Transcription profiling of human post-mortem brain tissue (orbitofrontal cortex) from patients with bipolar disorder and matched healthy controls

Project description:Bipolar affective disorder is a severe psychiatric disorder with a strong genetic component but unknown pathophysiology. We used microarray technology (Affymetrix HG-U133A GeneChips) to determine the expression of approximately 22 000 mRNA transcripts in post-mortem brain tissue (orbitofrontal cortex) from patients with bipolar disorder and matched healthy controls. Orbitofrontal cortex tissue from a cohort of 30 subjects was investigated and the final analysis included 10 bipolar and 11 control subjects. Differences between disease and control groups were identified using a rigorous statistical analysis with correction for confounding variables and multiple testing.

Cell-type-specific ageing effects in the human OFC and implication for psychiatric disease

Project description:We evaluated biological aging using two epigenetic clocks (Horvath’s multi-tissue clock and CorticalClock (Shireby et al.)Brain 2020) calculated from DNA methylation measured in human post-mortem brain (orbitofrontal cortex; BA11) in a trans-diagnostic psychiatric sample including healthy controls. Epigenome-wide DNA methylation profiling was performed with the Illumina Infinium MethylationEPIC BeadChip (version 1.0; Illumina, San Diego, CA, USA) according to manufacturer’s guidelines. The total cohort consisted of 91 individuals. Five individuals were run in duplicate (as internal control). Individuals were either diagnosed a psychiatric disease (1) or psychiatrically healthy (0). Independent of the epigenetic clock used, we did not find evidence for accelerated epigenetic aging in individuals diagnosed with a psychiatric disorder.

QUANTITATIVE SUBCELLULAR PROTEOMICS OF THE ORBITOFRONTAL CORTEX OF SCHIZOPHRENIA PATIENTS

Project description:We analyzed OFC post-mortem brain tissue from patients classified as residual schizophrenia according to the DMS-IV manual. In this classification, the patients do not manifest prominent positive psychotic symptoms but prevail characteristic disturbance of negative symptoms. Through the comparison of patient´s samples against a pool of control subjects without any history of psychiatric illness we generated an organellar proteome map from different subcellular fractions such as mitochondria (MIT), crude nuclear fraction (NUC) and cytoplasm (CYT) with the aim to monitor the subproteome changes using iTRAQ labelling for relative quantification and Selected Reaction Monitoring (SRM) for targeted quantification.

Profiling Neuronal Methylome and Hydroxymethylome of Opioid Use Disorder in the Human Orbitofrontal Cortex

Project description:Opioid use disorder (OUD) is influenced by genetic and environmental factors. While recent research suggests epigenetic disturbances in OUD, this is mostly limited to DNA methylation (5mC). DNA hydroxymethylation (5hmC) has been widely understudied. We conducted a multi-omics profiling of OUD in a male cohort, integrating neuronal-specific 5mC and 5hmC as well as gene expression profiles from human postmortem orbitofrontal cortex (OUD=12; non-OUD=26). Single locus methylomic analysis and co-methylation analysis showed a higher number of OUD-associated genes and gene networks for 5hmC compared to 5mC; these were enriched for GPCR, Wnt, neurogenesis, and opioid signaling. 5hmC marks also showed a higher correlation with gene expression patterns and enriched for GWAS of psychiatric traits. Drug interaction analysis revealed interactions with opioid-related drugs, some used as OUD treatments. Our multi-omics findings suggest an important role of 5hmC and reveal loci epigenetically dysregulated in OFC neurons of individuals with OUD.
