ABSTRACT: Comparison of three populations of OECs derived from the ONL of rats. The three populations were cultured employing the same culture medium but were maintained in vitro for a different amount of time (number of population doublings or PDs). OEC Ep cells were maintained for less than one week in vitro (less than 3 PDs); OEC Lp were maintained for over a year (more than 50 PDs) and TEG3 OEC cell line has been serially passed in culture for long-periods of time after genetic immortalization. Transcription profiling of rat olfactory ensheathing cells (OEC) comparison of three populations

Project description:Comparison of three populations of OECs derived from the ONL of rats. The three populations were cultured employing the same culture medium but were maintained in vitro for a different amount of time (number of population doublings or PDs). OEC Ep cells were maintained for less than one week in vitro (less than 3 PDs); OEC Lp were maintained for over a year (more than 50 PDs) and TEG3 OEC cell line has been serially passed in culture for long-periods of time after genetic immortalization. Experiment Overall Design: RNA sample preparation and experimental design. Total RNA was isolated with TRIZOL (Invitrogen, Gibco-BRL) from primary OEC cultures (OEC Ep and Lp) and the OEC cell line (TEG3). Cultured cells were harvested at confluency with a rubber scrapper, resuspended in the TRIZOL reagent and processed following the manufacturersâ?? instructions. Two RNA pools were obtained from each OEC population (OEC Ep, OEC Lp and TEG3 cells). Each RNA pool was purified from three individual cultures (three 100mm dishes) and in the case of OEC Ep, each culture was derived from one rat. The total RNA was resuspended in DEPC (Sigma) water and stored at -70ÂºC prior to use. Total RNA concentration and purity were determined using Agilent2100 Bioanalizer. Experiment Overall Design: Microarray analysis. Total RNA pools were cleaned using Rneasy Kit from Qiagen. Biotinylated cRNA probes were generated from each RNA pool, fragmented, and applied to two different sets of Rat Genome Affymetrix GeneChip U34A Array. Affymetrix software (Microarray Suite version 5.0) was used to filter inaccurately represented probe sets.

EPC senescence

Project description:Transcription profiling of human umbilical cord blood derived Outgrowth Endothelial Cells (OECs) at early and late passages. Outgrowth endothelial cells were isolated from the mononuclear fraction of umbilical cord blood and cultured on collagen coated flasks. Once OEC colonies emerged they were expanded in culture to study cell growth kinetics. Total RNA was extracted from OECs at an early passage and OECs at a late passage which were becoming senescent . The purpose of this experiment was to were compared early and late passage OECs to determine how senescence affects the gene expression profile of OECs.

Single cell RNA-sequencing reveals five subtypes of olfactory bulb-derived olfactory ensheathing cells that secrete Reelin

Project description:Olfactory ensheathing cells (OECs) are the only glial cells that support the olfactory sensory neurons which undergo adult neurogenesis and continually project their axons to glomeruli in the olfactory bulbs. We used single cell RNA sequencing to study the gene expression programs of OECs and to determine the diversity of purified OECs previously shown to promote spinal cord injury repair. Our analyses revealed five subtypes of OECs, each expressing unique marker genes and pathways indicative of progenitor, axonal regeneration, migration, or microglia-like functions. As expected, we found substantial overlap of OEC genes with those of Schwann cells, but also with astrocytes, oligodendrocytes and microglia. We experimentally confirmed the classic marker genes of the OEC subtypes and provide evidence that Reelin and Connective Tissue Growth Factors are secreted by multiple OEC subtypes. Our results support that adult OECs are a hybrid glia including some with progenitor characteristics and that they likely carry out diverse functions related to injury repair and axonal regeneration.

Insights into olfactory ensheathing cell development from a laser-microdissection and transcriptome-profiling approach

Project description:Olfactory ensheathing cells (OECs) are neural crest-derived glia that ensheath bundles of olfactory axons from their peripheral origins in the olfactory epithelium to their central targets in the olfactory bulb. We took an unbiased laser microdissection and differential RNA-seq approach, validated by in situ hybridisation, to identify candidate molecular mechanisms underlying mouse OEC development and differences with the neural crest-derived Schwann cells developing on other peripheral nerves. We identified 25 novel markers for developing OECs in the olfactory mucosa and/or the olfactory nerve layer surrounding the olfactory bulb, of which 15 were OEC-specific, i.e., not expressed by Schwann cells. One pan-OEC-specific gene, Ptprz1, encodes a receptor-like tyrosine phosphatase that blocks oligodendrocyte differentiation. Mutant analysis suggests Ptprz1 may also act as a brake on OEC differentiation, and that its loss disrupts olfactory axon targeting. Overall, our results provide new insights into OEC development and the diversification of neural crest-derived glia.

Gene expression of LDS-OECs

Project description:The Loeys-Dietz syndrome (LDS) is an inherited connective tissue disorder caused by mutations in the transforming growth factor Î² (TGF-Î²) receptors TGFBR1 or TGFBR2. Most patients with LDS develop severe aortic aneurysms resulting in early need of surgical intervention. We investigated circulating outgrowth endothelial cells (OEC) from the peripheral blood of LDS to gain further insight into the pathophysiology of the disorder. We performed gene expression profiling using microarray analysis followed by quantitative PCR for verification of gene expression. OECs isolated from age- and sex-matched healthy donors served as reference control. 3 OEC clones from different LDS patients were compared to their respective age- and sex-matched OEC clone isolated from healthy donors.

Changes in gene expression of Imr90 cells with age and senescence under 3% and 20% oxygen growth conditions

Project description:Senescent is an irreversible form of cell cycle arrest initiated by damaged cell constituents and subsequent pro-oncogenic signaling. Replicative senescence in vitro can be considered a model for human aging. When fibroblasts are cultured under atmospheric oxygen conditions of 20%, typical of normal tissue culture procedure, fibroblasts generally reach their replicative capacity at 50-60 population doublings (PDs). When fibroblasts are cultured under normal physiological oxygen conditions of 3%, PDs increase about 30% relative to atmospheric levels. Hence while oxygen is a requirement for normal aerobic respiration, it can contribute to the total amount of oxidative stress to which cells are exposed to, leading to a long-term adverse effect in vitro. Inasmuch, cultures maintained under hyperoxic and hypoxic conditions provide a convenient model system for assessing the relationship between oxygen/oxidative stress and senescence. We used microarrays to profile the changes in global gene expression during aging and senescence of Imr90 cells under growth oxygen conditions of 3% and 20%. Imr90 cells at various population doubling timepoints (young, old, and senescent) grown separately under 3 and 20% oxygen growth conditions were selected for RNA extraction and hybridization on Affymetrix microarrays. Timepoints from cells grown under 3% and 20% oxygen conditions were age matched via population doublings to ensure accurate cross sample comparison.
