Effects of estrogen and progesterone on glycosyltransferase expression in mice

ABSTRACT: This laboratory studies the structural basis of carbohydrate function in terms of adhesion and other biological processes. The Fisher lab is testing the hypothesis that the mucin-coated oral and uterine cavities present similar carbohydrate receptors that specify the bacterial ecology of both regions and the repertoire of these oligosaccharide species is hormonally regulated. This theory also suggests that certain individuals express carbohydrate receptors that make them susceptible to both periodontal disease and preterm labor.

Project description:This laboratory studies the structural basis of carbohydrate function in terms of adhesion and other biological processes. The Fisher lab is testing the hypothesis that the mucin-coated oral and uterine cavities present similar carbohydrate receptors that specify the bacterial ecology of both regions and the repertoire of these oligosaccharide species is hormonally regulated. This theory also suggests that certain individuals express carbohydrate receptors that make them susceptible to both periodontal disease and preterm labor. Experimental procedure: 40 mature female mice were ovariectomize . The mice were allowed to rest for two weeks to eliminate any remaining endogenous estrogen (E2) and progesterone (P4). Then the mice were separated into four groups (10 each) that received the treatment indicated. Group 1: ovariectomized mice with no supplement, only sesame oil vehicle. Group 2: ovariectomized mice with only P4 supplement (2 mg/day/mouse). Group 3: ovariectomized mice with E2 supplement (100 ng/day/mouse). Group 4: ovariectomized mice with both P4 (2 mg/day/mouse) and E2 (100 ng/day/mouse) supplement. The treatment lasted for 4 days; the steroids were dissolved in sesame oil and injected subcutaneously. After treatment, salivary glands (parotid, submandibular, and sublingual gland), submandibular lymph nodes and uterine horns with cervix were collected. Total RNA was extracted from these samples using Trizol.

Cranberry proanthocyanidins and dietary oligosaccharides synergistically modulate Lactobacillus plantarum physiology

Project description:Plant-based foods contain bioactive compounds such as polyphenols that resist digestion and potentially benefit the host through interactions with their gut microbiome. Based on previous observations, we hypothesized thatprobiotic Lactobacillus plantarum interact with cranberry polyphenols and dietary oligosaccharides to synergistically impact its physiology. In this study, L. plantarum ATCC BAA-793 was grown on dietary oligosaccharides including cranberry xyloglucans, fructooligosaccharides, and human milk oligosaccharidesin conjunction with proanthocyanidins (PACs) extracted from cranberry. As a result, L. plantarum exhibits a differential physiological response to cranberry PACs dependent on the carbohydrate source and polyphenol fraction introduced. Of two extracts evaluated, the PAC1 fraction increased growth regardless of oligosaccharide whereas PAC2 positively modulates growth during xyloglucan metabolism. Interestingly, PAC1 enables ATCC BAA-793 to utilize fructooligosaccharides efficiently as it is unable to ferment this substrate ordinarily. Relative to glucose, oligosaccharide metabolism increases the ratio of secreted acetic acid to lactic acid. The PAC2 fraction differentially increases this ratio during cranberry xyloglucan fermentation compared with PAC1. RNA-seq transcriptomics link expression of putative polyphenol degradation genes, polyphenol degradation profiles, and physiological phenotypes.

Project description:alpha-dystroglycan is a component of the dystrophin associated glycoprotein complex that binds components of the extracellular matrix (including laminin, perlecan and neurexin) via its carbohydrate groups. Recently, a group of muscular dystrophies have been identified due to the aberrant glycosylation of this molecule (dystroglycanopathies). The overexpression of the glycosyltransferase LARGE in several cell lines results in alpha-dystroglycan hyperglycosylation and enhanced ligand binding. The glycan structures induced by LARGE overexpression are unknown. We have injected into mouse legs two LARGE constructs, one wild type and the second in which a DxD motif has been mutated to NNN as a control. We aim to identify genes unregulated during LARGE induced hyperglycosylation in order to gain insight into the enzymes involved in this process and ultimatley the nature of the glycans responsible for ligand binding. RNA preparations from two LARGE constructs, were sent to Microarray Core (E). One wild type and the second in which a DxD motif has been mutated to NNN as a control. The aim is to identify genes unregulated during LARGE induced hyperglycosylation in order to gain insight into the enzymes involved in this process and ultimatley the nature of the glycans responsible for ligand binding. The RNA was amplified, labeled, and hybridized to teh GLYCOv3 microarrays.

Metabolism of sialic acid by Bifidobacterium breve UCC2003

Project description:Bifidobacteria constitute a specific group of commensal bacteria which inhabit the gastrointestinal tract of humans and other mammals. Bifidobacterium breve UCC2003 has previously been shown to utilise several plant-derived carbohydrates that include cellodextrins, starch and galactan. In the current study, we investigate the ability of this strain to utilise the mucin- and human milk oligosaccharide (HMO)-derived carbohydrate, sialic acid. Using a combination of transcriptomic and functional genomic approaches, we identified a gene cluster dedicated to the uptake and metabolism of sialic acid. Furthermore, we demonstrate that B. breve UCC2003 can cross feed on sialic acid derived from the metabolism of 3’ sialyllactose, a HMO, by Bifidobacterium bifidum PRL2010.
