Human Pancreatic Islets Expressing HNF1A Variant Have Defective β cell Transcriptional Regulatory Networks

Project description:Using an integrated approach to characterize the pancreatic tissue and isolated islets from a 33-year-old with 17 years of type 1 diabetes (T1D), we found donor islets contained β cells without insulitis and lacked glucose-stimulated insulin secretion despite a normal insulin response to cAMP-evoked stimulation. With these unexpected findings for T1D, we sequenced the donor DNA and found a pathogenic heterozygous variant in hepatocyte nuclear factor 1 alpha (HNF1A). In one of the first studies of human pancreatic islets with a disease-causing HNF1A variant associated with the most common form of monogenic diabetes, we found that HNF1A dysfunction leads to insulin-insufficient diabetes reminiscent of T1D by impacting the regulatory processes critical for glucose-stimulated insulin secretion and suggest a rationale for a therapeutic alternative to current treatment.

miR-7a is regulated in beta-cell dysfunction and couples early and late stages of pancreatic beta-cell differentiation to insulin secretion

Project description:Transcriptional and posttranscriptional regulatory networks play a crucial role in the maintenance and adaptation of pancreatic beta-cell function. In this study we show that the levels of the prototypic neuroendocrine miRNA-7 are regulated in islets of obese, diabetic and aged mouse models. Using gain- and loss-of-function models we demonstrate that miR-7 regulates crucial members of the endocrine pancreatic transcriptional network controlling differentiation and insulin synthesis. Importantly, it also directly regulates key proteins in the insulin granule secretory machinery. These results reveal an interconnecting miR-7 genomic circuit that influences beta-cell differentiation, insulin synthesis and release and define a role for miR-7 as an endocrine checkpoint to stabilize beta-cell function during metabolic stress. These findings have implications for miR-7 inhibitors as potential therapies for type 2 diabetes and neurodegenerative diseases. Either miR-7a2 or miR-7b were over-expressed in MIN6 cells using an adenoviral vector. The miR-7a infection was performed in duplicates. In addition, a GFP over-expression in MIN6 using the same viral vector served as control. We also explored the consequence of miR-7a2 deletion in pancreatic beta-cells by generating a beta-cells specific miR-7a2 knock-out using the Lox/Cre system in a C57BL/6 background. We profiled gene expression in mutant and wild-type (control) islets.

Myc is required for adaptive beta cell replication in young mice but is not sufficient in one year old mice fed with a high fat

Project description:Purpose: To test the role of Myc in adaptive expansion of beta cells Methods: Islets were isolated from 8 week old or one year old male C57BL6 mice fed a standard or high fat diet. RNAseq and DNA mehtylseq was performed. Results: RNAseq showed that Myc target genes are upregulted in young, but not old mice after a short term high fat diet. DNA methylation analysis shows hypomethylation of cell cycle genes that are Myc targets in islets from young mice fed with a HFD. Importantly, DNA hypomethylation of Myc response elements does not occur in islets from one-year-old mice fed short-term HFD Conclusions: We conclude that Myc is required for metabolic stress-mediated beta cell expansion in young mice, but with aging Myc upregulation in beta cells is not sufficient to induce beta cell replication by, at least partly, an epigenetically-mediated resistance to Myc action.

IGF2R-initiated Proton Rechanneling Dictates an Anti-inflammatory Property in Macrophages

Project description:Metabolic trait in macrophages can be rewired by insulin like growth factor 2 (IGF2), however, how IGF2 modulates the cellular dynamics and functionality remains unclear. We demonstrate here that IGF2 exhibits dual and opposing roles in controlling inflammatory phenotypes in macrophages via regulating glucose metabolism. Such difference relies on the dominant activation of IGF2R by low dose IGF2 and IGF1R by high dose IGF2. IGF2R activation leads to proton rechanneling to the mitochondrial intermembrane space, and enables sustained oxidative phosphorylation. Mechanistically,low dose IGF2 induces nucleus translocation of IGF2R that promotes Dnmt3a-mediated DNA methylation via activating GSK3β, and subsequently impairs the expression ofvacuolar-type H+-ATPase (v-ATPase).This sequestrated assembly of v-ATPase inhibits the channeling of protons to lysosomes and leads to their rechanneling to mitochondria. An IGF2R-specific IGF2 mutant only induces anti-inflammatory response and inhibits colitis progression. Altogether, our findings highlight a novel role of IGF2R activation in dictating anti-inflammatory macrophages.

Project description:This dataset consists of single-cell RNA-seq (10X) data from disperesed pancreatic islets of healthy and STZ induced diabetic mice. STZ (Sigma) was injected intraperitoneally in 8-week old male C57BLJ/6 mice at 50 mg/kg for five consecutive days. Islets were isolated from healthy mice and STZ diabetic mice after 100 days of either vehicle or drug treatment.
