miRNA profiling of human saphenous vein pericyte exosomes

Project description:Human saphenous vein perciytes (hSVPs) were proved increase angiogenesis in mouse myocardial infarction model by producing and releasing miR-132, which has proangiogenic, prosurvival, and antifibrotic activity. To investigate whether their exosomes have a role in their proangiogenic activity, we examined exosomes secreted by these cells. Nanoparticle tracking analysis (NTA) results indicated the presence of exosome size particles in the preparations. Experiments performed with hypoxic (1% O2) human umbilical vein endothelial cells (HUVECs) confirmed that the pericyte exosomes had proangiogenic and anti-apoptotic features. To understand the miRNA cargo of these exosomes that is important in their unique properties, we performed a miRNA array for profiling of 752 miRNAs.

Exosomes secreted by the CCND2OECMs promote proliferation and hypoxia tolerance in cultured CMs

Project description:CCND2OEExos (exosomes from CCND2-overexpressing hiPSC-derived cardiomyocytes (CCND2OECMs)) were found to be able to promote proliferation of not only cardiomyocytes, but also human umbilical vein endothelial cells (HUVEC) and human vascular smooth muscle cells (HVSMC) in vitro, as indicated by the large number of ki67+ and PH3+ HUVECs and SMCs in the CCND2OEExos-treated group, relative to those in the vehicle only- or CCND2WTExos-treated groups. A comparative bulk sequence analysis of miRNAs from CCND2WTExos and CCND2OEExos (exosomes from wild type hiPSC-derived cardiomyocytes) was performed to identify some underlying mechanisms about proliferation induction.

Endothelial cell-secreted bone targeting exosomes promote angiogenesis coupling with osteogenesis via the PERK-ATF4-CRELD2 pathway

Project description:The role of endoplasmic reticulum (ER) stress in bone metabolism and the management of associated diseases has garnered significant interest. However, its role in regulating bone homeostasis and skeletal development remains largely unclear. Osteoblast development and bone formation are enhanced by a particular subtype of CD31hi endomucinhi (CD31hiEMCNhi) endothelium. However, it is still unclear how endothelial exosomes contribute to the production of CD31hiEMCNhi endothelium and bone formation. This research revealed that human umbilical vein endothelial cells (HUVECs)-exosomes (Exos) enhanced the formation of osteoblast and angiogenic effects in vitro. Furthermore, in mice treated with HUVECs-Exos, osteoblast production, and CD31hiEmcnhi vessels were significantly increased. The mechanism by which HUVECs-Exos CRELD2 improved angiogenesis coupling with osteogenesis involved triggering the PERK-ATF4-CRELD2 pathway's ER stress. As a result, HUVECs-Exos CRELD2 may be used as a potential bone metabolic disease nanodrug.

Comprehensive proteomic analysis of exosome mimetic vesicles and exosomes derived from human umbilical cord mesenchymal stem cells

Project description:Exosomes derived from mesenchymal stem cells (MSCs) have shown to have effective application prospects in the medical field, but exosome yield is very low. The production of exosome mimetic vesicles (EMVs) by continuous cell extrusion leads to more EMVs than exosomes, but whether the protein compositions of MSC-derived EMVs (MSC-EMVs) and exosomes (MSC-exosomes) are substantially different remains unknown. The purpose of this study was to conduct a comprehensive proteomic analysis of MSC-EMVs and MSC-exosomes and to simply explore the effects of exosomes and EMVs on wound healing ability. This study provides a theoretical basis for the application of EMVs and exosomes.In this study, EMVs from human umbilical cord MSCs (hUC MSCs) were isolated by continuous extrusion, and exosomes were identified after hUC MSC ultracentrifugation. A proteomic analysis was performed, and 2,315 proteins were identified. The effects of EMVs and exosomes on the proliferation, migration and angiogenesis of human umbilical vein endothelial cells (HUVECs) were evaluated by cell counting kit-8, scratch wound, Transwell and tubule formation assays. A mouse mode was used to evaluate the effects of EMVs and exosomes on wound healing . Bioinformatics analyses revealed that 1,669 proteins in both hUC MSC-EMVs and hUC MSC-exosomes play roles in retrograde vesicle-mediated transport and vesicle budding from the membrane. The 382 proteins unique to exosomes participate in extracellular matrix organization and extracellular structural organization, and the 264 proteins unique to EMVs target the cell membrane. EMVs and exosomes can promote wound healing and angiogenesis in mice and promote the proliferation, migration and angiogenesis of HUVECs.

Dynamics of intercellular and extracellular miRNAs in HUVECs co-cultured with K562 cells

Project description:MicroRNAs (miRNAs) are small (18-22 nucleotides), non-coding RNAs that suppress the translation of target mRNAs by binding to their 3` untranslated region, and regulate gene networks, helping to control cell function and phenotype. Recently, the existence of extracellular miRNAs enclosed in exosomes has raised the possibility that they play an important role in cell-cell communication. The goals of this study were to elucidate whether or not miRNAs secreted from neoplastic cells transfer into endothelial cells and works functionally active in the recipient cells. In the present study, we investigated the interaction of a leukemia cell line (K562) and human umbilical vein endothelial cells (HUVECs) using a non-contact co-culture system, and assess the expression profiles of intercellular and extracellular miRNAs in HUVECs and K562 cells using Taqman MicroRNA Array v2.0 (Applied Biosystems, Bedford, MA). We compared the miRNA expression profiles between 1) K562 cells or HUVECs; 2) intercellular and extracellular; 3) mono-culture medium and co-culture medium.
