the advancement of the cryogel matrix digestibility (> 64%). The viable counts of LGG in the intestinal phases remained satisfactorily high (> 8.2 logCFU g-1). Peptidomic profiling of the gastric and intestinal phases confirmed the release of ACE and DPP-IV inhibitory, antimicrobial and antioxidant peptides.

Project description:The goal was to assess the impact of carbon source and cross-feeding on transcription profile of both bacteria. R. gnavus was grown in monoculture with glucose (Glc). R. bromii was grown in monoculture with soluble (SS) or resistant (RS) starch. R. bromii and R. gnavus were co-cultured with SS or RS. Total RNA was extracted from a culture sample taken at mid- to late exponential phase of growth. rRNA was depleted and mRNA sequenced. 3 biological replicates were prepared for each condition.

Antifungal defense of probiotic Lactobacillus rhamnosus GG is mediated by blocking adhesion and nutrient depletion

Project description:Candida albicans is an inhabitant of mucosal surfaces in healthy humans but also the most common cause of fungal nosocomial blood stream infections, associated with high morbidity and mortality. As such life-threatening infections often disseminate from superficial mucosal infections we aimed to study the use of probiotic Lactobacillus rhamnosus GG (LGG) in prevention of mucosal C. albicans infections. Here, we demonstrate that LGG protects oral epithelial tissue from damage caused by C. albicans in our in vitro model of oral candidiasis. Furthermore, we provide insights into the mechanisms behind this protection and dissect direct and indirect effects of LGG on C. albicans pathogenicity. C. albicans viability was not affected by LGG. Instead, transcriptional profiling using RNA-Seq indicated dramatic metabolic reprogramming of C. albicans. Additionally, LGG had a significant impact on major virulence attributes, including adhesion, invasion, and hyphal extension, whose reduction, consequently, prevented epithelial damage. This was accompanied by glucose depletion and repression of ergosterol synthesis, caused by LGG, but also due to blocked adhesion sites. Therefore, LGG protects oral epithelia against C. albicans infection by preventing fungal adhesion, invasion and damage, driven, at least in parts, by metabolic reprogramming due to nutrient limitation caused by LGG.

Transcription profiling of mouse adult colon cells treated with probiotic Lactobacillus GG

Project description:YAMC (young adult mouse colon) cells are a conditionally immortalized mouse colonic intestinal epithelial cell line derived from the Immortimouse.Prior to each experiment, cells were plated at a density of 7.5 x 105 cells per p100mm dish.Cells were treated with LGG (the probiotic Lactobacillus GG ) conditioned media overnight, then harvested the following day. RNA were extracted with Trizol and purified with Qiagen kit. Affymetrix microarray chip Mouse 430A containing 19,000 murine genes was run in duplicate(experiment on 7-24-03 and 1-22-04 ). Data were analyzed using Affymetrix Genechip Operating Software (GCOS, Version 1.0) In each case; LGG treatment was compared to mock treatment controls. Results are expressed as fold change of treated cells as compared to controls, as calculated using GENESPRING software (Version 4.2.1, Silicon Genetics, Mountain View, CA), Statistical analysis was performed using D chip. DNA microarray experiments demonstrate that epithelial cell hsp70 is one of the most highly upregulated genes in response to LGG-CM treatment

Transcription profiling of mouse ileum in response to colonization with a zebrafish or mouse gut microbiota

Project description:We compared gene expression in the small intestine (ileum) of mice that were either (i) germ-free, (ii) colonized with a conventional mouse cecal microbiota, (iii) colonized with a conventional zebrafish gut microbiota, or (iv) colonized with Pseudomonas aeruginosa PAO1. Experiment Overall Design: Adult germ-free NMRI mice were colonized with either (i) a conventional mouse cecal microbiota harvested from adult Swiss-Webster mice (5 biological replicates), (ii) a conventional zebrafish intestinal microbiota harvested from adult C32 zebrafish (3 biological replicates), or (iii) a culture of Pseudomonas aeruginosa PAO1 (5 biological replicates). 14 days after colonization, total RNA was prepared from the ileum of each animal, with total RNA prepared from adult germ-free NMRI mouse ileum serving as negative controls (5 biological replicates). RNA was used as template to generate cRNA for hybridization to Affymetrix 430 v2 Mouse GeneChips.

Lactobacillus rhamnosus GG modifies the metabolome of pathobionts in gnotobiotic mice

Project description:Lactobacillus rhamnosus GG (LGG) is the most widely used probiotic, but the mechanisms underlying its beneficial effects remain unresolved. Previous studies typically inoculated LGG in hosts with established gut microbiota, limiting the understanding of specific impacts of LGG on host due to numerous interactions among LGG, commensal microbes, and the host. There has been a scarcity of studies that used gnotobiotic animals to elucidate LGG-host interaction, in particular for gaining specific insights about how it modifies metabolome. To evaluate whether LGG affects the metabolite output of pathobionts, we inoculated LGG into gnotobiotic mice along with a gut-disruptive consortium containing Propionibacterium acnes, Turicibacter sanguinis, and Staphylococcus aureus (PTS).
