Comparative transcriptomic analysis of in vivo and in vitro produced bovine 4-cell stage and 8-cell stage embryos

ABSTRACT: Pre-implantation embryos derived by in vitro fertilization differ in their developmental potential from embryos obtained in vivo. In order to characterize changes in gene expression profiles caused by in vitro culture environment, we employed microarray constructed from bovine embryo-specific cDNAs (BlueChip v., Université Laval, Québec) to compare in vitro cultured and in vivo derived bovine embryos at 4-cell and 8-cell stage.

In order to characterize changes in gene expression profiles caused by in vitro culture environment, we employed microarray constructed from bovine embryo-specific cDNAs (BlueChip v., Université Laval, Québec) to compare in vitro cultured and in vivo derived bovine embryos at 4-cell and 8-cell stage. Analysis directly compared RNA samples from pool of 20 in vivo derived 4-cell stage embryos to RNA samples from pool of 20 in vitro cultured 4-cell stage embryos and RNA samples from pool of 20 in vivo derived 8-cell stage embryos to RNA samples from pool of 20 in vitro cultured 8-cell stage embryos. Three technical replicates, including one dye-swap array were prepared for each developmental stage (4-cell stage, 8-cell stage, respectively) .

Effect of vitrification using the Cryotop method on the gene expression profile of in vitro-produced bovine embryos

Project description:Changes in gene expression induced by the Cryotop vitrification procedure in bovine blastocysts using Agilent EmbryoGENE microarray slides. Bovine in vitro-produced embryos at the blastocyst stage (144 to 156 hours post insemination) were vitrified using the Cryotop system and compared with non-vitrified (control) embryos. After vitrification, the embryos were warmed and cultured for an additional 4 hours. Embryos that re-expanded or developed to the expanded blastocyst stage were used for microarray analysis.

Effects of treatment with bovine oviductal extracellular vesicles on the transcriptome of bovine day 7 in vitro produced embryos

Project description:Oviductal extracellular vesicles (oEVs) are emerging as key players in gamete/embryo-oviduct interactions contributing to successful pregnancy. Various positive effects of oEVs on gametes and early embryos have been found in vitro. To find out if these effects are associated with changes of embryonic gene expression, the embryonic transcriptome was analyzed after supplementation of bovine embryos with bovine oEVs during in vitro culture (IVC) in comparison to corresponding controls without oEVs. Embryos were cultured during 7 days of IVC with fresh oEVs, oEVs frozen after isolation from oviductal fluid or without oEVs (control), since a previous study showed different effects of fresh and frozen oEVs. Five pools of 10-14 embryos were analyzed for each group by low-input Illumina RNA-sequencing. Analysis of differential gene expression revealed 221 differentially expressed genes (DEGs) for the comparison of frozen oEVs vs control, 67 DEGs for fresh vs frozen oEVs, and only minor differences for the comparison fresh oEVs vs control (28 DEGs). An integrative analysis with the previously studied mRNA and small ncRNA content of bovine oEVs suggested direct effects of oEVs content on the embryonic transcriptome for the supplementation with frozen oEVs, i.e., increase of transcripts found in higher concentrations in oEVs and decrease of transcripts which are potential targets of miRNAs found in oEVs.

Comparison of transcriptome of in vitro and in vivo-produced bovine fetuses; specifically in liver and placental tissues.

Project description:During the past several decades, in vitro fertilization (IVF) has been increasingly used both in animal production and human infertility treatment. Animals derived from in vitro manipulation are occasionally associated with abnormal offspring syndrome (AOS) and other developmental abnormalities. By studying gene expression of in vitro-produced (IVP) embryos/animals, we gain an indicator of how well this procedure mimics the in vivo environment. Most previous studies of this nature have focused on only a few genes at a time or have been limited to studying the pre-implantation stage; thus, a global view of how gene transcription may be influenced by in vitro procedures during fetal development has yet to be ascertained. To this end, we collected liver and placental tissue samples from in vitro-produced and in vivo control bovine fetuses at day 90 and 180 of gestation. We used a bovine 13K oligonucleotide microarray to investigate the transcriptional profiles in both tissues and compared them with those of their age-matched in vivo counterparts. Surprisingly, in both liver and placental tissues, the transcriptional profiles between IVP and control fetuses, at either 90 or 180 days of gestation, were indistinguishable. A total of 879 genes were found to be significantly regulated during liver development from 90 to 180 days of gestation, but there were no gene expression changes in the placental tissue during this developmental period. Quantitative real time RT-PCR on 11 selected genes confirmed these results. Our results have certain implications for IVF technologies, both in agriculture and in human medicine. A total of 18 samples for each tissue type (liver & placenta) was analyzed (8 biological replicates for in vivo-produced fetuses and 10 biological replicates for in vitro ones). Two technical replicates were done for each sample.
