Analysis of human liver transcriptome after exposure to okadaic acid

ABSTRACT: Purpose: Not much research has been done on the effects of okadaic acid on the liver, but it has been shown, that it not only reaches the liver, but has an especially long retention time in it. In this study, differentiated HepaRG cells were incubated bith different, non-cytotoxic okadaic acid concentrations for 24h and the total RNA was used for sequencing. Project description:Okadaic acid (OA) is a marine biotoxin that can induce diarrheic shellfish poisoning upon consumption. It can accumulate in filter-feeding shellfish, thereby reaching the human food chain. In addition to inducing severe gastrointestinal symptoms, OA can pass the intestinal barrier into the bloodstream, thereby reaching the liver. However, the effects of OA on the liver have not been much in the focus of research so far. Therefore, this study now focuses on elucidating changes in the proteome of HepaRG cells induced by OA. Furthermore, as a potent phosphatase inhibitor, we expected changes in the phosphoproteome of the cells. Therefore, we further investigated the phosphoproteome of the HepaRG cells after OA exposure. Results revealed changes in enzyme activity regulation, cytoskeleton organization and energy metabolism.

Project description:Mytilus galloprovincialis (Lmk, 1819) is economically relevant bivalve specie. In Adriatic Sea, periodical temperatures increases define optimal growth conditions for Dinoflagellate spp which can reach high concentrations also in filter-feeding mussels, thus causing potential human health problems. The most commonly used methods for the detection of Diarrhoeic Shellfish Poisoning biotoxins have either a low sensitivity or are too expensive to be used for routine tests. Genomic tools, such as microarray platforms, provide a reliable and alternative solution to overcome these problems. In this study we used a mussel cDNA microarray for studying gene expression changes in mussels exposed to Okadaic acid. Mussels collected in the Gulf of Trieste, located in Northern Adriatic Sea, were fed with Okadaic acid-spiked invertebrates for five weeks. In a time course experiment we were able to describe an early acute response just from the first 4th day time point. Among the differentially expressed genes we found a general up-regulation of stress proteins and proteins involved in cellular synthesis. Overall, we identified 34 transcripts candidate as useful markers to monitor OA-induced stress in mussels. This study contributes to the characterization of many potential genetic markers that could be used in future environmental monitoring, and could lead to explore new mechanisms of stress tolerance in marine mollusc species. Biological replicates: two treated replicates and two control replicates per tissue.

We investigated the impact of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)-mediated AhR activation on a human model of bipotent liver progenitors, undifferentiated HepaRG cells. We used both intact undifferentiated HepaRG cells, and HepaRG cells with silenced Hippo pathway effectors, YAP1 and TAZ, which play key role(s) in tissue specific progenitor cell self-renewal and expansion, including liver, cardiac or respiratory progenitors.
