While it has been observed that Plasmodium falciparum infection of late-stage erythroblasts can delay terminal erythroid differentiation and enucleation, the mechanism(s) underlying this phenomenon are unknown. Here, we apply RNA-seq after fluorescence-activated cell sorting (FACS) of infected erythroblasts to identify transcriptional responses to direct and indirect interaction with P. falciparum.

Changes in the erythroblast miRNA profile in the bone marrow of a Plasmodium vivax-infected patient

Project description:Human studies of Plasmodium vivax in the bone marrow are scarce. Here, we present a detailed characterization of bone marrow aspirates taken from a P. vivax patient with high parasitaemia on admission and 42 days after treatment. Analysis of miRNAs related to erythropoiesis revealed a distinct series of differentially expressed miRNAs during infection compared to at convalescence. These results suggest that parasites in the bone marrow affect erythropoiesis.

Single cell data for plasmodium replication in mouse red blood cells

Project description:Hypoglycemia is a clinical hallmark of severe malaria, the often-lethal outcome of Plasmodium falciparum infection. Yet, the underlying mechanisms driving the pathogenesis of malaria-associated hypoglycemia remain poorly understood. Here we report that labile heme, an alarmin generated as a byproduct of hemolysis during the blood stage of Plasmodium spp. infection, plays a central role in the development of malaria-associated hypoglycemia. Labile heme recapitulated the hypometabolic response to Plasmodium (chabaudi chabaudi; Pcc) infection in mice, including the development of anorexia, transcriptional repression of hepatic glucose production (HGP) and reduction of glycemia, energy expenditure (EE) as well as core body temperature. While this hypometabolic response is protective against immune-mediated liver damage and anemia, when sustained over time it can lead to hypoglycemia and compromise EE as well as thermoregulation. I response, asexual stages of Plasmodium spp. activate a transcriptional program that reduces virulence in favor of sexual commitment and presumably malaria transmission. In conclusion, malaria-associated hypoglycemia represents a trade-off of a hypometabolic defense strategy against Plasmodium infection.

Circadian Control of the Response of Macrophages to Plasmodium spp.-Infected Red Blood Cells.

Project description:In this study, we sought to determine whether the intensity of the immune response to Plasmodium spp., the parasite causing malaria, depends on time of infection. For this, we considered the known existence of a circadian clock in immune cells which regulates several aspects of the immune response. Experiments were performed using mouse bone marrow-derived macrophages (BMDMs) stimulated ex vivo with red blood cells infected with Plasmodium berghei ANKA (iRBCs) or uninfected RBCs. First, we identified that lysed iRBCs triggered an immune response in macrophages. Then, by stimulating at 4 different circadian time points (16 h, 22 h, 28 h or 34 h post-synchronization of the cells clock), rhythms in reactive oxygen species and cytokines/chemokines were found. Furthermore, the analysis of the macrophage proteome and phosphoproteome revealed global changes according to treatment (iRBC or RBC) and to the circadian time point. In summary, our findings showed that the circadian clock within macrophages determines the magnitude of immune response upon stimulation with iRBCs, along with changes of the cell proteome and phosphoproteome.

Searching for new molecules involved in Anopheles mosquitoes’ response to Plasmodium infection

Project description:Plasmodium parasites within mosquitoes are exposed to various physiological processes, such as blood meal digestion activity, the gonotrophic cycle, and host responses preventing the entry of parasites into the midgut wall. However, when in vitro-cultured ookinetes are injected into the hemocoel of mosquitoes, Plasmodium parasites are not affected by the vertebrate host’s blood contents and do not pass through the midgut epithelial cells. This infection method might aid in identifying mosquito-derived factors affecting Plasmodium development within mosquitoes. This study investigated novel mosquito-derived molecules related to parasite development in Anopheles mosquitoes. We injected in vitro-cultured Plasmodium berghei (ANKA strain) ookinetes into female and male Anopheles stephensi (STE2 strain) mosquitoes and found that the oocyst number was significantly higher in males than in females, suggesting that male mosquitoes better support the development of parasites. Next, RNA-seq analysis was performed on the injected female and male mosquitoes to identify genes exhibiting changes in expression. Five genes with different expression patterns between sexes and greatest expression changes were identified as being potentially associated with Plasmodium infection. Two of the five genes also showed expression changes with infection by blood-feeding, indicating that these genes could affect the development of Plasmodium parasites in mosquitoes.

Transcription profiling of mouse hepatocytes reinfected with Plasmodium berghei

Project description:We show that an ongoing malaria blood stage infection impairs the establishment of Plasmodium sporozoites in hepatocytes and that secondary infections can only be established after a previous infection has been cleared from circulation. Using control mice, mice infected with sporozoites only, mice infected by iRBCs only or mice reinfected, we show that this impairment is not due to an effect of the acquired host immune response or to a decrease in host cell survival. Instead, an ongoing blood stage infection leads to a significant increase in the expression of hepcidin, a peptide hormone that is secreted by the liver and controls body iron homeostasis. A rapid increase of hepcidin levels during blood stage infection causes sequestration of iron in storage forms within cells of reticuloendothelial system decreasing its availability in hepatocytes, where it is required for Plasmodium sporozoite establishment.

Splenic immune response to Plasmodium infection initiated by two different routes

Project description:Plasmodium chabaudi parasite obtained from recently mosquito transmitted infections give rise to an attenuated blood-stage infection when compared to serially blood passaged parasites. We compared the immune response in the spleens induced by these two types of parasites in order to determine what may cause this attenuation. Spleens were taken at several timepoints during the blood-stage infection and RNA-seq analysis was performed.
