The role of nucleosome dynamics during the monocyte to macrophage transition

ABSTRACT: We used MNase Transcription Start Site Sequence Capture (mTSS-seq) to map nucleosome distribution and sensitivity to MNase digestion over pol II promoters in THP-1 monocytes during phorbol 12-myristate 13-acetate (PMA) induced differentiation. We found that differentially expressed macrophage genes have stronger nucleosome positioning and greater occupancy prior to differentiation suggesting that these genes are poised for future expression. We found dynamic alterations in nucleosome sensitivity during early differentiation and the establishment of a new sensitivity profile in macrophages. We further observed that alterations in sensitivity corresponded to gains and losses of regulatory factor footprints within the promoter region.
