Gene expression profiling of dendritic cells in different physiological stages under Cordyceps sinensis treatment

ABSTRACT: Cordyceps sinensis (CS) has been commonly used as a herbal medicine as well as a health supplement in China for over two thousand years. Although previous studies demonstrated that CS has benefit in immunoregulation and anti-inflammation, the precise mechanism by which CS affects immunomodulation is still unclear. In this study, we adopted duplicate sets of loop-design microarray experiments to examine two different batches of CS and analyzed the effects of CS on dendritic cells (DCs) in different physiology stages, naïve stage and inflammatory stage.

We used duplicate sets of loop-design microarray in this study. Each loop experiment consisted of 4 RNA samples, including untreated samples, CS 1μg/ml, LPS 1μg/ml, and LPS 1μg/ml+ CS 1μg/ml.

Immunomodulatory mechanism of Cordyceps militaris polypeptide through regulating gene in mice

Project description:Cordyceps militaris and Cordyceps sinensis belong to belong to the same genus, but the different species, with immunity improvement, antibacterial, and antihypertensive effect, and the studies on the functions of Cordyceps militaris mainly focus on those of its polysaccharides and polypeptides. The latest studies have found that some of the polypeptides with immunomodulatory effect can widely regulate immune functions at multiple levels, improve immunity, and enhance immune functions to ensure the healthy body, showing an important significance. Cordyceps militaris polypeptide prepared with the enzymolysis method was taken as the research object in this study. The differentially expressed genes and the related cell signal transduction pathway were screened by mRNA expression microarray. STEM 1.3.6 software was used for the clustering of the gene functions, and David and KEGG database were applied for the analysis of the related functions. 1748 differentially expressed genes were selected finally and three of them were validated by qPCR. The results showed that gene Hist1h2bp, Ctsg, and Elane were involved in the regulation of Cordyceps militaris on the immune activity of mice. Gene Hist1h2bp, Ctsg and Elan may be the potential targets of Cordyceps militaris polypeptide, which may provide an important theory basis for the further research and development of Cordyceps militaris polypeptide.

1H NMR spectroscopy-based metabolic profiling of Ophiocordyceps sinensis and Cordyceps militaris of water-boiled and 50% ethanol-soaked extracts

Project description:Introduction Ophiocordyceps sinensis, a well-known Chinese complementary herb, is a rare and valuable therapeutic resource. Cordyceps militaris (C. militaris) is a commonly used substitute for O. sinensis. A metabolomic-based approach for exploring the similarities and differences in the metabolites of O. sinensis and C. militaris in water-boiled and 50% ethanol-soaked extracts is of great significance. Objectives To determine a vital role of extraction methodologies in influencing the metabolic composition of herbs, 1HNMR-based profiling was used to characterize the metabolic fingerprints of O. sinensis and C. militaris. Methods To make a distinction between the global metabolite profiling of O. sinensis and C. militaris extracts obtained from either the water-boiled or 50% ethanol-soaked methods, we screened the herbs samples using 1HNMR-based metabolic fingerprints combined with multivariate statistical analysis. Results This study revealed that a total of 43 (82.69% of 52) metabolites were detectable in both O. sinensis and C. militaris. According to the variable importance in projection (VIP) value and p-value from the Mann-Whitney test, 7 metabolites (alanine, aspartate, glutamate, mannitol, ornithine, serine, and trehalose) differed between O. sinensis and C. militaris. Arginine, glucose, putrescine, pyroglutamate, betaine, O-phosphocholine, and xylose differed significantly between the water-boiled and 50% ethanol-soaked methods used to prepare the herb extracts. Conclusion A total of 52 primary metabolites were identified and quantified from O. sinensis and C. militaris samples. The study suggests that a water-boiled extraction is much faster method and strongly recommended over the 50% ethanol-soaked method for both O. sinensis and C. militaris.

Cordyceps promotes MKK7-JNK signaling pathway by reducing NF-κB/GADD45b signaling activation to induce apoptosis in TK-10 renal cell carcinoma

Project description:Cordyceps participates in various pharmacological activities including anti-tumor, and is involved in the regulation of NF-κB signaling pathway. However, the detailed role of cordycepin in suppression of NF-κB signaling pathway is less clear. In this study, we first analyzed the effect of cordyceps on NF-κB activity in TK-10 cells, and found that cordyceps resulted in a dose-dependent reduction in TNF-α-induced NF-κB activation. Here, we show that cordyceps mediated NF-kB inhibition induces apoptosis in TK-10 cells involved the serial activation of caspases. Moreover, we demonstrate that in addition to activating caspases, the cordyceps negatively modulates TNF-α-mediated NF-κB signaling to promote JNK activation, which results in apoptosis, and that NF-kB regulates antiapoptotic factor GADD45b and the JNK kinase MKK7. When the TNFα cytokine binds to the TNF receptor, IκB dissociates from NF-κB. As a result, the active NF-κB translocates to the nucleus. Cordyceps clearly prevented NF-κB from mobilizing to the nucleus, resulting in downregulation of GADD45b, whereas upregulation of MKK7 and phosphorylation of JNK (p-JNK). This increased Bax activation, leading to marked cordyceps-induced apoptosis. Bax subfamily proteins induced apoptosis through caspase-3. Furthermore, siRNA mediated inhibition of MKK7 downregulated p-JNK and The JNK inhibitor SP600125 strongly inhibited Bax. Thus, these results indicate that cordyceps inhibits NF-κB/GADD45b signaling activation to upregulate MKK7-JNK signaling pathway to induce apoptosis in TK-10 cells and support the potential of cordyceps as a therapeutic agent for renal cancer.

Developmental gene expression in Clonorchis sinensis using Custom-fabricated 12K oligonucleotide microarray

Project description:Developmental gene expression patterns (9812 genes) of juvenile and adult C. sinensis were analyzed using a 12K oligonucleotide microarray (a total of 11617 genes). A number of significantly differentially expressed genes having more than 2-fold difference were observed in adult stage compared to juvenile stage. This finding suggests that the expression levels of different gene sets are controlled in C. sinensis for adaptation to environmental change, such as developmental stages.

Transcriptome of Chinese sturgeon (Acipenser sinensis) from the hypothalamic-pituitary-gonad axis in two sex development stages

Project description:Here, the HPG axis transcriptomes of A. sinensis were sequenced using Illumina HiSeq 2000 and 4000 platforms. A total of 271.19 Gb filtered data from 45 samples belonging to 15 individuals (six individuals in stage I, six males and three females in stage II) were obtained, and 74.50% of 121,952 assembled unigenes were annotated to the non-redundant (NR), Swiss-Prot, Kyoto Encyclopedia of Genes and Genomes (KEGG), Cluster of Orthologous Groups of proteins (COG), InterPro, Gene Ontology (GO) and NT databases. Additionally, 78,770 unigenes contained complete open reading frames (ORFs). A total of 51,570 simple sequence repeats (SSRs) were detected, and the number of differentially expressed genes (DEGs) from 1,531 to 34,439 in 18 pairwise comparisons were identified for HPG samples of male and female A. sinensis in two stages. We reported the first integrated transcriptome data from HPG samples of male and female A. sinensis in stage I and II of gonadal development. These data offer valuable resources for research into reproductive regulation and the HPG axis interaction in A. sinensis and other sturgeons.
