A combination treatment based on drug repurposing demonstrates mutation agnostic efficacy in pre-clinical retinopathy models. [RNA-seq: Rd10]

ABSTRACT: To determine the gene expression changes in the retina of two separate groups (experimental groups: vehicle and TMB) of vivarium-reared 5-week-old Rd10 mice (strong disease), as well as one group of 4-week-old dark-reared Rd10 mice (mild disease) and one group of wild-type mice (healthy). TMB group received dietary administration of tamsulosin, metoprolol and bromocriptine drug cocktail between 4-5-weeks of age, for 9 days prior to sample collection. All other groups received base/vehicle diet.

Analysis of 20-HETE supplementation in calorically restricted C57Bl6 mice

Project description:Proteomic dataset of 15 murine kidneys of 14-week-old male C57Bl6 mice. Mice were divided into 3 treatment groups four weeks prior to nephrectomy and 5 mice had ad libitum access to food and water, while another 10 mice received 30% food restriction. Calorically restricted mice either received intraperitoneal supplementation of 20-HETE or vehicle (EtOH in sodiumchloride 0.9%) for 8 days on a daily basis prior to nephrectomy. Ad libitum fed mice received vehicle injections for 8 days on a daily basis prior to nephrectomy.

RNA-seq of Atlantic salmon (Salmo salar) liver sampled at various stages through smoltification

Project description:The anadromous Atlantic salmon undergo preparatory physiological transformations before seawater entry, referred to as smoltification. Little is known about the photoperiod-influence and genome regulatory processes driving smoltification such as the large scale changes in lipid metabolism and energy homeostasis in the developing smolt liver. To shed light on this, we performed a smoltification trial using contrasting photoperiod regimes and generate a transcriptome data from livers throughout smoltification and after seawater transfer. In this experiment two groups of Atlantic salmon were reared for a total of 46 weeks from the parr stage, through smoltification, and seawater transfer. After 21 week from first feeding, the experiment group was given artificial winter photoperiod (8 hours light, 16 hours dark) for 8 weeks to induce smoltification before returning to constant light. The second control group received constant light throughout the experiment. Liver tissue was sampled from individuals first at week 1, 21 weeks after first feeding, then again at week 10, after the winter period, at week 19, after the expected smoltification time, and lastly at week 25, after transfer to seawater.

Gene expression of Tamoxifen induced control (TC), dormancy-like (TD) or relapse (TR) cell samples

Project description:To generate a dormancy state in vivo, female BALB/c athymic mice with ER+MC cell (MCF7) derived xenografts were treated with Tamoxifen (TAM). Throughout the treatment period, no significant differences in the body weight of the TAM-treated groups (TD and TR) compared to the vehicle-treated groups (TC) were observed. Whereas xenografts in the vehicle-treated group (as control group: TC) continued to increase in volume, the xenograft volumes of the TAM-treated group regressed 20%-30% in the first week and then remained stable during the four-week administration of TAM. Thereafter, half of the xenografts in TAM-treated mice were resected at random and designated as a dormancy-like group (TD). For the remaining TAM treated xenografts, treatment was discontinued and xenograft volume was observed to gradually increase. This group was defined as the relapse group (TR) and would mimic relapses observed after discontinuation of anti-estrogen therapy in patients.
