Andes virus mRNA vaccines: comparison of unmodified and modified mRNA platforms [VDJ-seq]

Project description:Andes virus (ANDV) is a rodent-borne zoonotic orthohantavirus endemic in South America that causes hantavirus pulmonary syndrome in humans, with up to a 40% case fatality rate. We developed ANDV mRNA vaccines based on the M segment of the viral genome that codes for glycoproteins Gn and Gc in a single open reading frame of glycoprotein precursor (GPC). We generated RNAs either with regular uridine (U-mRNA) or N1-methylpseudouridine (m1Ψ-mRNA). Mice immunized by either ANDV U-mRNA or m1Ψ-mRNA developed similar germinal center responses in lymph nodes. Single cell RNA and BCR sequencing of germinal center B cells from vaccinated mice demonstrated similar levels of activation, except an additional cluster of cells exhibiting strong interferon response that was present in animals vaccinated with U-mRNA but not m1Ψ-mRNA. Furthermore, similar immunoglobulin class-switching and somatic hypermutations were observed for the two vaccines. Golden Syrian hamsters were immunized intramuscularly with 2 doses of the vaccines on days 0 and 21. The titers of Gn/Gc-binding antibodies were moderately greater for U-mRNA construct than for m1Ψ-mRNA construct, however, the titers of ANDV-neutralizing antibodies were equivalent. Vaccinated animals were challenged with a lethal dose of ANDV at 21 days after the boost, along with the naïve control group. All control animals succumbed to infection whereas all vaccinated animals survived without any detectable disease or viral load. The data demonstrate the development of effective vaccines against ANDV and the lack of a significant effect of m1Ψ mRNA modification on immunogenicity and protection in the hamster model.

Platynereis head proteome - Combined transcriptome and proteome profiling reveals specific molecular brain signatures for sex, maturation and circalunar clock phase

Project description:A maturation, sex and lunar phase locked Platynereis head proteome was generated by sampling six heads per biological replicate from male and female Platynereis at defined stages of sexual maturation (immature, premature and mature) at two different moon phases; New moon and Free-running Full moon, meaning the animals would expect a nocturnal light stimulus, Full moon, which was not given. From these heads RNA and protein were extracted simultaneously and used for both transcriptomics (ENA accession no. PRJEB27496) and quantitative proteomics. In total 2290 proteins were identified with two unique peptides as requirement; of these, 1064 proteins were found to be quantifiable (identification in two out of three biological replicates and three out of five technical replicates). Relative quantification revealed that the majority of quantifiable proteins changed during maturation (693 out of 1064 proteins), sexual differences accounted for only 17 differentially expressed proteins, whereas lunar phase differences showed 261 differentially expressed proteins. This finding is in contrast to what was found on the transcriptome levels where differences in between the lunar phases only accounted for only 64 out of 52059 transcripts. To confirm this high degree of lunar regulated proteins a second proteome from immature animals at New moon and Free-running Full moon was undertaken. In this second set, we identified 2095 proteins of which 1671 were considered quantifiable. Of these 173 proteins showed significant differences between the New moon and the Free running full moon phase, thus confirming the previous results of a higher regulation on the protein compared to the transcript levels.

Project description:The study is a Phase 1 Open-label Two-stage, Safety and Tolerability Study with Cancer Type-specific Cohorts, Evaluating Subcutaneous Administration of Andes-1537 for Injection in Patients with Advanced Solid Tumors that are Refractory to Standard Therapy or For Which No Standard Therapy Is Available. Patients with unresectable solid tumors that are refractory or have failed standard therapy and are deemed non-eligible or intolerant to further therapy or for which no standard therapy is available will be included in 5 cancer type-specific parallel cohorts. The following tumor types will be evaluated for potential inclusion in each cancer type-specific cohort: gallbladder & biliary tract carcinoma; cervical carcinoma; gastric carcinoma; pancreatic carcinoma, and colorectal carcinoma.
