SOX4 promotes T cell acute lymphoblastic leukemia progression

ABSTRACT: We report a previously unrecognized role of SOX4 in T cell acute lymphoblastic leukemia (T-ALL). To explore SOX4-dependent gene regulation, RNA-seq analysis was conducted and the differently expressed genes were revealed in the SOX4-knockdown Jurkat cells in comparison to control group.

SOX4 enables oncogenic survival signals in acute lymphoblastic leukemia.

Project description:The Sox4 transcription factor mediates early B-cell differentiation. Compared with normal pre-B cells, SOX4 promoter regions in Ph(+) ALL cells are significantly hypomethylated. Loss and gain-of-function experiments identified Sox4 as a critical activator of PI3K/AKT and MAPK signaling in ALL cells. ChIP experiments confirmed that SOX4 binds to and transcriptionally activates promoters of multiple components within the PI3K/AKT and MAPK signaling pathways. Cre-mediated deletion of Sox4 had little effect on normal pre-B cells but compromised proliferation and viability of leukemia cells, which was rescued by BCL2L1 and constitutively active AKT and p110 PI3K. Consistent with these findings, high levels of SOX4 expression in ALL cells at the time of diagnosis predicted poor outcome in a pediatric clinical trial (COG P9906). Collectively, these studies identify SOX4 as a central mediator of oncogenic PI3K/AKT and MAPK signaling in ALL.

Sox4 is a key oncogenic target in C/EBP? mutant Acute Myeloid Leukemia

Project description:Mutation or epigenetic silencing of the transcription factor C/EBP? is observed in ~10% of patients with acute myeloid leukemia (AML). In both cases, a common global gene expression profile is observed, but down-stream targets relevant for leukemogenesis are not known. Here we identify Sox4 as a direct target of C/EBP? whereby its expression is inversely correlated with C/EBP? activity. Downregulation of Sox4 abrogated increased self-renewal of leukemic cells and restored their differentiation. Gene expression profiles of leukemia initiating cells (LICs) from both Sox4 overexpression and murine mutant C/EBP? AML models clustered together, but differed from other types of AML. Our data demonstrate that Sox4 overexpression resulting from C/EBP? inactivation contributes to the development of leukemias with a distinct LIC phenotype. K/L (bi-allelic Cebpa mutations) leukemic mice and Sox4 overexprssing leukemic mice were used for RNA extraction and hybridization on Affymetrix microarrays. We compared these microarray samples with the C57/BL6 wild type mice.

Project description:Chromosomal instability is a characteristic of cancer, in acute lymphoblastic leukemia, several genes with aberrations have been reported that are currently used as diagnostic tools, prognostics and are even part of the treatment. The CGH microarrays performed in ten adult individuals with ALL, and the JURKAT and CEM cell lines, show that there are regions of shared loss on chromosomes 2, 9 and 14 and of gain on chromosomes 13 and 17 that are maintained in more than five of the samples.
