Project description:Autophagy, involved in protein degradation and amino acid recycling, plays a key role in plant development and stress responses. However, the relationship between autophagy and phytohormones is unclear. We used diverse methods, including CRISPR/Cas9, UPLC-MS/MS, chromatin immunoprecipitation, electrophoretic mobility shift assays, and dual-luciferase assays to explore the molecular mechanism of strigolactones in regulating autophagy and the degradation of ubiquitinated proteins under cold stress in tomato. We show that cold stress induced the accumulation of ubiquitinated proteins. Mutants deficient in strigolactone biosynthesis were more sensitive to cold stress with a higher accumulation of ubiquitinated proteins, while treatment with the synthetic strigolactone analog GR245DS enhanced cold tolerance in tomato, with a higher accumulation of autophagosomes and transcripts of autophagy-related genes (ATGs), and a lower accumulation of ubiquitinated proteins. Meanwhile, cold stress induced ELONGATED HYPOCOTYL 5 (HY5) accumulation, which was triggered by strigolactones. HY5 further trans-activated ATG18a promoter, resulting in autophagy formation. Mutation of ATG18a compromised strigolactone-induced cold tolerance, followed by a decreased formation of autophagosomes and an increased accumulation of ubiquitinated proteins. These findings reveal that strigolactones positively regulate autophagy in an HY5-dependent manner and promote the degradation of ubiquitinated proteins under cold conditions in tomato.

Project description:Both jasmonic acid (JA) and melatonin (MT) have been demonstrated to play positive roles in cold tolerance, however, whether and how they crosstalk in the cold responses in plants remain elusive. Here, we report that JA and MT act synergistically in the cold tolerance in tomato plants (Solanum lycopersicum). It was found that JA and MT were both substantially accumulated in response to cold stress and foliar applications of methyl jasmonate (MeJA) and MT promoted cold tolerance as evidenced by increased Fv/Fm, decreased relative electrolyte leakage (EL) and declined H2O2 accumulation in tomato plants. Inhibition of MT biosynthesis attenuated MeJA-induced cold tolerance, while inhibition of JA biosynthesis reduced MT accumulation in tomato plants under cold conditions. Furthermore, qRT-PCR analysis showed that the expressions of two MT biosynthetic genes, SlSNAT and SlAMST, were strongly induced by MeJA, whereas suppression of SlMYC2, a master JA signaling regulator, abated the expressions of SlSNAT and SlAMST under cold stress. Additionally, suppression of SlMYC2 reduced MT accumulation, decreased Fv/Fm and increased EL in cold-stressed tomato plants. Interestingly, exogenous MT promoted JA accumulation, while inhibition of MT biosynthesis significantly reduced JA accumulation in tomato plants under the cold condition. Taken together, these results suggest that JA and MT act cooperatively in cold tolerance and form a positive feedback loop, amplifying the cold responses of tomato plants. Our findings might be translated into the development of cold-resistant tomato cultivars by genetically manipulating JA and MT pathways.

Project description:The transcriptome of leaves from two Anthurium cultivars, Oregon (Elegang, E, cold tolerant) and Fantasy love (Menghuan, MH, cold sensitive) under cold stress were sequenced by Illumina Novaseq™ 6000. Sequencing generated a total of 129.44 Gb of raw reads and an average of 7.19 Gb of reads for each sample. The analysis showed differences of transcriptomes between the control and the cold treatment samples. We identified numerous differentially expressed genes that exhibited distinct expression patterns. These genes have known or potential roles inAnthurium cold tolerance.Therefore, we are appealing candidates for further investigation of the gene expression and associated regulatory mechanisms related to the cold stress response of A. andraeanum.

Project description:Two small RNA libraries were generated from micropropagated â??Muscat Hamburgâ?? (Vitis vinifera) plantlets under normal and low temperatures (4 °C). A total of 163 known miRNAs and 299 putative novel miRNAs were detected from two small RNA libraries by Solexa sequencing. Forty-four cold-inducible miRNAs were identified through differentially expressed miRNAs (DEMs) analysis; among which, 13 belonged to upregulated DEMs while 31 belonged downregulated DEMs. This study indicated that a diverse set of miRNAs in V. vinifera are cold-inducible and may play an important role in cold stress response. Examination of small RNA populations in grapevine under cold treatment and none cold treatment.

Project description:The cold acclimation process is regulated by many factors like ambient temperature, day length, light intensity, or hormonal status. Experiments with plants grown under different light-quality conditions indicate that the plant response to cold is also a light-quality-dependent process. Here, the role of light quality in the cold response was studied in one-month-old Arabidopsis thaliana (Col‐0) plants exposed for one week to 4 °C at short‐day conditions under white (100 and 20 μmol m‐2s‐1), blue or red (20 μmol m‐2s‐1) light conditions. An upregulated expression of CBF1, an inhibition of photosynthesis, and an increase in membrane damage showed that blue light enhanced the effect of low temperature. Interestingly, cold-treated plants under blue and red light showed only limited freezing tolerance compared to white light cold-treated plants. Next, the specificity of the light quality signal in cold response was evaluated in Arabidopsis accessions originating from different and contrasting latitudes. In all but one Arabidopsis accessions, blue light increased the effect of cold on photosynthetic parameters and electrolyte leakage. This effect was not found for Ws-0, which lacks functional CRY2 protein, indicating its role in the cold response. Proteomics data confirmed significant differences between red and blue light treated plants at low temperature and showed that the cold response is highly accession specific. In general, blue light increased mainly the cold-stress related proteins and red light induced higher expression of chloroplast-related proteins, which correlated with higher photosynthetic parameters in red light cold-treated plants. Altogether, our data suggest that light modulates two distinct mechanisms during the cold treatment - red light driven cell function maintaining program and blue light activated specific cold response. The importance of mutual complementarity of these mechanisms was demonstrated by significantly higher freezing tolerance of cold-treated plants under white light.

Project description:To understand mRNA expression pattern during cold acclimation and deacclimation, transcriptional profiling of cold acclimation and deacclimation-treated plants were analyzed using Agilent-015059 Arabidopsis 3 Oligo Microarray 4x44K G2519F. Arabidopsis Col-0 were grown on MS plate for 2 weeks (16 hours light / 8 hours dark). Two week-old Arabidopsis samples (NA, non acclimation) were treated with cold (2 M-BM-:C) for 7 days (CA7d) under 12h/12h light/dark conditions. Deacclimation-treated samples (DA6h, DA12h, DA24h) were grown at normal growth temperature under long day conditions after cold treatment for 7 days. Then total RNA was prepared from the whole seedling and used for the microarray hybridization. Three replicative hybridization experiments for each array were carried out using the independent biological samples.
