Phase separation of N-myc differentially modulates the transcriptom

ABSTRACT: SPARK-ON tag does not perturb the core transcriptional function of n-Myc.~2260 DEGs with ≥ 1.5-fold change in transcript level, including ~1270~6900 DEGs with ≥ 1.5-fold change in transcript level, including ~3100 up-regulated. By compare n-Myc condensed phase and dilute phase, 99 DEGs that were differentially expressed with ≥ 1.5-fold change. in transcript levels, including 88 up-regulated genes and 11 down-regulated genes and ~3800 down-regulated genes.

Project description:Transcriptional profiling of Drosophila melanogaster larval testes with and without the wMel strain of Wolbachia and found that 296 genes had at least a 1.5 fold change [q-value (%)<5%] in transcript levels, with 167 genes up-regulated and 129 genes down-regulated when comparing Wolbachia-infected flies to uninfected ones. Differential expression of genes related to metabolism, immunity, reproduction and other functions were observed.

Bovine blood gamma/delta T cell stimulation study

Project description:Blood gamma/delta T cells were purfiied from holstein calves less than six months old. Blood from 4 individual calves were pooled and gamma/delta T cells were purified by immunomagnetic separation, rested in RPMI medium for 12h, and underwent ConA & hIL-2 stimulation. Total RNAs were extracted by using Trizol reagents. 15 ug of total RNA was used for cDNA synthesis and microarray hybridization. Table 1: regulated genes These genes were identified, in GeneSpring software, via filtering of genes with relative expression ratio of 1.5 and over or 0.667 or below, equivalent to up- and down-regulation by fold change 1.5 or more, in both experiments. Table 2: down-regulated genes These genes were identified, in GeneSpring software, via filtering of genes with relative expression ratio of 0.667 or below, equivalent to down-regulation by fold change 1.5 or more, in both experiments. The avergae fold change was then converted to negative value. Gene annotation was performed by using GeneSpring's "Build Simplified Ontology" constructor. Transcriptome analysis using RNA sequencing conducted for wild type (Col-0) and a stable transgenic line with ectopic expression of AT3G11290 (35S:HIN1) under control conditions and after 96 hours at moderate low water potential stress (-0.7 MPa)

Project description:RNA sequencing was used to identify alternative splicing events and differentially expressed genes (DEG) in 35S:HIN1 stable transgenic line compared to Col-0 wild type. Two treatments were analyzed for each genotype: unstressed control and a 96 hours moderate severity low water potential (-0.7 MPa) treatment. RNA sequencing analysis found that overexpression of HIN1 in unstressed plants was sufficient to duplicate the effects of stress on nearly 40% of the genes where intron retention was normally suppressed by stress in wild type. Differentially expressed genes (DEGs) were also analyzed (P < 0.05 and fold change ≥ 1.5 used as cut off values for DEGs). Differentially expressed genes in 35S:HIN1has limited overlap of DEGs between 35S:HIN1and wild type. This indicated that HIN1 plays a lesser role in stress-regulated gene expression than in splicing regulation.
