Elucidation and Engineering of Sphingolipid Biosynthesis Pathway inYarrowia lipolyticaforProduction of Sphingoid Bases and Glycosylceramides

ABSTRACT: This study focused on understanding and modifying the sphingolipid biosynthesis pathway in the yeast Yarrowia lipolytica. By altering specific biological processes within the yeast, the creation of new types of lipid molecules became possible. The research findings highlight the high potential of this yeast in producing non-acetylated sphingoid bases and human-type glucosylceramides, which could serve as valuable components in cosmetics or nutritional supplements.

Project description:Investigation of whole genome gene expression level changes in a Yarrowia lipolytica Y4184 snf1 mutant, compared to the Y4184U+. The Y4184 is an engineered strain to produce eicosapentaenoic acid (EPA) via expression of a M-NM-^T9 elongase/M-NM-^T8 desaturase pathway, and is derived from Yarrowia lipolytica ATCC#20362. A six chip study using total RNA recovered from three separate cultures of Yarrowia lipolytica Y4184Ura+ strains and three separate cultures of Y4184 snf1 mutant strains in which YlSNF1 gene is deleted entirely. Each chip measures the expression level of 6708 genes from Yarrowia lipolytica ATCC#20362 with fourteen 24-mer probe pairs (PM/MM) per gene, with three-fold technical redundancy.

Dietary long-chain omega 3 fatty acids modify sphingolipid metabolism to facilitate asthmatic airway hyperreactivity

Project description:We report the effects of dietary n-3 polyunsaturated fatty acids (PUFAs) on sphingolipid metabolism and airway reactivity in the lung. Mice were fed a n3-PUFA enriched fishoil diet or an isocaloric control coconut oil-enriched diet (CO) for 8 weeks. The n-3 PUFA diet affected pulmonary sphingolipid metabolism, and airway reactivity, independent of inflammation or allergic sensitization. These findings support a role for pulmonary sphingolipid homeostasis in asthma and could have implications for the use of n-3 PUFA dietary supplements in asthma.
