Single-cell transcriptome analysis of tumor infiltrated CD45+ immune cells isolated from ctrl- or thonzonium bromide- treated mice

ABSTRACT: As a monocationic detergent, thonzonium bromide (TB) has been approved as an additive to pharmacologic formulations, serving to promote tissue contact of the administered drugs by dispersion and penetration of the cellular debris and exudate, but has not been used for cancer treatment. In this study, we performed single-cell RNA sequencing (scRNA-Seq) to identify the composition of the immune microenvironment after TB treatment.

Mepenzolate bromide displays beneficial effects in a mouse model of chronic obstructive pulmonary disease

Project description:For the clinical treatment of chronic obstructive pulmonary disease (COPD), it is important not only to improve the airflow limitation by bronchodilation but also to suppress emphysema by controlling inflammation. In this study, we have screened for compounds that prevent elastase-induced airspace enlargement in mice from medicines already used clinically. Mepenzolate bromide, a muscarinic antagonist used to treat gastrointestinal disorders was selected. Intratracheal administration or inhalation of mepenzolate bromide decreased the severity of elastase-induced airspace enlargement, alteration of lung mechanics and respiratory dysfunction. While mepenzolate bromide showed bronchodilatory activity, most of other muscarinic antagonists tested did not improve the elastase-induced pulmonary disorders. Mepenzolate bromide suppressed elastase-induced pulmonary inflammatory responses and production of superoxide anions, and reduced the level of cigarette smoke-induced airspace enlargement and alteration of lung mechanics. Based on these results, we propose that this drug is therapeutically effective for COPD as a consequence of both its anti-inflammatory and bronchodilatory activities.

Uncover New Reactivity of Genetically Encoded Alkyl Bromide Unnatural Amino Acids

Project description:Genetically encoded unnatural amino acids (Uaas) with electrophilic moieties are excellent tools to investigate protein-protein interactions (PPIs) both in vitro and in vivo. These Uaas, including a series of alkyl bromide-based Uaas, mainly target cysteine residues to form protein-protein cross-links. Although some reactivities towards lysine and tyrosine residues have been reported, a comprehensive understanding of their reactivity towards a broad range of nucleophilic amino acids is lacking. Here we used a recently developed OpenUaa search engine to perform an in-depth analysis of mass spec data generated for Thioredoxin and its direct binding proteins cross-linked with an alkyl bromide-based Uaa, BprY. The analysis showed that, besides cysteine residues, BprY also targeted a broad range of nucleophilic amino acids. We validated this broad reactivity of BprY with Affibody/Z protein complex. We then successfully applied BprY to map a binding interface between SUMO2 and SUMO-interacting motifs (SIMs). BprY was further applied to probe SUMO2 interaction partners. We identified 264 SUMO2 binders, including several validated SUMO2 binders and many new binders. Our data demonstrated that BprY can be effectively used to probe protein-protein interfaces even without cysteine residues, which will greatly expand the power of BprY in studying PPIs.

Expression data from Caenorhabditis elegans L1-stage larvae exposed to ethidium bromide and/or serial ultraviolet C radiation

Project description:Ultraviolet C radiation (UVC) damages the nuclear and mitochondrial genomes; this damage is repaired in the nuclear but not mitochondrial genome. Ethidium bromide (EtBr) inhibits mitochondrial DNA replication. We were interested in the transcriptomic response to exposure to UVC, EtBr, and the combination. The UVC exposure protocol results in a high level of mitochondrial DNA damage, and a low level of nuclear DNA damage (because of repair). We exposed age-matched L1-stage Caenorhabditis elegans to ultraviolet C radiation (UVC ) three times, separated in time by 24 h, in the absence of food. After the third exposure, larvae were placed on K agar plates with OP50 bacterial food. In some cases ethidium bromide was also used. Nematodes were sampled for RNA isolation several times.

Project description:Cancer associated fibroblasts (CAFs) as the main stromal cells in tumor microenvironment can provide metabolic links with tumor cells through metabolic derivatives such as cytokines released, creating a favorable metabolic microenvironment for tumor progression. Recent in vitro and in vivo studies have discovered that anesthetics can affect tumor malignancy. However, the mechanism is unknown, and the total-body dosage and short-term treatment duration of anesthetic drugs are still far from tumor treatment. Now, our study focuses on the effect of rocuronium bromide (RB) on CAFs. In order to restore the true effects of CAFs by RB, we studied the global transcriptomic signatures of human fibroblasts derived from different tissues (skin HS-27; lung TIG-1; dermal CRL-7815) by 160 μg/mL RB to find the target genes responding to RB.

Effect of choline kinase inhibitor hexadecyltrimethylammonium bromide on Plasmodium falciparum gene expression

Project description:Investigations on the fundamental of malaria parasite biology, such as invasion, growth cycle, metabolism and cell signalling have uncovered a number of potential antimalarial drug targets, including choline kinase, a key enzyme involved in the synthesis of phosphatidylcholine, an important component in parasite membrane compartment. The effect on gene expression of Plasmodium falciparum K1 strain following 72 hours exposure to 2 μM (IC50 concentration) of the choline kinase inhibitor, hexadecyltrimethylammonium bromide (HDTAB) was evaluated by DNA microarray analysis. Genes important in P. falciparum intra-erythrocytic life cycle, such as invasion, cytoadherance and growth were among those affected by at least 2-fold changes in their expression levels compared with non HDTAB-treated control.

Microarray whole transcriptome profiling of Sepantronium bromide (YM-155) treated MDA-MB-231 cells

Project description:Sepantronium bromide (YM-155) is believed to trigger apoptosis in tumor cells by inducing a major reduction in baculoviral inhibitor of apoptosis repeat-containing 5 (BIRC 5) also known as survivin. In MDA-MB-231 cells, the toxic effects of YM-155 were equi potent to standard chemotherapy drugs , having an LC50 < 50ng/ml. In this study we investigate whole transcriptome changes occurring at 8 hours of incubation with YM-155, preceding apoptosis as well as at a 20 hour time point. Microarrays were acquired for mRNAs and long intergenic non-coding RNA transcripts using the GeneChip™ Human Gene 2.1 ST Arrays by Affymetrix Inc.
