Adaptive evolution under modest thermal stress improves the thermotolerance and ethanol fermentability of Saccharomyces cerevisiae

Project description:Thermotolerance development of robust Saccharomyces cerevisiae is necessary to enhance enzyme activity of cellulase, lower cooling costs, and reduce cell harm from the bad-distributed heat transfer in large-scale fermentation. The process-based studies of adaptive evolution have been well documented, but it remains unknown for the underlying molecular mechanism of the improved thermotolerance and the facilitated ethanol fermentability derived from adaptive evolution. Here, a robust thermotolerant S. cerevisiae Z100 was obtained with significantly improved ethanol fermentability under the stress of high temperature (50 oC) after 91 days’ adaptive evolution. RNA sequencing showed that adaptive evolution and its derived thermotolerance contributed to the unique gene transcriptional landscapes of the evolved strain. An interesting phenomenon was that the gene transcriptional signals of carbon metabolism were strengthened not at 50 oC but at 30 oC in S. cerevisiae Z100, and thus suggested that the improved thermotolerance led to the enhanced ethanol fermentability at 30 oC. The deeply repressed gene transcriptional expression indicated ribosome would be another key thermotolerant mechanism for the evolved strain. This study would provide a robust thermotolerant S. cerevisiae for bioethanol production and an important clue for future synthetic biology to thermotolerance engineering of fermentation strains.

Project description:In Arabidposis thaliana, the msh1 recA3 double mutant shows an extensive mitochondrial genome rearrangement and displays pronounced thermotolerance. We use microarray analysis to characterize genes and their response in the double mutant to a heat shock experiment in which plants were exposed to a temperature of 37 C for 2 hours. Wild type and double mutant plant were exposed to a temperature of 37 C for 2 hours and RNA samples were collected for the wild type and the double mutant before and after the heat shock.

Phosphoproteome dynamics of Saccharomyces cerevisiae under heat shock and cold stress.

Project description:The ability of cells and organisms to survive and function through changes in temperature evolved from their specific adaptations to nonoptimal growth conditions. Responses to elevated temperatures have been studied in yeast and other model organisms using transcriptome profiling and provided valuable biological insights on molecular mechanisms involved in stress tolerance and adaptation to adverse environment. In contrast, little is known about rapid signaling events associated with changes in temperature. To gain a better understanding of global changes in protein phosphorylation in response to heat and cold, we developed a high temporal resolution phosphoproteomics protocol to study cell signaling in Saccharomyces cerevisiae. The method allowed for quantitative analysis of phosphodynamics on 2,777 phosphosites from 1,228 proteins. The correlation of kinetic profiles between kinases and their substrates provided a predictive tool to identify new putative substrates for kinases such as Cdc28 and PKA. Cell cycle analyses revealed that the increased phosphorylation of Cdc28 at its inhibitory site Y19 during heat shock is an adaptive response that delays cell cycle progression under stress conditions. The cellular responses to heat and cold were associated with extensive changes in phosphorylation on proteins implicated in transcription, protein folding and degradation, cell cycle regulation and morphogenesis.

Time-resolved heat stress response of Saccharomyces cerevisiae

Project description:Whole-genome transcriptional response of S. cerevisiae to an increase in temperature from 28°C to 41°C under well-controlled conditions. Two subsequent phases of response with very different dynamics: a short term response for the first hour after the temperature increase and a long term one for up to six hours. The initial response was strongest with almost half of the ORFs being induced or repressed to a statistically significant level (here 1.5 fold). The data was grouped based on the function of the encoded proteins. Analysis showed that the cells overexpressed genes involved in energy conservation processes. Genes encoding molecular chaperones were overexpressed as well, presumably to counteract the effect of the temperature increase on protein denaturation. Furthermore, genes encoding parts of the translation and transcription systems were repressed temporarily, in line with the observed lag in growth. More detailed analysis of certain small groups of genes involved in energy metabolism supported the notion that, although the expression level of genes represent a part of the stress response, they cannot be directly linked to the level of activity of their products.
