Project description:Global gene-expression profiles analysis of hESCs, patient-specific iPSCs, gene-corrected iPSCs, and patient fibroblast cells. Affymetrix GeneChip® Human Genome U133 plus 2.0 Array (Affymetrix, Inc.) was used for microarray hybridizations to examine the global gene expression of hES cells, piPS7 cells, ciPS5 cells and patient fibroblast cells. Briefly, total RNA was isolated using Trizol reagent (Invitrogen). cDNA was synthesized and amplified with WT cDNA Synthesis and Amplification Kit (Affymetrix, Inc.), labeled with biotin using WT Terminal Labeling Kit (Affymetrix, Inc.), hybridized to Affymetrix GeneChip® Human Genome U133 plus 2.0 Array and analyzed by GeneChip® Scanner 3000 7G3000 (Affymetrix, Inc.). All samples were prepared in three biological repeats. Data were analyzed using GCOS 1.4 software provided by Affymetrix.

Global gene expression profiles of mouse ESCs, disease-specific iPSCs, and gene-corrected iPSCs

Project description:The use of patient-derived induced pluripotent stem (iPS) cells as treatment for genetic diseases entails genetic repair or transfer of genetic information as a prerequisite. We have chosen the murine model of tyrosinemia type 1 (fumarylacetoacetate hydrolase deficiency; FAH(-/-) mice) as a paradigm for hereditary metabolic liver disorders and evaluated fibroblast-derived FAH(-/-)-iPS cell lines as targets for gene correction. By aggregating FAH(-/-)-iPS cells with tetraploid embryos, we obtained FAH-/--iPS cellâderived mice, which exhibited the phenotype of the founding FAH(-/-)-mice. We then rescued the diseased phenotype by lentiviral transduction of FAH-cDNA and performed embryo aggregation with these gene-corrected FAHgc-iPS cells to obtain viable healthy mice. Our results demonstrate that iPS cell technology is a valid approach to establish mouse disease models directly from somatic cells bearing genetic defects. Furthermore, established iPS cell lines can be genetically manipulated without loss of pluripotency for treatment of genetic diseases. 6 samples: 1 MEF, 1ESC, 4 iPSCs

Concurrent progress of reprogramming and gene-correction to overcome limitation of gene therapy of ALK2 mutant iPSCs

Project description:To ameliorate the cumbersome processes of reprogramming and subsequent gene-editing in vulnerable iPSCs, we have developed a greatly simplified one-step procedure, simultaneously introducing reprogramming and gene-editing components into human fibroblast cells. This not only serves to save time, labor, and costs, but opens up a new arena of research that is beyond the current application of gene-editing methodologies due to restrictive reprogramming concerns, inhibitory pluripotency maintenance requirements, and vulnerability of single-cell dissociated iPSCs. We generated iPSCs (mALK2-iPSCs) derived from Fibrodysplasia ossificans progressiva (FOP)-fibroblast cells carrying the ACVR1 p.R206H mutation and gene-corrected ALK2-iPSCs (cALK2-iPSCs). To identify gene-correction effects on global gene expression, gene expression profiling was measured in cALK2-iPSCs and mALK2-iPSCs, calibrated to WT-iPSCs with duplication.

Expression data of the iPSCs derived from foreskin fibroblast cells of normal person and KS patient

Project description:Klinefelter’s Syndrome (KS) is one of the common chromosome aneuploidy diseases in males with unexplained physiological mechanism. iPSCs, are similar to ESCs in terms of indefinitive self-renewal and pluripotency, provided an alternative choice for modeling disease to facilitate the disease research in vitro. We used microarray to detect the global reprogramming of KS and normal fibroblast cells to iPSCs. Also we used microarray to explore the possible molecular varieties between KS patient and normal person in the early development. Fibroblast cells from both normal person and KS patient were reprogrammed into iPSCs by ectopic expression of OCT4, SOX2, KLF4 and C-MYC. The expression profiles of normal and KS fibroblast cells, a line of normal iPSCs and two lines of KS iPSCs as well as a line of human ESCs were detected.
