Transcriptome sequencing of tumor associated macrophages cells in HCC liver of OXCT1mKO mice.

ABSTRACT: 6-8 weeks male LysCre/+ OXCT1fl/fl (OXCT1mKO or mKO for short) and its littermates Lys+/+ OXCT1fl/fl (control or ctrl) mice were used to build HCC implantation model (Hep1-6 in Portal Vein injection). RNA was extracted from tumor associated macrophages of OXCT1mKO and their littermate control mice for transcriptome analysis of macrophage function.

Keyword: Circadian Transcriptome, time course 72 samples were analyzed, comprised of 4 experimental groups (Ctrl SOL, mKO SOL, Ctrl TA, mKO TA), with 3 biological replicates for each time point sampled every 4 hours for 24 hours. Identification of a portal vessel-associated fetal liver hematopoietic stem cell niche

Project description:Whereas the cellular basis of the hematopoietic stem cell (HSC) niche in the bone marrow has been characterized, the nature of the fetal liver (FL) niche is not yet elucidated. We show that Nestin+NG2+ pericytes associate with portal vessels, forming a niche promoting HSC expansion. Nestin+NG2+ cells and HSCs scale during development with the fractal branching patterns of portal vessels, tributaries of the umbilical vein. After closure of the umbilical inlet at birth, portal vessels undergo a transition from Neuropilin-1+Ephrin-B2+ artery to EphB4+ vein phenotype, associated with a loss of peri-portal Nestin+NG2+ cells and emigration of HSCs away from portal vessels. These data support a model in which HSCs are titrated against a peri-portal vascular niche with a fractal-like organization enabled by placental circulation.

Quadriceps femoris muscles were harvested from 22-month-old muscle-specfic ATF4 knockout (ATF4 mKO) mice and littermate controls. mRNA levels in ATF4 mKO muscles were normalized to levels in littermate control muscles.

The effect of IL-37 in HCC cell line Sk-hep1

Project description:We performed whole microarray expressiong prolife of Sk-hep1 cells which could over-express IL-37. This project is to explore the effect of interleukin-37 on hepatocellular carcinoma(HCC) development. We transfected human HCC cell line Sk-hep1 with lentiviral vector encoding IL-37 or control. The successfully transfected cells were sorted by FACS and the cells were further cultured for 24 hours. The cells were harvested and send for microarray analysis.
