Project description:Eyes are complex, delicate sensory organs that can be irreversibly damaged by disease or aging. Finding solutions to repair eyes has been difficult because no animal model has been available to study the complete regeneration of a camera-type eye. To study camera-type eye regeneration we can study partial or limited regeneration in vertebrates or complete regeneration of different types of eyes with simpler structure in animals with higher regenerative potential. I discovered an invertebrate organism, the freshwater apple snail Pomacea canaliculata, with camera-type eyes, yet capable of full regeneration after complete amputation. After cellular and molecular study of P. canaliculata eye and their regeneration dynamic, we developed tools to edit genes and test their functions. We found that, like in humans, eye development in these animals is controlled by the expression of the gene pax6. It is now possible to unravel the genetic basis of camera-type eye regeneration, which may provide new opportunities for therapeutic interventions. This work represents the foundation to use P. canaliculata as a valuable research system in the field of regeneration, not only to bring more knowledge about complex organs regeneration, but also to shed light, for the first time, on the process of complete camera-type eye regeneration.

Function and evolutionary origin of unicellular camera-type eye structure.

Project description:The ocelloid is an extraordinary eyespot organelle found only in the dinoflagellate family Warnowiaceae. It contains retina- and lens-like structures called the retinal body and the hyalosome. The ocelloid has been an evolutionary enigma because of its remarkable resemblance to the multicellular camera-type eye. To determine if the ocelloid is functionally photoreceptive, we investigated the warnowiid dinoflagellate Erythropsidinium. Here, we show that the morphology of the retinal body changed depending on different illumination conditions and the hyalosome manifests the refractile nature. Identifying a rhodopsin gene fragment in Erythropsidinium ESTs that is expressed in the retinal body by in situ hybridization, we also show that ocelloids are actually light sensitive photoreceptors. The rhodopsin gene identified is most closely related to bacterial rhodopsins. Taken together, we suggest that the ocelloid is an intracellular camera-type eye, which might be originated from endosymbiotic origin.

Dynamically tunable hemispherical electronic eye camera system with adjustable zoom capability.

Project description:Imaging systems that exploit arrays of photodetectors in curvilinear layouts are attractive due to their ability to match the strongly nonplanar image surfaces (i.e., Petzval surfaces) that form with simple lenses, thereby creating new design options. Recent work has yielded significant progress in the realization of such "eyeball" cameras, including examples of fully functional silicon devices capable of collecting realistic images. Although these systems provide advantages compared to those with conventional, planar designs, their fixed detector curvature renders them incompatible with changes in the Petzval surface that accompany variable zoom achieved with simple lenses. This paper describes a class of digital imaging device that overcomes this limitation, through the use of photodetector arrays on thin elastomeric membranes, capable of reversible deformation into hemispherical shapes with radii of curvature that can be adjusted dynamically, via hydraulics. Combining this type of detector with a similarly tunable, fluidic plano-convex lens yields a hemispherical camera with variable zoom and excellent imaging characteristics. Systematic experimental and theoretical studies of the mechanics and optics reveal all underlying principles of operation. This type of technology could be useful for night-vision surveillance, endoscopic imaging, and other areas that require compact cameras with simple zoom optics and wide-angle fields of view.

Assembly of the cnidarian camera-type eye from vertebrate-like components.

Project description:Animal eyes are morphologically diverse. Their assembly, however, always relies on the same basic principle, i.e., photoreceptors located in the vicinity of dark shielding pigment. Cnidaria as the likely sister group to the Bilateria are the earliest branching phylum with a well developed visual system. Here, we show that camera-type eyes of the cubozoan jellyfish, Tripedalia cystophora, use genetic building blocks typical of vertebrate eyes, namely, a ciliary phototransduction cascade and melanogenic pathway. Our findings indicative of parallelism provide an insight into eye evolution. Combined, the available data favor the possibility that vertebrate and cubozoan eyes arose by independent recruitment of orthologous genes during evolution.
