Project description:JoMa1 cells are pluripotent precursor cells, derived from the neural crest of mice transgenic for tamoxifen-inducible c-Myc. Following transfection with a cDNA encoding for MYCN, cells become immortlized even in the absence of tamoxifen. The expression profile of JoMa1 cells was compared to cell lines derived from tumors induced by JoMa1 transfected with MYCN.

Project description:Neuroblastoma is a common childhood malignant tumor of neural crest origin with remarkable heterogeneity in outcomes. Amplification of the oncogene MYCN is strongly associated with highly malignant behaviour and poor prognosis. Here, we used human ESC-derived neural crest model to recapitulate the initiation of MYCN-driven neuroblastoma and to identify MYCN downstream effectors. Our results show that induced deregulation of MYCN in human neural crest progenitor cells is sufficient to induce tumors that recapitulate the pathological and molecular features of human MYCN-amplified neuroblastoma(MNA-NB). By using this platform, we are able to identify a group of 28 genes that are associated with MYCN expression level only in MNA-NB.

Project description:Osteosarcoma, derived from mesenchymal stem cells or osteoblasts, is the most common malignant bone tumor and the highly metastatic malignant phenotypes often with poor prognosis are related to modulation of TP53- and cell cycle-related pathways. MYC, which regulates the transcription of cell-cycle modulating genes, e.g. CCNE1, is used as a representative prognostic marker. MYCN, a member of MYC oncoprotein family, is highly expressed in a subset of osteosarcoma, however, its roles in osteosarcoma have not been elucidated extensively. Here, we attempted to make an in vitro tumorigenesis model by using hiPSCs derived neural crest cells, which is a precursor cell of mesenchymal stem cells, by MYCN overexpression in the background of heterozygousTP53 hotspot mutation (c.733G>A; p.G245S). MYCN-expressing transformed clones with TP53 mutation were isolated by soft agar colony formation, and injected subcutaneously and into periadrenal adipose tissue of immune-deficient mice, resulted in development of chondroblastic osteosarcoma. MYCN suppression decreased proliferation in MYCN-induced osteosarcoma cells, suggesting that MYCN seems to be one of the targets of osteosarcoma treatment. Further, comprehensive analysis of gene expression and exome sequencing of the MYCN-induced transformed clones indicated molecular context of osteosarcoma-specific feature such as activation of TGF-β signaling and DNA copy number amplification of GLI1. The model of MYCN-expressing chondroblastic osteosarcoma was developed from hiPSC-based neural crest cells, allowing us to provide useful tool for development of new tumor models using hiPSC-derived progenitor cells with gene modification and in vitro transformation.

Project description:Neural crest cells are embryonic progenitors that generate numerous cell types in vertebrates. With single cell analysis, we show that mouse trunk neural crest cells become biased toward neuronal lineages when they delaminate from the neural tube, whereas cranial neural crest cells acquire ectomesenchyme potential dependent on activation of the transcription factor Twist1. The choices that neural crest cells make to become sensory, glial, autonomic, or mesenchymal cells can be formalized as a series of sequential binary decisions. Each branch of the decision tree involves initial co-activation of bipotential properties followed by gradual shifts towards commitment. Competing fate programs are co-activated before cells acquire fate-specific phenotypic traits. Determination of a specific fate is achieved by increased synchronization of relevant programs and concurrent repression of competing fate programs.

Project description:Brain pericytes are one of the critical cell types that regulate endothelial barrier function and activity, thus ensuring adequate blood flow to the brain. The genetic pathways guiding undifferentiated cells into mature pericytes are not well understood. We show here that pericyte precursor populations from both neural crest and head mesoderm of zebrafish express the transcription factor nkx3.1 develop into brain pericytes. We identify the gene signature of these precursors and show that an nkx3.1-, foxf2a-, and cxcl12b-expressing pericyte precursor population is present around the basilar artery prior to artery formation and pericyte recruitment. The precursors later spread throughout the brain and differentiate to express canonical pericyte markers. Cxcl12b-Cxcr4 signaling is required for pericyte attachment and differentiation. Further, both nkx3.1 and cxcl12b are necessary and sufficient in regulating pericyte number as loss inhibits and gain increases pericyte number. Through genetic experiments, we have defined a precursor population for brain pericytes and identified genes critical for their differentiation. Project description:We report that cancer associated protein HIF-2a is expressed in trunk neural crest neuroblastoma precursor cells in the developing embryo in three different species; human, mouse and avian. Dysregulation of HIF-2a leads to alterations in embryonic development, and neural crest cell migration, proliferation and self-renewal capacity. With RNAsequencing we report that alterations of HIF-2a expression affects the global transcriptome and that gene ontology enrich for the same processes observed in vivo.

Project description:Ectoderm-derived neural crest is a transient structure arising during early embryogenesis in vertebrates. Neural crest consists of four derivatives based on their anterior- to posterior location along the body axis; cranial, vagal, trunk and sacral, respectively. We recently showed that trunk neural crest-specific gene MOXD1 functions as a tumor suppressor in trunk neural crest-derived childhood cancer form neuroblastoma and is essential for proper development of healthy adrenal glands. However, the role of MOXD1 during early embryogenesis is not known. Here, we conditionally knocked out MOXD1 in trunk neural crest cells before they become lineage-committed, using a CRISPR/Cas9 approach in chick embryos. Assessment of embryo growth showed that knockout of MOXD1 delayed development with knockout embryos being smaller. RNA sequencing of trunk-derived neural crest cells from control and knockout embryos showed enrichment of genes connected to gland development, copper ion metabolism and neuroblastoma progression. In conclusion, MOXD1 is important during early and prolonged embryonic development with effects on gland formation, possibly mediated via its role in copper metabolism.
