Expression data of exosomal miRNAs extracted from plasma [Affymetrix miRNA 4.0]

ABSTRACT: Exosomal miRNAs have emerged as promising disease biomarkers. We aim to investigate the potential of exosomal miRNAs to assist with AIS clinical diagnosis. In the present research, we used the Affymetrix Genechip miRNA 4.0 Array to investgate the profiles of differentially expressed miRNAs (DEMs) in the exosomes of peripheral blood plasma. As a result, exosomal miRNA profiling yielded a total of 05 DEMs between the AIS (Acute Ischemic Stroke) patients and controls.

Project description:Tuberculosis (TB) is difficult to diagnose under complex clinical conditions. Exosomal miRNAs have emerged as promising disease biomarkers. We aim to investigate the potential of exosomal miRNAs to assist with TB clinical diagnosis. In the present research, we used the Affymetrix Genechip miRNA 4.0 Array to investgate the profiles of differentially expressed miRNAs (DEMs) in the exosomes of peripheral blood plasma. As a result, exosomal miRNA profiling yielded a total of 102 DEMs (98 with up-expression and 4 with down-expression) between the TB (pulmonary tuberculosis and tuberculosis meningitis) patients and controls.

Project description:Diffuse large B-cell lymphoma (DLBCL) is the most common subtype of non-Hodgkin Lymphoma with complex clinical symptoms. Currently, the diagnosis of DLBCL depends largely on pathologic analysis of biopsy tissue, which is an invasive procedure and often poses some risk to patients. Exosomal miRNAs have emerged as promising noninvasive biomarkers for detecting tumor or monitoring disease progress. We aim to investigate the potential of circulating exosomal miRNAs to assist with diagnosis of DLBCL.In the present research, we used the miRCURY LNA™ microRNA Array to investgate the profiles of differentially expressed miRNAs in the exosomes of peripheral blood serum. As a result, circulating exosomal miRNA profiling yielded a total of 332 differentially expressed miRNAs (157 with up-expression and 175 with down-expression) between the DLBCL patients and healthy controls.

Different expression of microRNAs in human sepsis

Project description:To obtain novel biomarkers that serve as prompt indicators of sepsis, we aimed to screen out the differentially expressed miRNAs (DEMs) contribute to sepsis susceptibility and analyze the correlation of DEMs with inflammatory response. This case-control study included 40 sepsis patients and 40 physical examination donors as a control group. RNA-seq technology and bioinformatics analysis were used to screen out the DEMs between sepsis patients and normal people. The expression of these DEMs were subsequently verified by qRT-PCR. There were 305 DEMs (p<0. 05, |Fold change| ≥2) between sepsis and normal people were identified and top eighteen upregulated or downregulated miRNAs were picked out for QRT-PCR verification. Among them, 11 DEMs were high expression while 7 DEMs were low expression. Collectively, miRNA-3663-3p may be a potentially powerful diagnostic and predictive biomarker for sepsis.

Project description:Variability of meningioma growth, even within benign subgroup, makes some remain unaltered, while others grow fast despite an appended radiotherapy. Biomarkers that differentiate between less and more aggressive meningiomas would therefore have a significant clinical benefit in prediction of their biological behaviour. The aim of the study was to identify new candidate miRNAs enabling better and proper prediction of meningioma recurrence. We used miRNA 4.0 array (Applied Biosystems, Foster City, CA, USA) to detect miRNA profiles in 44 primary meningioma patients (19 with subsequent recurrence) and 5 healthy controls. Moreover, miRNA profiles were analyzed in 15 secondary meningiomas.

Analysis and Verification of Exosomal miRNA Expression Profile of Serum in Patients with Acute Ischemic Stroke

Project description:To understand the role of exosome miRNAs in AIS disease and to explore the miRNAs and the potential mechanisms related to the occurrence and development of the disease. This research collected serum samples from AIS patients and healthy volunteers and extracted exosome miRNA. The exosome miRNA associated with AIS was screened by illumina high-throughput sequencing . The differentially expressed target genes related to miRNAs were analyzed using bioinformatics analysis including GO functional enrichment analysis, KEGG pathway enrichment analysis, and the screening of key genes via PPI diagram. These differentially expressed mRNAs were indicated an important role in a variety of biological processes and multiple pathways of AIS disease.

Our study identified 2509 DEGs, 34 DEMs, and 432 DELs in LC patients. miRNA-mRNA pairs, including 1 miRNA (hsa-miR-21-5p) and 5 targeted genes (RECK, TIMP3, EHD1, RASGRP1 and ERG), were figured out. We finally found the hub subnetwork (LINC00632/has-miR-21-5p/TIMP3) by combining lncRNA-miRNA pairs, miRNA-mRNA pairs and lncRNA-mRNA pairs.
