Uncovering the potential molecular mechanism of liraglutide to alleviate the effects of high glucose on myoblasts based on high-throughput transcriptome sequencing technique

ABSTRACT: Myoblasts play an important role in muscle growth and repair, but the high glucose environment severely affects the function of myoblasts. The purpose of this study is to explore the potential molecular mechanism of liraglutide in alleviating the effects of high glucose environments on myoblasts.High-throughput transcriptome sequencing technique was used to sequence C2C12 myoblast from different treated groups.

RNA-Seq analysis on the liver of mice treated with high-fat diet and liraglutide

Project description:GLP-1 based diabetes drugs are effective to reduce hepatic lipid accumulation beyong glycemic control. As GLP-1 receptor (GLP-1R) is not expressed in hepatocytes, the mechanism behind the beneficial effects of GLP-1 based drugs on the liver remained elusive. Several studies have shown that the expression of hepatic fibroblast growth factor 21 (FGF21) can be stimulated by GLP-1-based drugs. Therefore, the present study aimed to assess such stimulation in mice and in mouse primary hepatocytes, determine whether hepatic FGF21 mediates functions of the GLP-1R agonist liraglutide, and to explore the potential mechanisms of liraglutide regulating the expression of FGF21. In high-fat diet induced obese mice, we observed hepatic and plasma FGF21 elevation, improved glucose disposal, and reduced plasma triglyceride levels by liraglutide treatment. Moreover, RNA-sequencing on the liver suggested that Fgf21 was the most upregulated gene after liraglutide treatment. In liver-specific FGF21 knock-out mice on high-fat and high-fructose diet, the body-weight gain attenuation and lipid homeostatic effects of liraglutide was lost or significantly reduced. These studies implied that hepatic FGF21 was the critical mediator of liraglutide-induced metabolic improvement.

Liraglutide inhibits osteoclastogenesis and improves bone loss by down-regulating Trem2 in female type 1 diabetic mice: findings from transcriptomics

Project description:Aims:We aimed to explore possible mechanisms of bone loss in T1D and clarify whether liraglutide have effects on bone quality of T1D mice using transcriptomics, . Methods:Female streptozotocin-induced diabetic C57BL/6J mice were randomly divided into 4 groups and received the following treatments daily for 8 weeks: saline as controls; insulin; liraglutide; liraglutide combined with insulin. These groups were also compared to non-STZ-treated normal glucose tolerance (NGT) group. Trunk blood and bone tissues were collected for analysis. Three tibia from each of the NGT, saline treated and liraglutide treated group were randomly selected for transcriptomics. Results:Compared with NGT mice,saline treated T1D mice manifested markedly hyperglycemia and weight loss, and micro-CT revealed significantly lower bone mineral density(BMD) and deficient microarchitectures in tibias.8 weeks’ treatment with liraglutide alone or combined with insulin rescued the decreased BMD and partly corrected compromised trabecular microarchitectures. Transcriptomics analysis showed there were 789 differentially expressed genes mainly mapped to osteoclastogenesis and inflammation pathways.RT-qPCR verified gene expression of Trem2, Nfatc1, Trap and Ctsk were significantly increased in tibia of T1D compared to that in NGT group. Liraglutide treatment alone or combined with insulin could effectively suppress osteoclastogenesis by down-regulating gene expression of Trem2, Nfatc1, Ctsk and Trap.

Gene expression profiling of pancreatic cancer cell conditioned myoblasts.

Project description:We verified whether conditioned media (CM) from pancreatic cancer cell lines (MIAPaCa2, CAPAN-1, PANC-1, BxPC3) alter glucose metabolism and gene expression profiles (microarray experiment with a platform of 5000 skeletal muscle cDNA) in mice myoblasts. Myoblasts were incubated with control or pancreatic cancer CM for 14 and 24 hours. Lactate significantly increased in CM compared with non-conditioned myoblasts. No variations in expression levels of the main genes involved in glycolysis were found in CM myoblasts. Propionyl coenzyme A carboxylase and isocitrate dehydrogenase 3 beta genes, which encode enzymes of the tricarboxylic acid cycle, were overexpressed, while IGFIIR and VAMP5 genes were underexpressed in CM myoblasts. PAFAH1B1 and BCL-2 genes (intracellular signal transduction) and the serine protease cathepsin G (proteolysis), were overexpressed in CM myoblasts. Tyrosine accumulation in CM myoblasts suggested that proteolysis overcomes protein synthesis. Sorcin, actin alpha, troponin T1, and filamin A were underexpressed in CM myoblasts. Our findings demonstrate that pancreatic cancer cell conditioned media enhanced lactate production and induced proteolysis, possibly by altering expression levels of a large number of genes, not only those involved in protein biosynthesis and degradation or glucose metabolism, but also those involved in the tricarboxylic acid cycle and in vesicle traffic.
